Recurrent airway obstruction (RAO), or 'heaves', is a common performance-limiting allergic respiratory disease of mature horses. It is related to sensitization and exposure to mouldy hay and has a familial basis with a complex mode of inheritance. In a previous study, we detected a QTL for RAO on ECA 13 in a half-sib family of European Warmblood horses. In this study, we genotyped additional markers in the family and narrowed the QTL down to about 1.5 Mb (23.7-25.2 Mb). We detected the strongest association with SNP BIEC2-224511 (24,309,405 bp). We also obtained SNP genotypes in an independent cohort of 646 unrelated Warmblood horses. There was no genome-wide significant association with RAO in these unrelated horses. However, we performed a genotypic association study of the SNPs on ECA 13 in these unrelated horses, and the SNP BIEC2-224511 also showed the strongest association with RAO in the unrelated horses (p(raw) = 0.00037). The T allele at this SNP was associated with RAO both in the family and the unrelated horses. Thus, the association study in the unrelated animals provides independent support for the previously detected QTL. The association study allows further narrowing of the QTL interval to about 0.5 Mb (24.0-24.5 Mb). We sequenced the coding regions of the genes in the critical region but did not find any associated coding variants. Therefore, the causative variant underlying this QTL is likely to be a regulatory mutation.

In addition to maintaining hemostasis, platelets have an important role in modulating innate and adaptive immune responses. A low platelet count has been found to be a negative prognostic factor for survival in humans and horses with critical illnesses, such as sepsis or systemic inflammatory response syndrome (SIRS). Decreased platelet aggregation, caused by in vivo activation, has been found in human patients with severe sepsis. In our prospective controlled study, we assessed platelet biology in blood samples from 20 equine SIRS cases and 120 healthy control horses. Platelet variables such as platelet count, large platelet count, clumps, plateletcrit, mean platelet volume, and mean platelet component concentration were analyzed by laser flow cytometry (Advia 2120) from K3EDTA blood and from citrate blood. Hirudin blood samples were analyzed by impedance aggregometry (Multiplate analyzer; Roche) for platelet aggregation, including spontaneous aggregation and aggregation by 4 different agonists: adenosine diphosphate (ADPtest), ADP + prostaglandin E1 (ADPtestHS), arachidonic acid (ASPItest), and collagen (COLtest). SIRS cases had significantly lower platelet counts in K3EDTA blood ( p < 0.0001) compared to control horses. There were no significant differences in aggregation values between SIRS cases and controls. Non-surviving SIRS horses did not have statistically significant lower platelet counts or lower aggregation values for COLtest, ADPtest, or ADPtestHS compared to surviving SIRS horses, although 5 non-survivors were thrombocytopenic.

ZusammenfassungDie equine Piroplasmose, ausgelöst durch die protozoären Erreger Theileria equi oder Babesia caballi, ist weltweit in (sub-)tropischen Regionen einschließlich Südeuropa endemisch. In Deutschland hat sie vor allem als Importparasitose Bedeutung. Da Zeckenarten mit Vektorkompetenz für equine Piroplasmen allerdings auch hierzulande vorkommen, ist eine langfristige Etablierung autochthoner Zyklen nach Einschleppung durch Equiden oder Zecken nicht ausgeschlossen. Im vorliegenden Fall erkrankten 2 Pferde nach einem Wanderrittaufenthalt in Südfrankreich an einer T. equi-Infektion. Während der Behandlung wurde bei einem der Pferde eine saugende Zecke (Rhipicephalus bursa) entdeckt. Auch wenn sich diese Zeckenart unter den derzeitigen Bedingungen in Deutschland nicht etablieren kann, verdeutlicht dieser Fund das Potenzial der Einschleppung infizierter Zecken. Die Therapie der Pferde erfolgte mit Imidocarb (3 mg/kg i. m.; 3 Behandlungen im Abstand von je 3 Tagen). Unerwünschte Arzneimittelwirkungen wurden nicht beobachtet. Um eine Etablierung des Infektionszyklus in Deutschland sowie eine chronische Manifestation der Piroplasmose zu verhindern, sollte trotz möglicher Nebenwirkungen von Imidocarb eine Erregerelimination angestrebt werden.

ZusammenfassungEin 15-jähriger Warmblutwallach wurde wegen multipler großflächiger, ulzerativer und krustiger Hautveränderungen vorgestellt, die seit 4 Jahren bestanden. Anhand der histologischen Untersuchung von Hautbioptaten wurde eine subepidermale Spaltbildung unterhalb der Basalzellen und oberhalb der Basalmembran festgestellt und ein bullöses Pemphigoid diagnostiziert. Durch eine immunsuppressive Therapie mit Dexamethason und Azathioprin konnte nach 14 Wochen eine vollständige Remission der Hautläsionen erreicht werden, wobei die betroffenen Hautareale durch Narbengewebe ersetzt wurden. Die Dauertherapie wurde nach schrittweiser Dosisreduktion über 5 Monate mit einer Erhaltungsdosis von 0,5 mg/kg Azathioprin täglich fortgesetzt. Nach 63 Wochen erfolgreicher Stabilisierung kam es zu einem Rezidiv sowie zusätzlich zu unerwünschten Arzneimittelnebenwirkungen (akute Hufrehe und erhöhte Infektanfälligkeit), sodass der Wallach aus Tierschutzgründen euthanasiert wurde.
