Salinity stress is a barrier to crop production, quality yield, and sustainable agriculture. The current study investigated the plant growth promotion, biochemical and molecular characterization of bacterial strain Enterobacter cloacae PM23 under salinity stress (i.e., 0, 300, 600, and 900 mM). E. cloacae PM23 showed tolerance of up to 3 M NaCl when subjected to salinity stress. Antibiotic-resistant Iturin C (ItuC) and bio-surfactant-producing genes (sfp and srfAA) were amplified in E. cloacae PM23, indicating its multi-stress resistance potential under biotic and abiotic stresses. Moreover, the upregulation of stress-related genes (APX and SOD) helped to mitigate salinity stress and improved plant growth. Inoculation of E. cloacae PM23 enhanced plant growth, biomass, and photosynthetic pigments under salinity stress. Bacterial strain E. cloacae PM23 showed distinctive salinity tolerance and plant growth-promoting traits such as indole-3-acetic acid (IAA), siderophore, ACC deaminase, and exopolysaccharides production under salinity stress. To alleviate salinity stress, E. cloacae PM23 inoculation enhanced radical scavenging capacity, relative water content, soluble sugars, proteins, total phenolic, and flavonoid content in maize compared to uninoculated (control) plants. Moreover, elevated levels of antioxidant enzymes and osmoprotectants (Free amino acids, glycine betaine, and proline) were noticed in E. cloacae PM23 inoculated plants compared to control plants. The inoculation of E. cloacae PM23 significantly reduced oxidative stress markers under salinity stress. These findings suggest that multi-stress tolerant E. cloacae PM23 could enhance plant growth by mitigating salt stress and provide a baseline and ecofriendly approach to address salinity stress for sustainable agriculture.
Resumo -Com o intento de selecionar progênies superiores para a efetiva concentração de alelos favoráveis em uma população de quinto ciclo de seleção recorrente intrapopulacional em famílias de irmãos-completos de milho pipoca, avaliaram-se 200 famílias quanto a seis características agronômicas, incluindo rendimento de grãos e capacidade de expansão, em delineamento aleatorizado em blocos com duas repetições dentro de 'sets' em dois ambientes distintos: Colégio Estadual Agrícola Antônio Sarlo, em Campos dos Goytacazes, e Estação Experimental da PESAGRO-RIO, em Itaocara, Rio de Janeiro, em 2007/2008. Houve diferenças significativas para famílias/"sets" para todas as características, indicando a presença de variabilidade genética a ser explorada nos futuros ciclos. Na seleção das progênies superiores o índice de Mulamba e Mock resultou em ganhos proporcionais mais adequados para capacidade de expansão e rendimento de grãos, com valores respectivos de 6,01 e 8,53%, na utilização de pesos econômicos arbitrários. Melhoramento genético.Abstract -Aiming to select superior progenies for concentration of favorable alleles in a popcorn population on the fifth cycle of intrapopulation full-sib recurrent selection, two hundred progenies were evaluated for six agronomic traits, including grain yield and popping expansion, in randomized complete block design with two replications within sets, in two distinct environments: Colégio Estadual Agrícola Antônio Sarlo, in Campos dos Goytacazes, and Experimental Station of PESAGRO-RIO, located in Itaocara, Rio de Janeiro State, in 2007/2008. There were significant differences among families within sets in all traits, indicating the presence of genetic variability to be exploited in the future cycles. The utilized selection indexes were the Mulamba and Mock; and Smith and Hazel. The Mulamba and Mock index propitiated more elevated magnitudes of the predicted gains for most of the traits, including popping expansion and grain yield, with values of 6.01 and 8.53%, respectively, utilizing arbitrary weight attributed by tentative.
ABSTRACT. Using only one type of marker to quantify genetic diversity generates results that have been questioned in terms of reliability, when compared to the combined use of different markers. To compare the efficiency of the use of single versus multiple markers, we quantified genetic diversity among 10 S 7 inbred popcorn lines using both RAPD and SSR markers, and we evaluated how well these two types of markers discriminated the popcorn genotypes. These popcorn genotypes: "Yellow Pearl Popcorn" (P1-1 and P1-5), "Zélia" (P1-2 and P1-4), "Curagua" (P1-3), "IAC 112" (P9-1 and P9-2), "Avati Pichinga" (P9-3 and P9-5), and "Pisankalla" (P9-4) have different soil and climate adaptations. Using RAPD marker analysis, each primer yielded bands of variable intensities that were easily detected, as well as non-specific bands, which were discarded from the analysis. The nine primers used yielded 126 bands, of which 104 were classified as polymorphic, giving an average of 11.6 polymorphisms per primer. Using SSR procedures, the number of alleles per locus ranged from two to five, giving a total of 47 alleles for the 14 SSR loci. When comparing the groups formed using SSR and RAPD markers, there were similarities in the combinations of genotypes from the same genealogy. Correlation between genetic distances obtained through RAPD and SSR markers was relatively high (0.5453), indicating that both techniques are efficient for evaluating genetic diversity in the genotypes of popcorn that we evaluated, though RAPDs yielded more polymorphisms.
Popcorn breeding programs in Brazil have emerged but despite some advances there is still a lack of material performance studies specially regard to seed quality, in different agricultural seasons. This research was carried out to estimate the popcorn combining ability from biparental crosses between eight tropical and one temperate lines in two agricultural seasons (2008 and 2009) with regard to eight traits related to seed quality, in addition to three agronomic traits related to popping expansion. Method 4, proposed by Griffing (Model 1), was used to verifying the importance of the dominance effect in the genetic control of the seed quality traits, favouring the identification of superior hybrids through seed tests in the laboratory with low cost. The lines from Zélia and CMS42 were promising in obtaining hybrids with superior seed quality and greater popping expansion. The hybrids Zélia 9 IAC-112, CMS42 9 UEM M2-1, CMS43-1 9 IAC-112 and UEM M2-2 9 Zaeli expressed superior SCA for grain yield, popping expansion and at least two seed quality traits. These hybrids are materials of interest for cultivation in different planting seasons.
Com o intento de selecionar progênies superiores para a efetiva concentração de alelos favoráveis em uma população de terceiro ciclo de seleção recorrente intrapopulacional em famílias de meios-irmãos de milho-pipoca, avaliaram-se 192 progênies em delineamento em blocos casualizados com duas repetições em dois ambientes distintos: Colégio Estadual Agrícola Antônio Sarlo, em Campos dos Goytacazes, e Estação Experimental da PESAGRO-RIO, em Itaocara, Rio de Janeiro, em 2004/05. Definiu-se a intensidade de seleção de 15,6%, correspondendo ao número de trinta famílias de meios-irmãos para continuidade do programa. Foram empregados os índices de seleção de Smith e Hazel, Pesek e Baker, Williams, e Mulamba e Mock. O índice de Mulamba e Mock proporcionou as magnitudes mais elevadas de ganhos preditos para a maioria das características, incluindo-se capacidade de expansão e produção, com valores respectivos de 7,16% e 10,00%, na utilização de pesos arbitrários atribuídos por tentativas.
E studos de divergência genética são importantes para o conhecimento da variabilidade genética das populações e possibilitam o monitoramento de bancos de germoplasmas (Cruz e Carneiro, 2003), pois geram informações úteis para preservação e uso dos acessos (Toquica et al., 2003 et al., 1998). Marshall (1989), estimou que 0,5% dos acessos de um banco devem ser requisitados por ano, e considerou que este seria o uso mínimo desejável. Porém, isto dificilmente acontece, e segundo Li et al. (1998), os fatores que dificultam o uso do banco de germoplasma são a não disponibilidade dos acessos aos melhoristas, a falta de conhecimento sobre os acessos conservados e o não conhecimento dos curadores sobre os programas realizados pelos melhoristas. Hammer (2003), afirma que um banco de germoplasma não pode ser um museu, devendo-se estimular a avaliação dos acessos por meio de análises genéticas.Para determinar quão distante geneticamente uma população ou genótipo é de outra são utilizados métodos biométricos, onde se quantifica ou se estima a heterose, que são analisados pela estatística multivariada permitindo unificar múltiplas informações de um conjunto de caracteres. Vários métodos podem ser utilizados, dentre eles estão a análise por componentes principais, variáveis canônicas e métodos aglomerativos. A escolha do método RESUMOTécnicas multivariadas foram utilizadas para avaliar a divergên-cia genética entre 56 acessos da coleção de germoplasma de Capsicum spp. da UENF. Foram utilizados onze descritores quantitativos propostos pelo International Plant Genetic Resources Institute, em um experimento conduzido em condições de campo, em Campos dos Goytacazes (RJ), no delineamento de blocos ao acaso com três repetições e 10 plantas por parcela. A distância generalizada de Mahalanobis (D 2 ) foi utilizada como medida de dissimilaridade. Foram aplicadas variáveis canônicas, método hierárquico do vizinho mais próximo, método de otimização de Tocher e projeção das distâncias no plano. As variáveis avaliadas foram comprimento e diâmetro do fruto, número de sementes por fruto, peso médio do fruto, altura de planta, diâmetro da copa, peso de 1000 sementes, dias para florescimento, dias para frutificação, número de frutos por planta e peso de frutos por planta. Houve diferença significativa entre os acessos para todos os descritores avaliados. Observou-se concordância entre todas as técnicas multivariadas utilizadas e foi possível separar os acessos em oito grupos distintos, indicando a existência de variabilidade genética entre os acessos. A maior distância generalizada de Mahalanobis foi 266,42. Observou-se que acessos têm potencial para serem utilizados como genitores em cruzamentos para obtenção de progênies com alta heterose. Pela análise das variáveis canônicas observou-se que os cruzamentos com maior potencial heterótico seriam 56x43, 34x08 e 59x41.Palavras-chave: Capsicum spp., germoplasma, variáveis canônicas, método de Tocher, projeção das distâncias no plano, recursos genéticos.
The development of cultivars with an improved nitrogen use efficiency (NUE) together with the application of plant growth-promoting bacteria is considered one of the main strategies for reduction of fertilizers use. In this sense, this study: i) evaluated the effect of Azospirillum brasilense on the initial development of maize genotypes; ii) investigated the influence of A . brasilense inoculation on NUE under nitrogen deficit; and iii) sought for more NUE genotypes with higher responsiveness to A . brasilense inoculation. Twenty-seven maize genotypes were evaluated in three independent experiments. The first evaluated the initial development of maize genotypes with and without A . brasilense (strain Ab-V5) inoculation of seeds on germination paper in a growth chamber. The second and third experiments were carried out in a greenhouse using Leonard pots and pots with substrate, respectively, and the genotypes were evaluated at high nitrogen, low nitrogen and low nitrogen plus A . brasilense Ab-V5 inoculation. The inoculation of seeds with A . brasilense Ab-V5 intensified plant growth, improved biochemical traits and raised NUE under nitrogen deficit. The inoculation of seeds with A . brasilense can be considered an economically viable and environmentally sustainable strategy for maize cultivation.
In plant breeding, correlations between the statistics of stability and adaptability of popcorn cultivars are not yet well understood. Therefore, the objectives of the present experiment was to investigate the correlations between r 2 di and b i from Eberhart and Russell, x i from Wricke, S ð1Þ i , S ð2Þ i and S ð3Þ i from Huehn, P i from Lin and Binns and the rank-sum from Kang, and indicate the most reliable method for selecting popcorn cultivars.
