Sixty-three Escherichia coli strains isolated from broilers with respiratory problems were examined for virulence factors, hemolysin synthesis ability, motility, hemagglutination capacity, operon pap presence, colicin production, and serum resistance. The capacity to hemagglutinate guinea pig erythrocytes was found in 53 (84.1%) of the samples, but only 30 (47.6%) agglutinated chicken erythrocytes. D-mannose-sensitive hemagglutination against guinea pig erythrocytes was found in 19 (30.2%) samples and against chicken erythrocytes, in 15 (23.8%) samples, whereas the D-mannose-resistant hemagglutination with guinea pig erythrocytes was found in 34 (54%) samples, and 13 of these (20.6%) showed this characteristic against chicken erythrocytes. Operon pap, P fimbria codifier, was detected in 26 samples in a total of 34 D-mannose-resistant samples. Colicin production was observed in 55 (87.3%) of the strains, and 41.8% presented V colicin production. Of the samples analyzed, 56 (88.9%) presented serum resistance, six (9.5%) were intermediate, and only one (1.6%) was sensitive to the action of the complement. The diversity of virulence profiles detected in the samples in this study explains in part the multifactorial characteristics of avian colibacillosis.
Resumo: Com o objetivo de identificar a patogenicidade e resistência a antimicrobianos da cepa de Escherichia coli BK125, foram utilizados os seguintes testes: sorotipagem, aglutinação em lâmina para detecção da fímbria F18, PCR para verificar a presença do gene Stx2e e teste de citoxicidade para avaliar a expressão da verotoxina, ensaio para detecção de hemolisinas, patogenicidade em leitões e antibiograma. A cepa BK125 apresentou o seguinte perfil: F18 + , Stx2e + , Hly + , resistente a estreptomicina, tetraciclina, sulfonamida e foi capaz de provocar a doença clínica e morte em leitões inoculados. Também foi possível o resgate dessa cepa de fezes diarréicas e do conteúdo intestinal dos leitões revelando assim, alto índice de recuperação de colônias inoculadas. Os resultados permitem concluir que a E. coli BK125 (O139:K82) é produtora de fatores de virulência e reproduz experimentalmente a doença do edema em suínos. As aflatoxinas prejudicam os parâmetros de produção, causam imunodepressão humoral e celular e afetam o mecanismo de coagulação. No sul do Brasil, a aflatoxicose representou de 15 a 29% dos casos examinados em laboratório de 1985 a 1991, sendo uma das principais enfermidades diagnosticadas no período em questão.Frangos de corte com 42 dias de idade, fêmeas, com peso médio de 1.800 g foram inoculados diretamente no inglúvio com 360 mg de aflatoxina B 1 através de dose única. Aos 30 minutos, 1, 2, 5, 8, 12 e 24 horas após a inoculação, cinco animais tratados e quatro controles foram sacrificados e coletados 40g de fígado, de cada um, para serem processados individualmente. Usou-se o "kit" comercial Veratox da Neogen Co. que emprega o ensaio imunoenzimático utilizando anticorpos monoclonais (ELISA). Nos fígados, houve diferenças significativas (P<0,05) entre as médias dos inoculados e controles em todos os intervalos p.i. Nas primeiras duas horas p.i. 46,6% da toxina foi detectada neste órgão. O método pode ser útil para o diagnóstico da aflatoxicose das aves de maneira simples, rápida e econômica, eliminando, assim, os inconvenientes relativos à amostragem e disponibilidade do alimento quando se buscam aflatoxinas em rações e matérias-primas.
