Nowadays, zoos and aquariums, along with the constant advancement of sociocultural moral values, are proactively committed to ensuring and safeguarding cetacean health standards. This entails developing new approaches to health assessments by embracing minimally invasive sampling methods and enhanced animal handling and management, among other aspects. Hence, in the present survey, to appraise skin diseases, the implementation of cytology cell samplers as a non-invasive skin sampling device on 18 bottlenose dolphins housed in two facilities in the Canary Islands during the months of April, October, and December 2019 was performed to isolate cetacean poxvirus in tattoo-like lesions through a real-time PCR-based method using the DNA polymerase gene. Samples were repeatedly collected over time from eleven tattoo-like lesions and from apparently healthy skin to serve as a control for all study animals. From a total of 55 skin samples, detection of the poxvirus was attained in 31 (56.36%); specifically, on 20 of 21 samples collected from tattoo-like lesions (95.23%) and on 11 of 34 samples acquired from apparently healthy skin (32.35%). Correspondingly, the current study constitutes the first report of the isolation of cetacean poxvirus in skin samples without macroscopical signs of tattoo lesions in cetaceans. Likewise, ten of the eleven dolphins that showed tattoo lesions housed in Facility 1 were positive for tattoo skin disease, while four dolphins held in Facility 2 were positive for cetacean poxvirus without ever showing clinical evidence of the disease. This raises the question of whether this pathogen can produce latent infections and whether progression of the disease may depend on environmental stimuli, viral load, or the good health/immunological status of individual animals. Accordingly, further scientific research on cetaceans under human care could provide the knowledge, skills, and resources to understand the host–pathogen dynamics of cetacean poxviruses and their effect on cetaceans’ health.
En la presente investigación se aisló una bacteria termófila productora de enzima amilasa, de los géiseres de Calientes, Candarave (Tacna-Perú), la cual por análisis de la secuencia del gen ARNr 16S presentó un 99 % de identidad con Bacillus licheniformis. Para determinar la capacidad amilolítica se evaluó en medio sólido presentando un halo de hidrólisis de 29,5 mm de diámetro a las 72 horas. En la evaluación de la producción de amilasa se obtuvo una concentración máxima de 1410,57µg/mL de azúcares reductores a inicios de la fase estacionaria a 54,6 horas de incubación a 60 °C y pH 7. El extracto amilolítico tuvo un grado de purificación de 2,3 veces con sulfato de amonio, actuando a una temperatura óptima de 58 °C con una actividad de 10,7 U/mL ya un pH óptimo de 6,7 con una actividad de 8,3 U/mL, además la velocidad máxima (Vm) fue de 229,5µg de almidón hidrolizado por ml de enzima por minuto y el valor de la constante de Michaelis (Km) fue de 8,7 mg de almidón por mL enzima.
El presente trabajo estudia las proteasas alcalinas producidas por dos bacterias termófilas designadas como BP-2 y BP-4. Estas fueron aisladas de sedimentos de los géiseres de Candarave, mostrando capacidad proteolítica en medio sólido. La cepa BP-2 mostró un diámetro de 46 mm del halo de hidrólisis a 50 °C, mientras que la cepa BP-4 mostró un diámetro de 85 mm del halo de hidrólisis a 60°C. Las bacterias seleccionadas BP-2 y BP-4 degradaron 2,9 g/L de caseína en un tiempo estimado de 51 horas de incubación y 1,5 g/L de caseína en 52 horas de incubación, respectivamente (concentración inicial de 10 g/L de caseína). La máxima actividad proteolítica se alcanzó al final de la fase logarítmica y comienzo de la fase estacionaria en las dos bacterias. Los extractos proteolíticos producidos por las bacterias BP-2 y BP-4 actuaron a una temperatura óptima estimada de 57°C y 64°C, con un pH óptimo alcalino de 7,7 y 8,0 respectivamente. Del análisis de las secuencias del gen ARNr 16S de cada bacteria se determinó que la bacteria BP-2 tiene 99% de identidad con la especie Bacillus licheniformis y la bacteria BP-4 tiene un 99% de identidad con la especie Geobacillus thermaparaffinivorans.
Para esta investigación, se estudió la cinética de la enzima celulasa producida por la bacteria Bacillus sp GCB-13, aislada de los géiseres de Candarave. La bacteria alcanzó una fase logarítmica a partir de las 24 horas de incubación. La producción enzimática fue desarrollada usando CMC como sustrato, determinándose la concentración de azúcares reductores mediante el método del Ácido Dinitrosalicílico (DNS) y la evaluación de proteínas totales con el método de Bradford. El extracto enzimático crudo obtenido fue aplicado sobre sustratos lignocelulósicos naturales como tallos y hojas del geranio (Pelargonium x hortorum), y sobre sustratos sintéticos como papel bond y Carboximetilcelulosa (CMC), para evaluar su potencial de degradación. Es así que se obtuvieron los valores de azúcares reductores de 3.499 mg/ml; 0.116 mg/ml, 0.083 mg/ml y 0.593 mg/ml. Los parámetros cinéticos fueron calculados empleando la ecuación de Eadie-Hofstee, obteniendo un valor de Km y V de 3.43 μM de CMC y 0.012 máx mg/ml.min, respectivamente. Finalmente, se evaluaron las actividades enzimáticas a diferentes valores de pH y temperaturas, correspondiendo los mayores niveles de actividades de 4.4 U/ml a 45 °C a pH 7.0, 3.04 U/ml a 45 °C a pH 5.0 y 3.6 U/ml a 45 °C a pH 10.
La investigación tuvo como objetivo determinar el diámetro del halo de hidrólisis amilolítico, la roducción de proteínas amilásicas y la actividad enzimática de las amilasas producidas por Geobacillus thermoparaffinivorans (CB-13) a diferentes tiempos de producción. La cepa amilolítica de Geobacillus thermoparaffinivorans (CB-13), aislada de los géiseres de Candarave, Tacna se reactivó e incubó en caldo Luria Bertani (LB) a 60 °C durante 24 horas y se sembró por puntura en agar almidón 1 % para determinar los halos de hidrólisis amilolíticos. Posteriormente, se inoculó 5 % (v/v) de medio caldo LB con crecimiento bacteriano en 50 ml de medio de caldo almidón 1 % por triplicado, después fue incubado en estufa con agitador orbital a 150 rpm a 60 °C durante 60 horas. Asimismo, se extrajeron de cada incubación, con periodos de tiempo de 12 horas, alícuotas de 1.5 ml para ser centrifugadas y obtener el sobrenadante que se utilizó para determinar la concentración de proteínas (mg ml-1) como indicativo de la producción de amilasas, y la concentración de azúcares reductores como indicativo de la actividad enzimática (U ml-1). Se obtuvo un diámetro de halo de hidrólisis de almidón formado alrededor de la colonia de la cepa amilolítica de 10.6 mm, una producción de amilasas máxima de 0.197 mg ml-1 a las 48 horas de producción y una actividad enzimática máxima a las 36 horas de producción de 0.505 U ml-1.
