Objective: Toxoplasmosis, in which obligate intracellular protozoa Toxoplasma gondii (T.gondii) is the causative organism, is a multisystemic disease that can be seen all over the world and can impair all vertebrates. The only hosts known for T.gondii are members of Felidae family. Our study aimed to determine anti-Toxoplasma gondii antibodies with Sabin-Feldman Dye Test (SFDT) in cats in Ankara. It's aimed to evaluate the current situation in terms of Toxoplasmosis spread by comparing our findings with previous studies in the same region. Methods: Rh strain of Toxoplasma used in our study is maintained in our laboratory. SFDT is still accepted as the gold standard. Material of the study was obtained by taking blood samples from cats who were admitted to the clinics between March 2016 and October 2016 in Ankara. Blood samples were inactivated and measurements were done with SFDT 1/4, 1/16, 1/64, 1/256, 1/1024 titers. Results: SFDT resulted positive in 56 (43.4%) cats at a dilution of 1/16, in 7 (5.4%) cats at a dilution of 1/64, in 23 (17.8%) cats at a dilution of 1/256 and negative in 43 (33.3%) cats. Comparison of demographic data with SFDT results showed that positive test results did not differ according to gender and age (P=0.803 and P=0.991, respectively). T. gondii için bilinen tek kesin konak Felidae ailesinin üyeleridir. Çalışmamızda Ankara'da kedilerde Sabin-Feldman Dye Testi (SFDT) ile anti-Toxoplasma gondii antikorlarının tespiti amaçlanmıştır. Aynı bölgede daha önceden yapılan çalışmalar ile karşılaştırmalar yapılarak Toxoplasmozun yayılımı açısından günümüzdeki durumun değerlendirilmesi hedeflenmiştir. Çalışmanın materyali Mart 2016-Ekim 2016 tarihleri arasında Ankara'da kliniklere müracaat eden kedilerden kan örnekleri alınarak sağlanmıştır. Kedilerden alınan kan örnekleri inaktive edilerek SFDT 1/4, 1/16, 1/64, 1/256, 1/1024 titrelerde çalışılmıştır. Bulgular: Toxoplasma gondii araştırması yapılan kedilerin 56'sında (%43,4) SFDT 1/16 titrede, 7'sinde (%5,4) 1/64 titrede, 23'ünde (%17,8) 1/256 titrede pozitif saptanırken, 43'ünde (%33,3) negatif çıkmıştır. Demografik bilgiler ile SFDT sonuçlarının karşılaştırılmasında; pozitif test sonuçlarının cinsiyet ve yaş ile ilişki göstermediği bulunmuştur (Sırasıyla P=0,803 ve P=0,991). Sokak kedilerinde ev kedilerine göre seropozitiflik fazladır (P<0,001). Sadece ticari kuru mama ile beslenen kedilerde test sonuçları negatiftir (P<0,001). Objective: European hares (Lepus europaeus) are among the most important animals that are connected with humans in many countries and natural life. Hares are important for public health, since they carry many zoonotic diseases, such as Encephalitozoon cuniculi, Francisella tularensis and Toxoplasma gondii. The study aimed to determine the seroprevalence of Encephalitozoon cuniculi, Francisella tularensis and T. gondii and the potential zoonotic risk posed by hares that live in provinces of Turkey.Methods: Blood samples were collected from hares during the official hunting season. Serum samples were examined serologically by enzyme-linked immunosorbent assay for E. cuniculi, Sabin-Feldman dye test was used to examine T. gondii, while micro-agglutination test was used to examine F. tularensis. Results:Of the total of 42 hares examined, one (2.4%) was found positive for E. cuniculi, two (4.8%) were found positive for T. gondii and one (2.4%) was found positive for F. tularensis. Conclusion:Anti-T. gondii and anti-E. cuniculi antibodies were serologically detected in hares for the first time in Turkey. Furthermore, this is the first study reporting the seropositivity of F. tularensis infection in hares.
Encephalitozoon cuniculi can cause latent disease, especially in lagomorphs and many wild and domestic animals in various countries. This infection is important for veterinary and public health because it is caused by a potentially zoonotic and opportunistic pathogen. The aim of this study was to investigate the relationship between seropositivity of E. cuniculi and renal function markers, which may be useful in predicting the disease in clinically healthy rabbits. In this study, the seropositivity of E. cuniculi infection in clinically healthy rabbits was determined, and necropsy findings were evaluated along with the results of renal function tests. In a laboratory rabbit breeding facility, enzyme-linked immunosorbent assay (ELISA) tests showed that 48 (49.5%) of 97 rabbits were seropositive against E. cuniculi. Blood urea nitrogen (BUN) and creatinine levels were significantly elevated in seropositive animals. Two seropositive rabbits were necropsied to confirm the infection. According to histopathological findings in the kidney, degenerative changes and E. cuniculi spores were identified in the tubule epithelia. Serum creatinine (p<0.001) and BUN (p<0.01) levels were found to have a statistically significant relationship with the serological status of rabbits. Serological and histopathological methods are not routinely used in rabbits to diagnose E. cuniculi infection. The kidneys were one of the most affected organs in encephalitozoonosis in rabbits. As revealed in this study, the testing blood urea nitrogen and creatinine levels would be useful for the evaluation of general health status and renal function of the seropositive rabbits, and clinical interpretation as well.
Working in a laboratory is very difficult and needs special attention. Laboratory workers can be exposed to numerous potential hazards including chemical, biological, physical, and radioactive. That is why it is really important to follow the working principles in laboratories for the sake of the lab analyzers and others who work with them in the lab. Laboratory safety includes the use of certain laboratory rules, methods, infrastructures, and devices during work to protect the working person and the working material. All studies show that > 70% of medical decisions are based on laboratory results. In such important laboratories, it is must to get safe and reliable results. This requires a well-established working system and strict observance of laboratory safety. Biosafety is very important in parasitology laboratories as well as in all microbiology laboratories. Usually, it takes a long time for people to detect parasitic diseases through laboratory accidents, who are working in laboratories. That is why, especially in parasitology laboratories, the issue of laboratory safety should be emphasized more sensitively. We will be reviewing the hazards, parasites, exposure routes, and protective measures imposed in parasitology laboratories.
Bu çalışmada 2021 yılında Çankırı il sınırlarını içerisindeki 9 farklı istasyondan avlanan 37 adet tatlı su balık türünün (20 Cyprinus carpio, 7 Capoeta tinca, 5 Squalius cephalaus, 5 Alburnus spp.) sindirim sistemi parazitler yönünden incelenmiştir. Bulunan helmintlerin morfolojik teşhisi yapılmış ve enfeksiyon parametreleri hesaplanmıştır. Parazitolojik incelemesi yapılan 37 balığın 9 (%24,32)’unun sindirim kanalında en az bir tür olmak üzere nematod parazitler toplanmıştır. Pseudocapillaria tomentosa (2), Rhabdochona gnedini (11), R. denudata (7) ve Rhabdochona spp. (4) olmak üzere 24 adet nematod türü morfolojik olarak teşhis edilmiştir. İncelenen toplam 20 C. carpio’ nun %15 (3/20)’inde, 7 C. tinca’nın %42,9 (3/7)’unda, 5 S. cephalus’un %40 (2/5)’ında ve 5 Alburnus spp.’ un %20 (1/5)’sinde nematod parazitler saptanmıştır. Bu çalışmada parazitolojik muayenesi yapılan balıklardan C. carpio türünde Rhabdochona spp. ve P. tomentosa; S. cephalaus’ da R. gnedini ve R. denudata; C. tinca’da R. gnedini ve R. denudata ile Alburnus spp.’ da R. gnedini türleri bulunmuştur. Bu araştırma ile Türkiye’de ilk kez R. gnedini türü rapor edilmiştir. Ayrıca, bu parazit türü Türkiye balık parazit faunası için yeni bir kayıttır.
Bu çalışmada T. gondii için tanıda kullanılan serolojik yöntemlerden anti-T. gondii IgG, IgM, IgG avidite ve Sabin-Feldman dye test (SFDT) sonuçları incelenmiştir. Anti-T. gondii IgG, IgM ve IgG avidite testleri için ELISA yöntemi kullanılmıştır. T. gondii tanısında referans test olan SFDT halen altın standart olarak geçerliliğini korumaktadır. Yapılan laboratuvar analizlerine ek olarak tüm olguların cinsiyet, yaş, numunenin geldiği il, yıl dağılımları, gönderilen numune tipi bilgileri de verilmiştir.Bulgular: Bu çalışmada değerlendirilen 2,778 olgunun 706'sı (%25,4) erkek, 2,072'si (%74,6) kadındır. Anti-T. gondii IgG değerleri incelendiğinde; tüm örneklerin %47,1'i, anti-T. gondii IgM'in %10,2'si pozitif bulunmuştur. SFDT'sinde ise 2,361 hastanın 1,228'i (%52) pozitif tespit edilmiştir. Başvuran 537 erkeğin 319'u (%59,4), 1,824 kadının 909'u (%49,8) pozitif bulunmuştur. On yıllık çalışmada SFDT'nin en sık seropozitiflik titresinin 1/64 düzeyinde olduğu görülmüştür. Çalışmamızda anti-T. gondii IgG, IgM ve IgG avidite testlerinin her üçünün de bir hastada birlikte çalışıldığı tüm olgular değerlendirildiğinde IgG avidite çalışılan olguların tamamında IgG düzeyleri pozitif bulunmuştur. Ek olarak anti-T. gondii IgG değerleri pozitif bulunan 293 olgunun %62,8'inde yüksek avidite, %24,2'sinde sınır değer ve %13'ünde düşük avidite bulunmuştur. Çalışmamızda anne-bebek birlikte gelen olgular değerlendirildiğinde; her ikisinde anti-T. gondii IgG seropozitifliği %80, her ikisinde anti-T. gondii IgM seropozitifliği %5 saptanmıştır. Bu yüksek oranlar, anne bebek arasında antikor transferini desteklemektedir. Numunelerin geldiği il dağılımı incelendiğinde en fazla olgunun %80,7 ile Ankara'dan geldiği görülmüştür. En fazla gönderilen örnek tipi kan örneği, ikinci sıklıkta ise beyin omurilik sıvısı olarak tespit edilmiştir. Sonuç: T. gondii yüksek pozitiflik oranları nedeniyle günümüzde halk sağlığı açısından önemini korumaktadır. On yıllık verilerin toplandığı bu çalışma göstermiştir ki; farkındalık artışına rağmen halen yüksek seropozitiflikler devan etmektedir. Bu nedenle toksoplazmoz tanısına yönelik yapılan laboratuvar analiz sonuçlarının sistematik olarak toplanması ve değerlendirilmesi, kontrol önlemlerinin alınmasına katkı sağlayacaktır.
