Background:Cell interaction between dendritic cells (DC) and natural killer (NK) cells in the periodontal milieu is not yet fully known, although these cells are individually known to contribute to the pathogenesis of periodontal disease.Materials and Methods:Fifty subjects (25 males and 25 females) were included in the study. The subjects were divided into three groups: Group A comprised 16 subjects with clinically healthy gingiva; group B 17 subjects with gingivitis; and group C 17 subjects with gingivitis; and group C 17 subjects with moderate periodontitis (PPD ≥ 5 mm and CAL ≥ 3 mm in at least six sites). Gingival samples were collected and immunohistochemical study was done using CD57 and CD1a antibody. Statistical analysis was done using analysis of variance (ANOVA), followed by Tukey-Kramer multiple comparison for CD1a and Tukey's highly significant difference (HSD) test for CD57.Results and Conclusion:The study showed an inverse relationship between the CD1a+ (langerhans) cells and CD57+ (natural killer) cells. There was a significant increase in CD57+ cells and reduction in CD1a levels as periodontal disease progressed. The significant reduction in CD1a levels in periodontal disease when compared to health could possibly be a result of NK cells down regulating it. Reduction in CD1a levels may result in a low inflammatory response subsequently resulting in tissue destruction.
Introduction. The interdental papilla may be lost or reduced in height, forming black triangles due to various reasons, which gives an unaesthetic appearance when the patient smiles. Various noninvasive and invasive techniques have been used to augment/reconstruct the interdental papilla, to reclaim the pink triangle. The most satisfactory and natural appearance can be obtained by augmenting or reconstructing the lost papilla as the pink esthetics is as important as the white esthetics. Cases. Two female patients and 1 male patient reported to the dental department with the complaint of a small black gap in the gum area between their upper front teeth since 6 months and 1 year, respectively. On examination, the interdental papillae in all three cases were classified as class 1 (Nordland and Tarnow’s). The interdental papilla was augmented surgically by using three different approaches in each case. Result and Conclusion. Postoperatively and after 1-month follow-up, there was a complete fill of the interdental area by the interdental papilla normal (Nordland and Tarnow’s). As a result, the black triangle was successfully converted into a natural pink triangle in all three cases.
Background:The differentiation of naοve T helper (Th) cells towards Th17 and regulatory T cells (Treg) is regulated by the transcription factors retinoic acid related orphan receptor gamma transcription (RORYt) and Forkhead box p3 (Foxp3), respectively. An imbalance in the activity of these transcription factors could result in the dysregulation of Th17/Treg response.Materials and Methods:Total RNA was isolated from gingival tissue obtained from 10 patients, each from periodontally healthy and diseased groups. The gene expression of RORYt and Foxp3 was measured by real-time reverse transcription polymerization chain reaction using total RNA isolates from gingival tissues group when compared to the healthy group, while Foxp3 demonstrated a 6.68 ± 0.03 fold decrease of expression in diseased group when compared to healthy group.Conclusion:Our results indicate a functional imbalance in the Th17/Treg response in periodontal disease group when compared to the periodontally healthy group.
