A general report on the use of the Allium test as cytotoxicological and genotoxicological assay is proposed, with particular emphasis about the standardization of the test in several common applications. The intraspecific variation in Allium cepa has been overlooked, as in most investigations no mention is made about origin and denomination of the onion cultivar used. A standardization of the used material would allow a better generalization of the results, since we cannot be sure that all cultivars would give the same response. A more frequent use of transmission electron microscopy (TEM) investigation is proposed. Even if relatively time consuming and not available in all laboratories, it may help to better understand the mechanism of cytotoxicity, since many morphological characters may appear similar but be arisen from different processes observable only with TEM. About statistical testing, tests other than chi-squared may be used in case of a lower amount of data. The most commonly used statistical tests are the parametric tests ANOVA and Student's t, and the nonparametric tests Kruskal-Wallis and Mann-Whitney U, for analysis of variance. Tests should be used also to assess the minimal sample dimension for obtaining significance, since data collection (microscope observation) appears to be one of the main bottle necks of the test. Also the use of the Allium test for testing liposomes and other nanovectors for drug delivery is proposed, in order to assess the cytotoxicity of these types of medium and the possible increase in cytotoxicity of the associated drug.
Diplopods feed organic matter in decomposition; however, some environmental factors can promote changes in tissues of these animals. Sewage sludge has been applied for recuperation of physical structure of degraded soil. This work analyzed the influence of the sludge from a city of São Paulo in the midgut of the diplopod Rhinocricus padbergi. After the exposition to sludge, the midgut was prepared for histological and ultra-structural analyses. After 1 week of exposition, there were various glycoprotein globules in the fat body, which appeared, ultrastructurally, little electron dense. In the animals exposed for 2 weeks, there was an intensive renovation of the epithelium with the invasion of regenerative cells, which was observed in the histological and ultrastructural analyses. These data showed that the sludge present various substances that were very hazardous for these animals; more studies were necessary before the application of this in agriculture.

The indiscriminate use of pesticides has become a serious environmental concern. Of them, imidacloprid (IMI) is one of the most widely used worldwide. In 2010 in Brazil, 1.934 tonnes of IMI were sold and mainly used for sugarcane crops. Several studies have examined the toxicity of IMI as well as its possible ecological effects. However, few studies have examined its toxicity at the genetic level. This is one of the biggest challenges for the scientific community, which is concerned about the impacts of these contaminants on the environment and human health. In this study, we evaluated the effects of IMI above the genetic material in Allium cepa and Tradescantia pallida following exposure to different concentrations of this insecticide. The results demonstrated that the concentrations tested induced chromosomal alterations and increased the frequency of micronuclei. Therefore, IMI in these concentrations was genotoxic to the tested organisms. These factors should be taken into account when applying this pesticide.
