This study sought to chart the ontogenesis of the goat rumen by histomorphometric examination, scanning electron microscopy and immunohistochemical analysis. A total of 140 goat embryos and fetuses were used, from the first stage of prenatal life until birth. The appearance of the rumen from the primitive gastric tube was observed at 35 days of prenatal life (CRL 3 cm, 23% gestation). By 38 days (CRL 4.3 cm CRL, 25% gestation) the ruminal wall comprised three layers: an internal epithelial layer, a middle layer of pluripotential blastemic tissue and an external layer or serosa. Ruminal pillars were visible at 46 days (CRL 6 cm, 30% gestation), and by 76 days (CRL 18 cm, 50% gestation) ruminal papillae were starting to appear. Under scanning electron microscopy, by 50 days (CRL 7.7 cm, 33% gestation) small ruminal papillae were observed protruding from the surface. Finally, neuroendocrine cells (synaptophysin, SYP) were detected at 53 days (CRL 9 cm CRL, 35% gestation), while glial cell markers (glial fibrillary acidic protein-GFAP, and vimentin-VIM) were found at 108 days (CRL 31 cm, 72% gestation) and 39 days (CRL 4.4 cm, 26% gestation), respectively. Neuropeptide Y (NPY) and vasoactive intestinal polypeptide (VIP) were detected immunohistochemically at 113 days (CRL 33 cm, 75% gestation) and 120 days (CRL 35 cm, 80% gestation), respectively. In conclusion, histomorphogenesis of the rumen in goats was similar to that reported in deer, but rather slower than observed for sheep or cattle. Anat Rec, 295:776-785, 2012. ABSTRACT:The main objective of this study is to estimate the probable maximum precipitation (PMP) in Barcelona for durations ranging from 5 min to 30 h. To this end, rain records from the Jardí gauge of the Fabra Observatory located in Barcelona (1927Barcelona ( -1992) and the urban pluviometric network supported by Clavegueram de Barcelona, S.A. (CLABSA, 1994(CLABSA, -2007 were analysed. Two different techniques were used and compared: a physical method based on the maximization of actual storms, and the Hershfield' statistical method. The PMP values obtained using the two techniques are very similar. In both cases, the expected increasing behaviour of the PMP with duration was found, with the increase especially notable for the mesoscale durations 2-9 h, and not significant from 12 h on up. This result seems to be related to the scale of the meteorological situations producing high intense rainfall amounts over our territory.
The functionality of the placenta may affect neonatal adiposity and fetal levels of key nutrients such as long-chain polyunsaturated fatty acids. Fetal macrosomia and its complications may occur even in adequately controlled gestational diabetic (GDM) mothers, suggesting that maternal glycemia is not the only determinant of fetal glycemic status and wellbeing. We studied in vivo the placental transfer of fatty acids (FA) labeled with stable isotopes administered to 11 control and 9 GDM pregnant women (6 treated with insulin). Subjects received orally <sup>13</sup>C-palmitic, <sup>13</sup>C-oleic, and <sup>13</sup>C-linoleic acids and <sup>13</sup>C-docosahexaenoic acid (<sup>13</sup>C-DHA) 12 h before an elective caesarean section. FA were quantified by gas chromatography and <sup>13</sup>C enrichments by gas chromatography-isotope ratio mass spectrometry. The <sup>13</sup>C-FA concentration was higher in total lipids of maternal plasma in GDM patients versus controls, except for <sup>13</sup>C-DHA. Moreover, <sup>13</sup>C-DHA showed a lower placenta/maternal plasma ratio in GDM patients versus controls and a significantly lower cord/maternal plasma ratio. Other FA ratios studied were not different between GDM and controls. A disturbed <sup>13</sup>C-DHA placental uptake occurred in GDM patients treated with diet or insulin, while the latter also had lower <sup>13</sup>C-DHA levels in the venous cord. The tracer study pointed towards an impaired placental DHA uptake as a critical step, while the transfer of other <sup>13</sup>C-FA was less affected. Patients with GDM treated with insulin could also have a greater fetal fat storage, which may have contributed to the reduced <sup>13</sup>C-DHA in the venous cord observed. The DHA transfer to the fetus was reduced in GDM pregnancies compared to controls. This might have an influence on fetal neurodevelopment and long-term consequences for the child.
Oxidative stress is considered a major mechanism causing sperm damage during cryopreservation and storage, and underlies male factor infertility. Currently, oxidative stress is no longer believed to be caused only by the overproduction of reactive oxygen species, but rather by the deregulation of redox signaling and control mechanisms. With this concept in mind, here, we describe for the first time the presence of the soluble carrier family 7 member 11 (SLC7A11) antiporter, which exchanges extracellular cystine (Cyss) for intracellular glutamate, in stallion spermatozoa, as well as its impact on sperm function using the specific inhibitor sulfasalazine. Spermatozoa incubated with Cyss exhibited an increased intracellular GSH content compared with controls (P < 0.01): 50% in fresh extended stallion spermatozoa and 30% in frozen-thawed spermatozoa. This effect was prevented by the addition of sulfasalazine to the media. Cystine supplementation also reduced the oxidation–reduction potential of spermatozoa, with sulfasalazine only preventing this effect on fresh spermatozoa that were incubated for 3 h at 37°C, but not in frozen-thawed spermatozoa. While sulfasalazine reduced the motility of frozen-thawed spermatozoa, it increased motility in fresh samples. The present findings provide new and relevant data on the mechanism regulating the redox status of spermatozoa and suggest that a different redox regulatory mechanism exists in cryopreserved spermatozoa, thus providing new clues to improve current cryopreservation technologies and treat male factor infertility.
Lymphoma is a very common lymphoid neoplasm in domestic animals, but few naturally occurring cases have been reported in rabbits. It presents at different sites within rabbits and, although the macroscopic pattern tends to be similar, different cell populations may be involved. This report describes a case of spontaneous lymphoma ocurring in a two-and-a-half-year-old pet Dutch dwarf rabbit. T and B lymphocyte infiltrates were observed in skin, lung, kidney, liver, intestine and lymph nodes, in each case affecting one or more tissue structures. The diagnosis, based on microscopic and immunocytochemical findings, was multicentric, T cell-rich B cell lymphoma with skin involvement.
