The aim of the work was to study the production of the exopolysaccharides by Agaricus brasiliensis and the isolation of exopolysaccharides (EPSs) with biological effects. A brasiliensis LPB03 was cultured in submerged fermentation in a medium containing glucose, yeast extract, hydrolyzed soybean protein, and salts (pH 6.1) at 29 degrees C and 120 rpm for 144 h. The maximum biomass and EPS yield was 7.80 +/- 0.01 and 1,430.70 +/- 26.75 mg/L, respectively. To isolate the produced EPSs, two methods were compared: (1) with alcohol precipitation and (2) treatment with tricloroacetic acid (TCA), followed by alcohol precipitation. The use of TCA facilitated the purification of the EPS, reducing the amount of the contaminant soy proteins. For monosaccharide identification, the EPSs were hydrolyzed, derivatized to alditol acetates, and analyzed by gas chromatography (GC) and GC-mass spectrometry, which showed the presence (in molar percentage) of mannose (58.7), galactose (21.4), and glucose (13.1) as major sugars, with lower amounts of rhamnose (3.9) and xylose (2.8). Scanning electron microscopy was used to observe the morphological structure of the EPS. The experiments in vivo including EPS in the mice diet during 8 weeks indicated the hipocholesteremic and hypoglycemic effects.
O vírus da Bronquite Infecciosa das galinhas (VBI) pertence ao grupo 3 da família Coronaviridae e é o causador de desordens respiratórias e renais em frangos de corte. A vacinação com vacinas vivas é praticada em matrizes e avós e muitas vezes também nos plantéis destinados ao abate. As vacinas utilizadas no Brasil são usualmente do sorogrupo Massachusetts e baseadas nas amostras H120 e H52. É comum que após a vacinação o vírus vacinal seja detectado por isolamento em ovos embrionados ou por métodos moleculares por até 4 semanas. Após essa data, normalmente, não há detecção de vírus e o VBI, quando encontrado, pode representar recirculação do vírus vacinal no plantel ou a introdução de uma nova cepa do vírus. No presente estudo, para avaliar a circulação do vírus em plantéis de frangos e reprodutoras nos estados do Rio Grande do Sul e Mato Grosso do Sul, foram coletadas 240 traqueias e rins de aves de 48 plantéis, sendo (20 exemplares/4 plantéis) de avós, (80 exemplares/16 plantéis) de matrizes e (140 exemplares/28 plantéis) de frangos de corte, as quais foram analisadas em misturas de cinco amostras. Todos os animais eram vacinados e as amostras foram coletadas ao redor de 2 a 48 semanas após a vacinação. A presença de VBI foi determinada com auxílio de uma reação em cadeia da polimerase tipo nested, direcionada ao gene da proteína S1, padronizada neste estudo. Das 48 amostras testadas, 14 resultaram positivas: cinco foram oriundas de aves vacinadas há menos de quatro semanas na data da coleta e nove eram de amostras de aves vacinadas há mais de quatro semanas, o que pode ser devido à recirculação do vírus vacinal ou mesmo introdução de vírus selvagem nos plantéis.
The objective of this study was to estimate adjustment factors of scrotal circumference for growth traits in Nellore cattle. Measurement at post-yearling of scrotal circumference from 100,107 Nellore was adjusted for six traits: age (A), body weight (BW), hip height (HH), conformation (C), precocity (P), musculature (M), and simultaneously for age and the other five traits (A-BW, A-HH, A-C, A-P, AM), body weight and the other four traits (BW-HH, BW-C, BW-P, BW-M), and hip height and the other three traits (HH-C, HH-P, HH-M). Standard values were 500 days, 300 kg, 135 cm, and 3, for age, body weight, hip height, and visual scores, respectively. Amplitudes of adjustment factor of scrotal circumference for age, body weight, and hip height were 0.6028, 0.4869, and 0.1572, respectively. Scrotal circumference is expected to grow, on average, 8.71 cm per 100 days of age, 6.51 cm per 100 kg of body weight, and 4.81 cm per 10 cm of hip height. The results showed higher amplitude in the adjustment factor of scrotal circumference for age and body weight (0.3429) compared with age and hip height and body weight and hip height (0.1271 and 0.0443, respectively). The amplitude observed in adjustment factor of scrotal circumference for conformation, precocity, and musculature were 0.1328, 0.1175, and 0.1160, respectively. Considering different adjustment factors of scrotal circumference can be useful for identifying better selection criteria for growth and/or sexual precocity in Nellore.
This study aimed to investigate the following environmental effects in Suffolk lambing: contemporary groups, type of birth, and age of animal and age of dam at lambing on conformation (C), precocity (P), musculature (M), and body weight at postweaning (W), and the heritability coefficients and genetic correlations among these traits. Contemporary groups, type of birth, and age of animal and age of dam at lambing were significant for W. For C, all the effects studied were significant, except linear and quadratic effects of age of the animal. For P, all effects studied were significant, except the quadratic effect of age of the animal. For M, the effects of contemporary group, type of birth, and the linear effect of the age of the animal were significant. Heritability estimates were 0.07 ± 0.03, 0.14 ± 0.03, 0.09 ± 0.03, and 0.11 ± 0.03 for C, P, M, and W, respectively, indicating a positive low response for direct selection. Estimates of genetic correlations among the visual scores (C, P, and M) and W were moderate to highly favorable and positive, ranging from 0.48 to 0.90. These results indicate that selection for visual scores will increase body weight.
The aim of this study was to evaluate the genotype x environment interaction (GxE) for scrotal circumference (SC) measured at different ages using pedigree-based (A -1) and pedigree and genomic-based (H -1) relationship matrices. Data from 1,515 Brahman bulls, from the Cooperative Research Centre for Beef Genetic Technologies (Beef CRC) experimental dataset were used in this study. SC was adjusted to age and body weight measured at 6 months (SC6), 12 months (SC12), 18 months (SC18) and 24 months of age (SC24). Body weight (BW) measured at 6 months (BW6), 12 months (BW12), 18 months (BW18) and 24 months of age (BW24) were used as criteria to describe the environment for SC in each age. All the animals measured were genotyped using medium-density SNP chips (“50k” or “70k” SNP) and their genotype were imputed using a reference panel with 729,068 SNP. The environment gradient (EG) was obtained by standardizing the solutions of the contemporary groups obtained by Animal Model with BW as the dependent variable. Then, the reaction norms (RN) were determined through a Random Regression Model. The breeding values (EBV) were estimated using either A -1 or H -1. The rank correlation was obtained using Spearman’s correlation among the EBV estimated for the traits in analysis. For SC6 and SC24, higher estimates of heritability (h²) were obtained using A -1, when compared to those observed with H -1. In those ages, the improvement of the environment decreases the h² coefficient. On the other hand, the h² for SC12 and SC18 increased as the environment became more favorable, regardless of the matrix used. The RN for SC6 and SC24 estimated using A -1 and H -1 showed a decrease of variance from the worst to the best environment, an indication of existence of GxE. On the other hand, for SC12 and SC18, there were no significant differences between the EBV estimated in the lower and in the higher environments, regardless of the kinship matrix used, suggesting absence of GxE on those ages. Spearman’s correlation among EBV estimated using A -1 and H -1 in different EG were practically equal to unity for all traits evaluated. In our study, there was weak evidence of GxE effect on SC in ages suitable for selection for sexual precocity. So, the absence of GxE at 12 and 18 months means these ages are advantageous for measuring SC to selection for sexual precocity. The advantage is that no changes in classification were observed when the sires were evaluated in different environments.
