Penelitian mengenai bakteri toleran uranium yang berpotensi dalam biopresipitasi uranium telah dilakukan. Untuk menetapkan bakteri yang paling toleran uranium maka dilakukan skrining bertingkat dengan medium TGY cair yang mengandung larutan uranium dengan konsentrasi 0,4 mM; 0,8 mM; 1 mM; dan 2 mM terhadap isolat bakteri yang telah diisolasi dari limbah radioaktif cair aktivitas rendah fase organik dan fase air yang dimiliki oleh PSTA BATAN Yogyakarta. Dari hasil skrining awal (preliminary screening) ini diperoleh 51 isolat bakteri toleran uranium. Setelah dilakukan karakterisasi yang meliputi pengecatan Gram, pengamatan bentuk sel dan morfologi koloni serta penghitungan kinetika pertumbuhan dan dikombinasikan dengan teknik rep-PCR maka diperoleh 26 klaster dari 51 isolat bakteri pada tingkat keserupaan (similarity) 83%. Selanjutnya dari tiap klaster dipilih satu isolat dengan laju pertumbuhan spesifik (µ) tertinggi. Dengan demikian didapatkan 26 isolat bakteri toleran uranium yang diuji selanjutnya sebagai bakteri polifosfat. Untuk mendapatkan bakteri polifosfat maka isolat bakteri ini ditumbuhkan dalam medium agar plate yang tidak mengandung P dan diinkubasikan selama 5 hari pada suhu ruang untuk diamati daya hidup (viability) dan perkembangannya. Berdasarkan hasil seleksi ini diperoleh 7 isolat bakteri polifosfat yang mampu tumbuh dan berkembang dengan menggunakan cadangan polifosfat di dalam selnya sebagai sumber energi dan berpotensi dalam biopresipitasi uranium. Bakteri polifosfat ini diberi kode : A-14, A-25, A- 66, A-67 I, O-21, O-27 dan O-29. Selain karakterisasi dilakukan pula identifikasi molekular berdasarkan urutan nukleotida gen 16S rRNA terhadap 7 isolat bakteri polifosfat. Hasil analisis hubungan filogenetik 7 isolat bakteri polifosfat menggunakan bakteri acuan Bacillus cereus ATCC 14579 dan Acinetobacter radioresisten DSM 6976 dengan melihat nilai keserupaan dan perbedaan nukleotida maka dapat ditetapkan bahwa isolat bakteri dengan kode A-35, A-14, A-66, O-27, O-21, dan O-29 memiliki kemiripan dengan Bacillus cereus sedangkan isolat bakteri dengan kode A-67 I memiliki kemiripan dengan Acinetobacter radioresistens.
This study aims to identify the genus of mold in the biodeterioration process of the photo archive of Memory of the World (MoW) restoration of Borobudur Temple and the potential for the enzymatic activity of these molds. The type of research method chosen is descriptive qualitative. Starting with survey sampling and sampling. Inoculation of fungi using the streak method on PDA medium. Mold identification based on macroscopic and microscopic observations of fungi. The results of characterization were then identified using the matching profile method using the mold identification reference book. The identification results resulted in six genera of contaminant molds in the biodeterioration of the MoW photo archive of the Borobudur Temple restoration. The genera identified included: Acremonium (69.66 %%), Penicillium (14.59%), Aspergillus (3.36%), Culvularia (2.24%) Fusarium (1.12%), and Pleurostomophora (1.12%) and some sterile mycelia. The types of biodeterioration in the photo collection include mold growth, discolored spots, peeling off layers, and damage to the substrate in the photo. Based on literature search, all mold genera found as the cause of biodeterioration has the potential to have proteinase, gelatinase, and cellulase enzymes. Kemiskinan unsur hara N dalam tanah dapat diatasi dengan memanfaatkan kelompok mikroba tanah yang dapat berguna sebagai penyedia unsur hara pada tanah yang nantinya tersedia untuk tanaman. Tujuan penelitian ini adalah untuk mengetahui hasil isolasi bakteri, karakteristik fenotipik bakteri, serta perbedaan keanekaragaman bakteri dari bintil akar aktif dan tidak aktif serta rhizosfer tanaman kacang tanah (Arachis hypogaea L.). Penelitian ini di Laboratorium Mikrobiologi FMIPA UNY. Sampel bintil akar dan rhizosfer diambil di sawah, di Kabuaten Magelang, Jawa Tengah. Pengujian Karakteristik fenotipik meliputi karakterisasi morfologi koloni, morfologi sel, dan uji fisiologis (biokimia). Identifikasi bakteri dilakukan dengan metode profile matching, untuk mengetahui indeks similaritasnya dengan genera bakteri acuan. Diperoleh total 15 isolat yang merupakan bakteri gram positif dan negatif, berbetuk coccus maupun bacil, dengan konfigurasi round, margin smooth, elevasi raised, berwarna putih hingga putih kekuningan, dengan hasil uji fisiologis (biokimia) yang beragam. Setelah dilakukan proses identifikasi, diperoleh 2 genera bakteri yang ada pada bintil akar aktif yaitu Azomonas dan Staphylococcus, 3 genera bakteri yang ada pada bintil akar tidak aktif yaitu Mesorhizobium, Azomonas, dan Bacillus, serta 3 genera bakteri yang ada pada rhizosfer tanaman kacang tanah yaitu Rhizobium, Pseudomonas, dan Neisseria.
Penelitian ini bertujuan untuk mengetahui nilai dan status higiene sanitasi, keberadaan bakteri Bacillus cereus, dan pengaruh higiene sanitasi dan lama penyimpanan terhadap kualitas fisik, kimia dan mikrobiologi susu kedelai di dua Industri Rumah Tangga. Jenis penelitian yang dilakukan adalah deskriptif kuantitatif dengan membandingkan higiene sanitasi pada industri susu kedelai di Depok, Sleman dan Caturharjo, Sleman dengan mengisi lembar checklist higiene sanitasi dan dilakukan wawancara. Uji laboratorium dilakukan untuk mengetahui kualitas susu kedelai meliputi syarat fisik, kimia dan mikrobiologis. Uji fisik berupa pengujian organoleptik oleh panelis yang meliputi rasa, bau, warna, dan penampakan. Uji kimia dilakukan adalah uji pH dan sakarin. Uji mikrobiologis dilakukan uji identifikasi bakteri Bacillus cereus dan Angka Lempeng Total (ALT). Hasil penelitian menunjukkan higiene sanitasi IRT A memenuhi standar syarat sedangkan IRT B tidak memenuhi syarat minimal higiene sanitasi. Nilai higiene sanitasi mempengaruhi Uji Lempeng Total. Sedangkan lama penyimpanan mempengaruhi kualitas organoleptik, Angka Lempeng total dan pH.Kata kunci : susu kedelai, higiene sanitasi, lama penyimpanan
