Hepatitis E is an important public health problem mostly in developing but occasionally also in industrialized countries. Domestic and wildlife animals are considered reservoirs of the hepatitis E virus (HEV). Since no information on the prevalence of autochthonous HEV infections in human and animal in Poland is available, the aim of the study was to investigate the HEV seroprevalence of different wildlife species as potential virus reservoirs in the country. No HEV antibodies were found in any of the sera collected from the red deer (Cervus elaphus), European bison (Bison bonasus), roe deer (Capreolus capreolus), elk (Alces alces), fallow deer (Dama dama), sika deer (Cervus nippon), Tatra chamois (Rupicapra rupicapra tatrica) or brown bear (Ursus arctos). HEV-specific antibodies were detected in 44.4% (95% CI 38.3-50.7) serum samples originated only from wild boars. The percentage of seropositive wild boars differed significantly between the provinces and was positively correlated with the wild boar density and rurality of the area. This study showed that HEV circulates among wild boar population in Poland, and this species should be considered as an important reservoir of the virus.
Antimicrobial resistance was tested in Escherichia coli isolated from feces (n = 660) of red deer, roe deer, fallow deer, European bison, and wild boar shot in regional forests in Poland during two winter hunting seasons. Indicator E. coli (n = 542) was resistant against 11 of 14 tested compounds, mostly sulfamethoxazole, streptomycin, ampicillin, trimethoprim, and tetracycline (1.3-6.6% range). No significant differences were observed between boar and ruminant isolates. Most of deer and bison isolates showed no resistance. Selective screening of wildlife samples revealed 1.7% prevalence of cephalosporin-resistant E. coli found mostly in wild boars. They produced extended-spectrum beta-lactamases (bla, bla) and plasmid-mediated AmpC-type cephalosporinase (bla). The majority of the isolates originated from boars shot in a narrow time frame and space; therefore, common antimicrobial selection pressure in the environment was assumed. Three E. coli isolates carried plasmid-mediated quinolone resistance genes (qnrS1/S3). No transferable colistin resistance mechanisms were found in two resistant E. coli. Transferability of resistance was proved in a single pAmpC-positive isolate carrying IncI1-alpha 95 kb plasmid. No cephalosporin-resistant E. coli harbored pathogenicity markers; therefore, they might be considered a vector of resistance determinants, but not a pathogen themselves.
A series of eleven cymantrene-and cyrhetrene-nucleobase conjugates together with the hitherto unreported N7-isomer of the ferrocene-adenine conjugate have been synthesised and characterized. The synthetic approach involved a Michael addition reaction of in situ generated acryloylcymantrene, acryloylcyrhetrene and acryloylferrocene with canonical nucleobases, such as thymine, uracil and adenine, respectively. The mechanism of these reactions was investigated by means of density functional theory (DFT) calculations.The respective products were characterized by spectroscopic methods and by electrochemical measurements. The molecular structure of one cymantrene-adenine conjugate (5) in the solid state was determined by single-crystal X-ray structure analysis, confirming the N9-substitution mode of the adenine moiety. It was found that the molecule adopts a bent conformation with the adenine and cyclopentadienyl planes in almost perpendicular orientation. The cymantrenyl nucleobases showed an irreversible redox behaviour which is associated with ligand exchange reactions of the radical cationic species. The newly synthesised compounds were also tested for their activity against the protozoan parasite Trypanosoma brucei and human myeloid leukaemia HL-60 cells. Some compounds showed promising antitrypanosomal activity while most of them were non-toxic to HL-60 cells. It was additionally found that cymantrene and cyrhetrene ketone nucleobases were more active than their alcohol congeners. These findings indicate the potential of cymantrene and cyrhetrene nucleobase conjugates as possible lead compounds for future antitrypanosomal drug development.
Background:Recently an increase in the population of wild boars (Sus scrofa) in Europe has been observed. This is important from a zoonotic perspective because it influences directly on the spread of many diseases. For the first time, an extensive survey on the prevalence of Leptospira infections in Polish wild boars was performed. During the hunting season 2012-2014, 3621 blood samples from wild boars were collected. The animals originated from different geographical areas across Poland. Serum samples were tested by a microscopic agglutination test (MAT) for the presence of specific antibodies to the following Leptospira serovars: Icterohaemorrhagiae, Grippotyphosa, Sejroe, Tarassovi, Pomona, Canicola, Bratislava, Autumnalis, Hardjo and Ballum.Results: Antibody titers to all Leptospira serovars except serovar Ballum were found in 377 serum samples (10.4 %). The highest number of seropositive wild boars was found in the south-eastern part of Poland and in highly urbanized areas such as Silesia and Łódź. Conclusions:The relatively high prevalence of Leptospira infections in wild boars may constitute a threat to hunters and people having contact with forest lakes or marshlands. The results also indicate that an increasing population of wild boar living close to borders of cities may create additional risk for inhabitants in large urban areas.
The most important wildlife species in the epidemiology of hepatitis E virus (HEV) infections are wild boars, which are also the main reservoir of the virus in a sylvatic environment. The aim of the study was a serological and molecular assessment of the prevalence of HEV infections in wild boars in Poland. In total, 470 pairs of samples (wild boar blood and livers) and 433 samples of faeces were tested. An ELISA (ID.vet, France) was used for serological analysis. For the detection of HEV RNA, real-time (RT)-qPCR was employed. The presence of specific anti-HEV IgG antibodies was found in 232 (49.4%; 95%CI: 44.7–54%) sera, with regional differences observed in the seroprevalence of infections. HEV RNA was detected in 57 (12.1%, 95%CI: 9.3–15.4%) livers and in 27 (6.2%, 95%CI: 4.1–8.9%) faecal samples, with the viral load ranging from 1.4 to 1.7 × 1011 G.C./g and 38 to 9.3 × 107 G.C./mL, respectively. A correlation between serological and molecular results of testing of wild boars infected with HEV was shown. HEV infections in wild boars appeared to be common in Poland.
A tulathromycin concentration and pharmacokinetic parameters in plasma and lung tissue from healthy pigs and Actinobacillus pleuropneumoniae (App)-infected pigs were compared. Tulathromycin was administered intramuscularly (i.m.) to all pigs at a single dose of 2.5 mg/kg. Blood and lung tissue samples were collected during 33 days postdrug application. Tulathromycin concentration in plasma and lung was determined by high-performance liquid chromatography with tandem mass spectrometry (LC-MS/MS) method. The mean maximum plasma concentration (Cmax ) in healthy pigs was 586 ± 71 ng/mL, reached by 0.5 h, while the mean value for Cmax of tulathromycin in infected pigs was 386 ± 97 ng/mL after 0.5 h. The mean maximum tulathromycin concentration in lung of healthy group was calculated as 3412 ± 748 ng/g, detected at 12 h, while in pigs with App, the highest concentration in lung was 3337 ± 937 ng/g, determined at 48 h postdosing. The higher plasma and lung concentrations in pigs with no pulmonary inflammation were observed at the first time points sampling after tulathromycin administration, but slower elimination with elimination half-life t1/2el = 126 h in plasma and t1/2el = 165 h in lung, as well as longer drug persistent in infected pigs, was found.
BackgroundLeptospira spp. infect humans and a wide range of domestic and wild animals, but certain species such as small rodents and red foxes (Vulpes vulpes) play a particular role as reservoirs and transmission of leptospirosis as they easily adapt to many habitats including human environments. To investigate the significance of red foxes in the epidemiology of leptospirosis in Poland, a seroprevalence survey was conducted. During the 2014–2015 hunting season, blood samples of 2134 red foxes originating from the central-eastern part of Poland were collected. Serum samples were tested by a microscopic agglutination test for the presence of specific antibodies to Leptospira serovars Icterohaemorrhagiae, Grippotyphosa, Sejroe, Tarassovi, Pomona, Canicola, Hardjo, Ballum, Australis, Bataviae, Saxkoebing and Poi.ResultsAntibodies to at least one serovar were detected in 561 sera (26.3%). The highest seroprevalence was found in the Subcarpathia (41.6%) and Warmia-Masuria (40.3%) provinces. Antibodies were mainly directed against serovars Poi (12.4%), Saxkoebing (11.3%), and Sejroe (6.0%).ConclusionsExposure of red foxes to certain Leptospira serovars seems to be common in central and eastern Poland. In addition, the high prevalence of antibodies against Leptospira spp. in foxes may indicate a potential risk of infection for humans and other species coming into contact with these animals.
