Stevioside and rebaudioside A, two intense natural sweeteners, that are constituents of the South American plant Stevia rebaudiana, were tested for cariogenicity in albino Sprague-Dawley rats. Sixty rat pups colonized with Streptococcus sobrinus were divided into four groups and fed stevioside, rebaudioside A or sucrose added to basal diet 2000 as follows: group 1,30% sucrose; group 2, 0.5% stevioside; group 3, 0.5% rebaudioside A, and group 4, no addition. All four groups were sacrificed after 5 weeks. S. sobrinus counts were made and caries was evaluated according to Keyes’ technique. There were no differences in food and water intake and weight gains between the four groups. There were significant differences in sulcal caries scores (p < 0.02) and S. sobrinus counts (p < 0.05) between group 1 and the other three groups. There were no significant differences between the stevioside, rebaudioside A and no-addition groups. It was concluded that neither stevioside nor rebaudioside A is cariogenic uner the conditions of this study.

Germin-like proteins (GLPs) are involved in biotic and abiotic stress tolerance in different plant species. Rice ( Oryza sativa L.) genome contains about 40 GLP family member proteins in nine chromosomes. Although some of the rice GLP ( OsGLP ) promoters have been studied through in silico analysis as well as experimentally, studies regarding the distribution pattern of the biotic and abiotic stress associated transcription factor binding sites (TFbs) in the promoter regions of OsGLP genes have not been attempted thoroughly. Several transcription factors (TFs) namely NAC, WRKY, bHLH, bZIP, MYB and AP2/ERF act as major TFs concerned with biotic as well as abiotic stress responses across various plant species. In the present study the in silico analysis was carried out using the 1.5 kilobases (kb) promoter regions from 40 different OsGLP genes for the presence of NAC, WRKY, bHLH, bZIP, MYB and AP2/ERF TFbs in it. Among various OsGLP gene promoters, OsGLP8-11 was found to contain highest number of tested TFbs in the promoter region whereas the promoter region of OsGLP5-1 depicted least amount of TFbs. Phylogenetic study of promoter regions of different OsGLP genes revealed four different clades. Our analyses could reveal the evolutionary significance of different OsGLP gene promoters. It can be presumed from the present findings as well as previous reports that OsGLP gene duplications and subsequent variations in the TFbs in OsGLP gene promoter regions might be the consequences of neofunctionalization of OsGLP genes and their promoters for biotic and abiotic stress tolerance in rice.

Food legumes are important for defeating malnutrition and sustaining agri-food systems globally. Breeding efforts in legume crops have been largely confined to the exploitation of genetic variation available within the primary genepool, resulting in narrow genetic base. Introgression as a breeding scheme has been remarkably successful for an array of inheritance and molecular studies in food legumes. Crop wild relatives (CWRs), landraces, and exotic germplasm offer great potential for introgression of novel variation not only to widen the genetic base of the elite genepool for continuous incremental gains over breeding cycles but also to discover the cryptic genetic variation hitherto unexpressed. CWRs also harbor positive quantitative trait loci (QTLs) for improving agronomic traits. However, for transferring polygenic traits, “specialized population concept” has been advocated for transferring QTLs from CWR into elite backgrounds. Recently, introgression breeding has been successful in developing improved cultivars in chickpea (Cicer arietinum), pigeonpea (Cajanus cajan), peanut (Arachis hypogaea), lentil (Lens culinaris), mungbean (Vigna radiata), urdbean (Vigna mungo), and common bean (Phaseolus vulgaris). Successful examples indicated that the usable genetic variation could be exploited by unleashing new gene recombination and hidden variability even in late filial generations. In mungbean alone, distant hybridization has been deployed to develop seven improved commercial cultivars, whereas in urdbean, three such cultivars have been reported. Similarly, in chickpea, three superior cultivars have been developed from crosses between C. arietinum and Cicer reticulatum. Pigeonpea has benefited the most where different cytoplasmic male sterility genes have been transferred from CWRs, whereas a number of disease-resistant germplasm have also been developed in Phaseolus. As vertical gene transfer has resulted in most of the useful gene introgressions of practical importance in food legumes, the horizontal gene transfer through transgenic technology, somatic hybridization, and, more recently, intragenesis also offer promise. The gains through introgression breeding are significant and underline the need of bringing it in the purview of mainstream breeding while deploying tools and techniques to increase the recombination rate in wide crosses and reduce the linkage drag. The resurgence of interest in introgression breeding needs to be capitalized for development of commercial food legume cultivars.

Comprehensive sexuality education (CSE) has been recognised globally as key to helping young people assert their sexual and reproductive rights. In India too, there is growing awareness of the importance of providing CSE not only to reduce sexually transmitted infections, unintended pregnancies and abortions but also to teach important life skills. Simultaneously, lack of political will and conflicting interests among certain religious and political factions have ensured that no uniform CSE curriculum has been implemented throughout the country. This paper analyses the Adolescent Education Programme teacher curriculum as revised in 2009-2010 by the National Council of Educational Research and Training and the United Nations Population Fund. It highlights some of the opportunities presented by the curriculum and argues that despite its intent of providing relevant sexuality education for young people, the language of the curriculum is vague, thus potentially exacerbating confusion, and excluding people who do not conform to societal stereotypes of sex, gender, and ability. In order to be holistic, any CSE programme must be inclusive, cater to diverse needs and present content in a rights-based language without adding to the socio-cultural context of mystery and shame attached to sexuality.

Rust caused by Uromyces viciae-fabae is a major biotic constraint to field pea ( Pisum sativum L.) cultivation worldwide. Deployment of host-pathogen interaction and resistant phenotype is a modest strategy for controlling this intricate disease. However, resistance against this pathogen is partial and influenced by environmental factors. Therefore, the magnitude of environmental and genotype-by-environment interaction was assessed to understand the dynamism of resistance and identification of durable resistant genotypes, as well as ideal testing locations for rust screening through multi-location and multi-year evaluation. Initial screening was conducted with 250 diverse genotypes at rust hot spots. A panel of 23 promising field pea genotypes extracted from initial evaluation was further assessed under inoculated conditions for rust disease for two consecutive years at six locations in India. Integration of GGE biplot analysis and multiple comparisons tests detected a higher proportion of variation in rust reaction due to environment (56.94%) as an interactive factor followed by genotype × environment interaction (35.02%), which justified the requisite of multi-year, and multi-location testing. Environmental component for disease reaction and dominance of cross over interaction (COI) were asserted by the inconsistent and non-repeatable genotypic response. The present study effectively allocated the testing locations into various categories considering their “repeatability” and “desirability index” over the years along with “discrimination power” and “representativeness.” “Mega environment” identification helped in restructuring the ecological zonation and location of specific breeding. Detection of non-redundant testing locations would expedite optimal resource utilization in future. The computation of the confidence limit (CL) at 95% level through bootstrapping strengthened the accuracy of the GGE biplot and legitimated the precision of genotypes recommendation. Genotype, IPF-2014-16, KPMR-936 and IPF-2014-13 identified as “ideal” genotypes, which can be recommended for release and exploited in a resistance breeding program for the region confronting field pea rust.

Susceptibility to root-knot nematodes (Meloidogyne spp.) is one of the major factors limiting mungbean production in South and South-east Asia. Host-pest-environment interaction in mungbean and rootknot nematode (M. incognita) was investigated in multi-location field evaluation using 38 promising mungbean genotypes extracted from initial evaluation of 250 genotypes under sick plots considering second stage freshly hatched juvenile as inoculants. the extent of environmental and genotypeby-environment interactions (GGe) was assessed to comprehend the dynamism of resistance and identification of durable resistant mungbean genotypes. Among environmental factors, nematode activity was highly influenced by rainfall and minimum temperature. The GGE biplot and multiple comparison tests detected a higher proportion of genotype × environment (Ge) interaction followed by genotype and environment on number of nematode galls, gall index and reproduction factor. The first two principal components (PCs) explained 64.33% and 66.99% of the total variation of the environment-centered gall scoring and reproduction factor data, respectively. the high Ge variation indicated the presence of non-cross over interactions which justify the necessities of multi-location testing. Detection of non-redundant testing locations would expedite optimum resource utilization in future. The GGE biplot analysis identified genotypes such as PM-10-12, IPM-410-3 and NVL-641 as the outperforming and desirable genotypes with durable resistance against M. incognita which can be exploited in mungbean breeding programmes globally. on the contrary, the highest gall scoring and reproduction factor were recorded in genotype IPM-9901-8.
