Zusammenfassung Zur Steigerung der Effizienz des Embryotransfers wurde in den vergangenen Jahren wiederholt versucht Verfahren zur Superovulation bei der Stute zu etablieren. Bisherige Studien zeigten jedoch, dass der Einsatz von equinem Choriogonadotropin (eCG), heterologem FSH, anti-Inhibin Vakzinen, GnRH-Analoga oder pocinem FSH-aufgrund mangelhafter Effektivität und Nebenwirkungen-wenig viel versprechend für den kommerziellen Einsatz bei der Stute zu sein schien. Verschiede Autoren berichteten, dass mit dem seit 2003 in den USA kommerziell erhältlichen gereinigten und standardisierten (10:1 Ratio von FSH:LH) equinem Hypophysenextrakt eFSH ®. Superovulationen bei der Stute ausgelöst werden können. In der vorliegenden Studie wurden sechs zyklische genitalgesunde Stuten über 5 Zyklen untersucht und die Anzahl der Ovulationen und gewonnenen Embryonen aus den stimulierten Zyklen mit denen der Kontrollzyklen verglichen. Zyklus 1 und 3 wurden als Kontrollzyklen ohne Stimulation und Besamung definiert. Im zweiten und vierten Zyklus wurden die Stuten jeweils ab einer Follikelgröße von 22-25 mm mit einer zweimal täglichen Gabe von 12,5 mg eFSH ® (Fa. Bioniche-Animalhealth, Athens, GA, USA) bis zu einer durchschnittlichen Follikelgröße von 32-35 mm stimuliert. Am zweiten Tag der eFSH ® Behandlung wurden 250 µg Cloprostenol (Estrumate ®) intramuskulär injiziert. Nach einer 36 Stunden dauernden "coasting period" und Ovulationsinduktion mit humanem Choriogonadotropin (Ovogest ®) wurden die Stuten 12 und 36 Stunden nach Ovulationsinduktion mit 750 Mio. progressiv motilen Spermien gekühlt (+5° C) gelagertem Sperma eines fertilen Warmbluthengstes inseminiert und eine Embryonengewinnung an Tag 6,5 nach Ovulation durchgeführt. Im fünften Zyklus wurde eine Embryonengewinnung nach Ovulationsinduktion und zweimaliger Besamung wie in den Zyklen 2 und 4, jedoch ohne vorherige Stimulation durchgeführt. In den 18 Kontrollzyklen und 12 eFSH ® stimulierten Zyklen lag die Anzahl Ovulationen (OV) pro Stute bei den eFSH ®-Zyklen mit durchschnittlich 4,4 Ovulationen deutlich über der durchschnittlichen Ovulationsrate in den Kontrollzyklen (1,3 OV). Bis zum Erreichen eines Follikeldurchmessers von >32-35 mm wurden im Mittel 4,0 Tage für die eFSH ®-Behandlung benötigt. Die Embryonengewinnungsrate nach eFSH ®-Stimulation lag im Gegensatz zu Kontrollzyklen ohne Superovulationsbehandlung (1,2 Embryonen) pro Stute bei ø 2,9 Embryonen. Das aufgeführte eFSH ®-Behandlungsschema ist geeignet um Superovulationen und höhere Embryonengewinnungsraten im Vergleich zu nicht stimulierten Zyklen bei der Stute zu erzielen. Nichtsdestotrotz sind die Embryonengewinnungsraten trotz guter Superovulationsergebnisse nicht konstant und von individuellen Unterschieden bestimmt. In aktuelle Studien wird daher der Einfluss der Follikeldynamik und Genitaldurchblutung auf die Embryonengewinnungsrate untersucht, um möglicherweise früher "gute" von "nicht guten" Spenderstuten im Rahmen eines Superovulationsprogrammes zu unterscheiden und damit einen effizienteren Einsatz z...
ZusammenfassungVerschiedene Ursachen können zu einer Azoospermie führen, die mit Infertilität des Hengstes einhergehen. Es werden das klinische Bild, weiterführende Untersuchungen und der Therapieverlauf bei einem dreijährigen Hengst mit einer Azoospermie aufgrund eines Verschlusses der Samenleiterampullen ("plugged ampullae") beschrieben. Bei ungestörter Begattungsfähigkeit wurden bei der Vorstellung des Hengstes spermienfreie Ejakulate mit einem niedrigen Gehalt an alkalischer Phosphatase (30 U/l) gewonnen. Bei der transrektalen Untersuchung erwiesen sich die Samenleiterampullen per palpationem als nicht schmerzhaft, daumenstark und von fester Konsistenz bei ultrasonographisch dilatiertem Lumen. Über einen Zeitraum von einer Woche wurde dreimal täglich eine transrektale Massage der Samenleiterampullen mit anschließender Samenentnahme durchgeführt. Das Ejakulat wurde zunehmend milchig, der drastisch erhöhte Anteil morphologisch abweichender Spermien reduzierte sich und die Anteile vorwärtsbeweglicher und membranintakter Spermien stiegen kontinuierlich während des achttägigen Klinikaufenthalts. Der vorliegende Fall verdeutlicht, dass durch regelmäßige transrektale Massage der Ampullen mit anschließender Samenentnahme eine vollständige Heilung der Azoospermie aufgrund eines Verschlusses der Samenleiterampullen erzielt werden kann.Schlüsselwörter: Hengst / Azoospermie / Samenleiterampullen / Ultrasonographie / Alkalische Phosphatase "Plugged ampullae" -a case report of azoospermia in the stallion Causes for azoospermia as a reason of infertility in the stallion include spermatogenic failure, obstruction of extracurrent ducts, failure of emission or ejaculation and retrograde ejaculation. Clinical signs, diagnostic approaches, therapy and progress of a 3-year-old stallion with azoospermia due to plugged ampullae are presented. The andrological examination of the stallion included clinical and sonographic examination of the testis and accessory glands, as well as spermatological examination and measurement of alkaline phosphatase as a marker of testicular and epididymal secretions. The stallion showed a slightly softened testicular consistence and sonographically a dilatation of the rete testis. Palpation per rectum revealed enlarged, tense ampullae with a sonographically dilatated lumen. During semen collection the stallion showed normal libido and breeding behaviour including urethral pulsation. The water-like ejaculates were lacking spermatozoa. After transrectal massage of the ampullae with subsequent semen collection three-times a day over a period of eight days the ejaculate included an increasing number of motile sperm besides densely packed spermatozoal casts up to a size of 1 x 2.5 cm.
