This study aimed to apply cellulose acetate (CA) films incorporated with oregano essential oil (OEO) to inhibit bacteria growth associated with spoilage of meat products (Weissella viridescens (microaerophilic) and Pseudomonas fluorescens (aerobic)) and evaluate its effect on the shelf life of vacuum-packed sliced ham (VPSH). CA films were produced using acetone solvent, adding 25, 35, 50, or 75 mg of OEO per film. Antimicrobial activity and mechanical properties of films were determined. CA films in Petri dishes showed a better antimicrobial effect against W. viridescens than P. fluorescens. As VPSH, presents a microaerophilic environment, product shelf life was determined fitting Baranyi and Roberts’ model to W. viridescens’ growth experimental data, at 8 °C. OEO did not modify films’ mechanical properties. Application of the CA film with 75 mg of OEO decreased value of W. viridescens, increased its value, resulting in a ham` shelf-life increased by eight days, demonstrating excellent application potential.
O Croton blachetianus, popularmente denominado marmeleiro, é um arbusto rico em metabólitos secundários que apresenta diversas atividades biológicas. O objetivo desse estudo foi determinar a composição dos compostos voláteis do óleo essencial das folhas secas de C. blanchetianus e sua ação antimicrobiana (dose mínima inibitória – DMI do vapor) sobre as cepas das bactérias ácido-lácticas (BAL), Weissella viridescens e Leuconostoc mesenteroides, deteriorantes de produtos cárneos. O óleo essencial das folhas secas de C. blanchetianus foi extraído pela técnica de hidrodestilação e os compostos voláteis foram analisados pela técnica de headspace, por cromatografia gasosa. A dose mínima inibitória na fase vapor foi definida como a dose mínima de solução de óleo essencial de C. blanchetianus em DMSO 5% cujo vapor inibe completamente o crescimento das BAL, durante 48 horas. Posteriormente, o efeito bactericida e bacteriostático foi determinado. Os compostos majoritários encontrados foram os terpenos eucaliptol (32,94%) e D-α-Pineno (29,43%). A dose mínima inibitória foi de 0,32g/mL, para ambos os microrganismos. O efeito bactericida para W. viridescens foi observado pela ação do vapor da solução de 0,32 g/mL e, para a L. mesenteroides, o efeito bacteriostático e bactericida foi pela ação do vapor da solução de 0,40 g/mL e 0,57 g/mL, respectivamente. Assim, foi possível observar a presença de terpenos como compostos majoritários na análise por headspace, bem como a atividade antimicrobiana, bacteriostática e bactericida do óleo essencial das folhas secas de C. blanchetianus frente as BAL W. viridescens e L. mesenteroides.
Aims: This study evaluated the behaviour of the Salmonella enterica serotypes in osmotically stressful BHI broth (0.940 ≤ a w ≤ 0.960), assessing inoculum from two stages of the bacterial life cycle (exponential and stationary) and two temperatures (25°C and 35°C).Methods and Results: Four S. enterica serotypes (Typhimurium, Enteritidis, Heidelberg and Minnesota) were grown in stressful BHI at 25°C. A mathematical model was proposed for describing the total microbial count as the sum of two subpopulations, inactivating and surviving-then-growing. When submitted to a w of 0.950 and 0.960, all strains showed a decreased count, followed by a period of unchanged count and then exponential growth (Phoenix Phenomenon). Strains inoculated at a w = 0.940 and 0.945 showed inactivation kinetics only. Cells cultivated at 25°C and inoculated from the exponential phase were the most reactive to the osmotic stress, showing a higher initial population reduction and shorter adaptation period. The proposed model described the inactivation data and the Phoenix Phenomenon accurately. Conclusions:The results quantified the complex response of S. enterica to the osmotic environment in detail, depending on the inoculum characteristic and serotype evaluated.Significance and impact of study: Quantifying these differences is truly relevant to food safety and improves risk analysis.
RESUMO -As bactérias ácido lácticas (BAL) estão entre os principais grupos de microrganismos responsáveis pela deterioração de produtos cárneos refrigerados, embalados a vácuo e em atmosfera modificada. O crescimento das BAL pode ser predito por modelos matemáticos que descrevem a influência de diferentes fatores ambientais (como condições não-isotérmicas) sobre o crescimento microbiano. O objetivo deste trabalho foi obter um modelo matemático capaz de predizer o crescimento de Lactobacillus viridescens em condições não-isotérmicas de cultivo em meio MRS. As curvas de crescimento foram descritas pelo modelo de Baranyi e Roberts, e a dependência do parâmetro µ max com a temperatura foi descrita pelo modelo da raiz quadrada. O modelo proposto foi validado com dados experimentais de L. viridescens, com a temperatura variando de 5 a 11 ºC, e de 6 a 10 ºC a cada 24 h. Os resultados mostraram que foi possível predizer de forma segura, confirmado através dos índices estatísticos, o crescimento de L. viridescens em meio MRS em condições não-isotérmicas.
