Resumen En el Perú, Cucurbita moschata Duchesne tiene una variedad local llamada Loche, que se propaga por esquejes y de la que se conoce muy poco sobre su reproducción sexual. Por ello, el objetivo del presente trabajo fue evaluar la biología floral y reproductiva del Loche de Lambayeque. En esta especie las flores femeninas poseen mayor diámetro y longitud de corola que las masculinas, sin embargo, estas últimas presentan pedicelos y sépalos más largos. La floración masculina inició antes que la femenina con la aparición de los botones florales. En la antesis, las flores femeninas iniciaron este proceso antes que las masculinas. Asimismo, durante la producción de flores en promedio se observó 10 flores masculinas por cada flor femenina. En relación al grano de polen, éste tiene una viabilidad del 98% desde la antesis, la que decae hasta un 20% al día siguiente. El número promedio de granos de polen por flor osciló entre 25 125 y 31 833 y el diámetro presentó de 0.1 a 0.2 mm. El estigma es receptivo durante tres días desde la antesis. La producción de néctar fue mayor en las flores femeninas que en las masculinas. El Loche presenta varios sistemas reproductivos: a nivel sexual, la geitonogamia y alogamia; y a nivel asexual, por medio de esquejes y apomixis facultativa, los cuales fueron determinados a partir de pruebas de polinización. Entre los polinizadores de C. moschata, el orden Hymenoptera tiene el mayor número de familias con afinidad por sus flores. Las familias Vespidae y Apidae son las de mayor regularidad en las visitas, siendo Drosophila sp. la de mayor duración en estancia.
ResumenEl café es el principal producto agrícola de exportación del Perú, con gran importancia económica y social. Un importante centro de producción de este cultivo con un clima particular por su humedad y temperaturas medias máximas y mínimas (en relación a otras zonas productoras de café), altitud (entre 1000 y 1500 m.s.n.m.) y flora nativa, es el Distrito de Villa Rica (Pasco), donde existen diferentes variedades, destacando entre ellas la variedad "Typica". En este estudio se analizó la diversidad genética mediante marcadores RAPD de 42 muestras que corresponden a 18 variedades de café (Coffea arabica L.) y como contraste se utilizó la variedad de café "Robusta" (Coffea canephora Pierre ex Froehner). Se obtuvieron 34 fragmentos de ADN polimórficos; el número de bandas polimórficas por iniciador varió entre uno (utilizando el cebador OPA-17) y cinco (con los cebadores OPA-11 y OPB-05). En el análisis de agrupamiento se construyó un dendograma utilizando el coeficiente de Jaccard y la técnica de ligamento promedio (UPGMA). Este análisis determinó que los cafés arábicos de la zona de Villa Rica presentan un bajo grado de diversidad genética y que no se observa una separación discreta entre las variedades arábicas; sin embargo, la diferenciación genética fue alta con respecto a la variedad Robusta. AbstractCoffee is the main agricultural export product of Peru, with great economic and social importance. Villa Rica (Pasco) is a major production center due to special climatic conditions of humidity, maximum and minimum average temperatures (in relation to other coffee producing regions), elevation (between 1000 and 1500m) and native flora. Different varieties can be found but "Typica" can be highlighted. Using RAPD markers the genetic diversity of Coffee (Coffea arabica L.) was examined. 42 samples from a total of 18 varieties were used and in contrast a variety of "Robusta" coffee (Coffea canephora Pierre ex Froehner) was employed. Thirty four polymorphic DNA fragments were obtained. The number of polymorphic bands per primer ranged from one (with primer OPA-17) to five (with primers OPA and OPB-11-05). In the cluster analysis a dendrogram was constructed using the Jaccard coefficient and UPGMA technique. This analysis found that Arabica beans from Villa Rica have a low degree of genetic diversity and no discrete separation is observed between Arabica varieties, but genetic differentiation is high with the Robusta variety.
Resumen El yacón es un cultivo de origen andino caracterizado por almacenar principalmente fructooligosacáridos en sus raíces; sin embargo, no se conoce aún cuanta diversidad genética a nivel molecular existe en la especie. En el presente estudio se caracterizó 30 accesiones de yacones cultivados provenientes del norte, centro y sur del Perú mediante la técnica de RAPDs, utilizando 34 iniciadores decaméricos; con los cuales se muestrearon 166 fragmentos genómicos, de las cuales el 30.7% fueron polimórficos. Al 0.58 de similitud se formaron 7 grupos de yacones. El primero de ellos con 22 accesiones agrupa a todas las provenientes del norte y del sur, y algunas del centro, los grupos restantes solamente contienen accesiones del centro. No se logró encontrar material duplicado y la mayor diversidad estuvo concentrada en las accesiones provenientes del centro del Perú. El AMOVA reportó un 21.14% de variación genética explicada por el componente interregional y la mayor variabilidad estuvo concentrada entre las accesiones de cada región (78.86%).
ResumenSe estudió la diversidad genética mediante la técnica de RAPD en 37 accesiones de una colección del norte del Perú de "chago" o "mauka" Mirabilis expansa; Se obtuvieron 60 marcadores polimórficos con 9 de 19 iniciadores decaméricos. Se calcularon los índices de iniciador RAPD, obteniéndose los valores más altos con los iniciadores OPA04, OPA09 y OPA13, lo cual sugiere su uso valioso en futuras investigaciones con RAPD en colecciones más grandes o para especies de Mirabilis. Con el coeficiente de Simple Matching y el algoritmo UPGMA se obtuvo un dendograma del cual, a un coeficiente de 1, se observan 31 grupos. Esto indicaría unos 16.216 % de posibles duplicados en la colección de Germoplasma. Con un índice de similitud de 0.85 se encontró que se forman 8 clusters o grupos, sin coincidir en su mayoría con los 5 morfotipos reportados. Además se realizó un Análisis Molecular de Variancia con 2 componentes: interregional y entre accesiones/ región, cuyos valores fueron 21.69% y 78.3%, respectivamente; valores que sugieren una considerable contribución de variación genética gracias a las muestras de diversas partes del país. Palabras clave: RAPD, Mirabilis expansa, raíz tuberosa, diversidad genética, AMOVA Abstract A collection of 37 accessions of Mirabilis expansa, "chago" or "mauka" from northern Perú, was analyzed by RAPD assays and the molecular genetic diversity determined. In order to know which primer was more informative, the PIC values were summed up and a RAPD primer index calculated, resulting in the primers OPA04, OPA09 y OPA13 with the highest indexes, suggesting their potential utility in further research in Mirabilis species. From 9 of the 19 RAPD primers selected, we obtained 60 polymorphic markers, which yielded a Simple Matching Coefficient Dendogram, which shows 31 groups at a similarity index of 1. It also indicates a 16.216% of possible duplicates in the collection and with a similarity index of 0,85 we observed 8 clusters, with a significant difference compared to the morphological characterization data. An Analysis of Molecular Variance was performed, separating 2 variation components: among regions and among accessions/within region. The values obtained were 21,69% and 78,3% respectively, being both relatively high and showing considerable contribution of genetic variation from samples of diverse collection places.
