Um fur pharmakologische Untersuchungen und die Diskussion von Struktur-Wirkungs-Beziehungen uber eine luckenlose homologe Reihe zu verfugen, sollten noch y-lactonisierte Diarylpropylamine zuganglich gemacht werden.Die friiher beschriebene Synthese von a-Amino-y-lactonen durch Aminolyse von a-Bromlactonen6) kann auf 6 nicht ubertragen werden, da bereits bei dem Versuch, y, y-Diphenylbutyrolacton zu bromieren, Elimination zum ungesattigten Lacton erfolgt6). Eine nachtragliche N-Alkylierung des N-unsubstituierten Analogons von 6a (R' = NH,) ist unzweckmaflig, da wir dieses Aminolacton nur in sehr geringer Ausbeute (5.9 %) erhielten' .). So priiften wir, ob uns die Umsetzung der a-Amino-y-ketosauren 5 rnit metalliertem Benzol zu 6 fuhren kann.
Im Rahmen unserer Untersuchungen über lactonisierte Diarylalkylamine synthetisierten wir kürzlich N-alkylierte Derivate von 5 ? Auffällig in den 'H-NMR-Spektren dieser Verbindungen war die erhebliche Tieffeldverschiebung des P-ständigen Protons. Um strukturelle Unsicherheiten zu beseitigen und darüberhinaus auch Über die unsubstituierten Basisverbindungen der lactonisierten Diarylethyl-und -propylamine verfügen zu können, nahmen wir die Synthese der isomeren Aminolactone 4 und 5 in Angriff. SynthesenWie für Amine beschrieben? setzt sich auch NH, mit 2 zum Aminoamid 3 um, das ohne Reinisolierung durch saure Hydrolyse und Lactonisierung in 5 überführt werden kann. -Problematischer ist die Darstellung von 4. Ben-Ishai und Mitarb. berichteten über die Synthese von a-N-Acylamino-y-lactonen wie 1, nicht jedoch über die erfolgreiche Verseifung zu a-Arninolactonen*! D a der Lactonring erhalten bleiben soll, liegt die saure Hydrolyse nahe. Sie gelingt weder in siedender óN-HCl(l2 h), 48proz. HBr (1 2 h) noch 2N-Salzsäure/Ethanol (4 d). Stets wird 1 zurückerhalten. Auch bei Erhitzen in 6N-NaOH (2.5 h) bleibt 1 in Suspension. Erst nach Behandeln in siedender 6N-NaOH/EtOH 1äBt sich eine geringe Menge 4 isolieren. H-NMR-Spektroskopie, StereochemieDie envelope-Konformation der Butyrolactone. bei der das P-C-Atom aus der Ebene der vier anderen Ringatome heraustritt. wurde mehrfach in Röntgenstrukturanalysen e~-mittelt~'~). Wir konnten sie auf diese Weise auch für ein a-aminoalkyliertes y. y-Diphenylbutyrolacton bestatigen? DaB diese Konformation auch in Lösung vorliegt. zeigen die 'H-NMR-Spektren Ca-und C B-substituierter Butyr~lactone~. 4. 6.7! Umsetzung der 5,5-0iaryl-2(5H)-furanone 4a-d mit überschüssigen Aminen liefert Aminoamide 5a-k, die durch Hydrolyse und Ringschluß in die gewünschten 4-Amino-5,5-diaryl-4,5-dihydro-2(3H)-furanooe 6a-k überführt werden. Ein wesentlicher Parameter in der Struktur-Wirkungs-Beziehung bei Pharmaka mit Diarylalkylamin-Struktur ist die Länge der Alkylkette. Im Rahmen unserer Untersuchungen über lactonisierte Diarylalkylamine berichteten wir über Synthese und Pharmakologie von Derivaten mit 4 2 • 31 und 5 41 C-Atomen zwischen Aromaten und Aminogruppe. Um in dieser Reihe vollständigere Aussagen über Struktur-Wirkungs-Beziehungen machen zu können, sollte die Synthese diarylierter Aminolactone mit 2 C-Atomen zwischen Aromaten und Aminogruppe in Angriff genommen werden. Eine Möglichkeit sahen wir in der Addition von Aminen an ~a. ß-Butenolide des Typs 4, die nach Cannone et al. durch Grignardisierung des Diels-Alder-Adduktes I und anschließende thermische Spaltung zugänglich sind 51 • Aus bisher durchgeführten Umsetzungen von Aminen mit a, ß-ungesättigten Lactonen wie ß-Angelicalacton und Strophanthidin ergibt sich allerdings eine recht unübersichtliche Reaktivität. Michael-Addukte können nur selten rein isoliert werden, es überwiegt die Bildung von ungesättigten Hydroxyamiden, ungesättigten Aminosäuren, y-Imino-oder y-Enaminoamiden und -im Falle primärer Amine -Pyrrolioonen o -91 • Lehmann und GossenArch.
