The present study was carried out to investigate the effect of plant growth promoting thermotolerant Pseudomonas putida strain AKMP7 on the growth of wheat plants to heat stress. The results indicated the superior performance by P. putida strain AKMP7 in improving survival and growth of wheat plants under heat stress. The bacterium significantly increased the root and shoot length, dry biomass, tiller, spike let and grain formation of wheat over uninoculated plants. Inoculation reduced membrane injury and the activity of several antioxidant enzymes such as SOD, APX and CAT under heat stress. Inoculation improved the levels of cellular metabolites like proline, chlorophyll, sugars, starch, amino acids, and proteins compared to uninoculated plants. Scanning electron microscopy studies confirmed the colonization of the organism on the root surface. This result suggests the possible role of microorganisms in mitigating adverse effects of climate changes on crop growth and may lead to development of microbe based climate-ready technology.

A reproducible and highly efficient protocol for genetic transformation mediated by Agrobacterium has been established for greengram (Vigna radiata L. Wilczek). Double cotyledonary node (DCN) explants were inoculated with Agrobacterium tumefaciens strain LBA 4404 harboring a binary vector pCAMBIA 2301 containing neomycin phosphotransferase (npt II) gene as selectable marker, β-glucuronidase (GUS) as a reporter (uidA) gene and annexin 1 bj gene. Important parameters like optical density of Agrobacterium culture, culture quantity, infection medium, infection and co-cultivation time and acetosyringone concentration were standardized to optimize the transformation frequency. Kanamycin at a concentration of 100 mg/l was used to select transformed cells. Transient and stable GUS expressions were studied in transformed explants and regenerated putative plants, respectively. Transformed shoot were produced on regeneration medium containing 100 mg/l kanamycin and 250 mg/l cefotaxime and rooted on ½ MS medium. Transient and constitutive GUS expression was observed in DCN explants and different tissues of T0 and T1 plants. Rooted T0 and T1 shoots confirming Polymerase Chain Reaction (PCR) positive for npt II and annexin 1bj genes were taken to maturity to collect the seeds. Integration of annexin gene into the greengram genome was confirmed by Southern blotting.

The effect of low light intensity (40 to 50% of natural light) at different growth stages and subsequent exposure to natural light was assessed on growth and productivity in 2 genotypes during khari] and 3 during rabi under 2 N levels. The dry matter accumulation was affected under shading at all stages, which was critical from primordial initiation (PI) onwards. The expression of panicle number appears to be a function of the environment particularly the light intensity within 26 to 39°C range of mean maximum temperatures. The grains/panicle were also reduced by shading from PI onwards. The yields were progressively reduced with low light intensity appearing in succession at different growth stages, particularly more critical during ripening phase. It was considered that low light intensity during monsoon season was an important constraint for higher productivity since yields as low as 3.2 to 4.4 tons/ha were recorded with varieties which yielded 8 to 10 tons/ha under high light intensity conditions as in rabi.
