Minyak kelapa murni (VCO) merupakan salah satu hasil olahan dari buah kelapa segar. Penggunaan biji, daun, dan kulit pepaya pada pembuatan VCO selain lebih murah dan mudah didapat, juga merupakan enzim golongan protease dan dapat diaplikasikan dalam pembuatan VCO. Penelitian ini bertujuan untuk mengetahui proses pembuatan enzim papain kasar dari biji, daun, dan kulit pepaya, mengetahui pengaruh penambahan enzim papain kasar terhadap rendemen VCO, dan kualitas VCO yang dihasilkan. Hasil penelitian menunjukan bahwa penambahan enzim kasar berpengaruh terhadap rendemen dan kualitas VCO yang dihasilkan. Rendemen tertinggi (33,75%) diperoleh dengan penambahan enzim papain kasar dari biji papaya dengan konsentrasi 30%, Sedangkan rendemen yang terendah (21,12%) dihasilkan dengan enzim papain kasar dari daun dengan konsentrasi 20%. Parameter yang diamati adalah uji kadar air, kadar asam lemak bebas, massa jenis, dan bilangan peroksida. Kadar air VCO yang dihasilkan dari penelitian ini semua sampel belum memenuhi syarat mutu VCO, yaitu maksimal 0,5% (SNI 7381-2008). Sedangkan Hasil uji massa jenis ini yang memenuhi standar mutu SNI 7381-2008 yaitu sampel penambahan enzim papain dari biji 10%, daun 20%, biji 30%, daun 30% dan kontrol. Sedangkan sampel daun 10%, kulit 10%, biji 20%, kulit 20%, dan kulit 30% belum memenuhi standar SNI 7381-2008 yaitu sebesar 0,915-0,920 kg/m3. Hasil uji bilangan peroksida dapat diketahui bahwa semua sampel belum memenuhi standar SNI 7381-2008 sebesar ≤ 3 meq/kg.
