Extracellular plaques of β-amyloid (Aβ) and intraneuronal neurofibrillary tangles made from tau are the histopathological signatures of Alzheimer's disease (AD). Plaques comprise Aβ fibrils that assemble from monomeric and oligomeric intermediates, and are prognostic indicators of AD. Despite the significance of plaques to AD, oligomers are considered to be the principal toxic forms of Aβ 1,2 . Interestingly, many adverse responses to Aβ, such as cytotoxicity 3 , microtubule loss 4 , impaired memory and learning 5 , and neuritic degeneration 6 , are greatly amplified by tau expression. A major obstacle to pre-symptomatic diagnosis and disease-modifying therapy for Alzheimer's disease (AD) is inadequate understanding of molecular mechanisms of AD pathogenesis. For example, impaired brain insulin signaling is an AD hallmark, but whether and how it might contribute to the synaptic dysfunction and neuron death that underlie memory and cognitive impairment has been mysterious. Neuron death in AD is often caused by cell cycle re-entry (CCR) mediated by amyloid-β oligomers (AβOs) and tau, the precursors of plaques and tangles. We now report that CCR results from AβO-induced activation of the protein kinase complex, mTORC1, at the plasma membrane and mTORC1-dependent tau phosphorylation, and that CCR can be prevented by insulin-stimulated activation of lysosomal mTORC1. AβOs were also shown previously to reduce neuronal insulin signaling. Our data therefore indicate that the decreased insulin signaling provoked by AβOs unleashes their toxic potential to cause neuronal CCR, and by extension, neuron death.
The mechanisms of mitochondrial dysfunction in Alzheimer's disease are incompletely understood. Using two‐photon fluorescence lifetime microscopy of the coenzymes, NADH and NADPH, and tracking brain oxygen metabolism with multi‐parametric photoacoustic microscopy, we show that activation of lysosomal mechanistic target of rapamycin complex 1 (mTORC1) by insulin or amino acids stimulates mitochondrial activity and regulates mitochondrial DNA synthesis in neurons. Amyloid‐β oligomers, which are precursors of amyloid plaques in Alzheimer's disease brain and stimulate mTORC1 protein kinase activity at the plasma membrane but not at lysosomes, block this Nutrient‐induced Mitochondrial Activity (NiMA) by a mechanism dependent on tau, which forms neurofibrillary tangles in Alzheimer's disease brain. NiMA was also disrupted in fibroblasts derived from two patients with tuberous sclerosis complex, a genetic disorder that causes dysregulation of lysosomal mTORC1. Thus, lysosomal mTORC1 couples nutrient availability to mitochondrial activity and links mitochondrial dysfunction to Alzheimer's disease by a mechanism dependent on the soluble building blocks of the poorly soluble plaques and tangles.

This novel control mechanism can trans-modulate EGFR function by heterologous stimuli of PLD.
Galectins have been implicated in T cell homeostasis playing complementary pro-apoptotic roles. Here we show that galectin-8 (Gal-8) is a potent pro-apoptotic agent in Jurkat T cells inducing a complex phospholipase D/phosphatidic acid signaling pathway that has not been reported for any galectin before. Gal-8 increases phosphatidic signaling, which enhances the activity of both ERK1/2 and type 4 phosphodiesterases (PDE4), with a subsequent decrease in basal protein kinase A activity. Strikingly, rolipram inhibition of PDE4 decreases ERK1/2 activity. Thus Gal-8-induced PDE4 activation releases a negative influence of cAMP/protein kinase A on ERK1/2. The resulting strong ERK1/2 activation leads to expression of the death factor Fas ligand and caspase-mediated apoptosis. Several conditions that decrease ERK1/2 activity also decrease apoptosis, such as anti-Fas ligand blocking antibodies. In addition, experiments with freshly isolated human peripheral blood mononuclear cells, previously stimulated with anti-CD3 and anti-CD28, show that Gal-8 is pro-apoptotic on activated T cells, most likely on a subpopulation of them. Anti-Gal-8 autoantibodies from patients with systemic lupus erythematosus block the apoptotic effect of Gal-8. These results implicate Gal-8 as a novel T cell suppressive factor, which can be counterbalanced by functionblocking autoantibodies in autoimmunity.
Our aim was to analyze the incidence of mutations in BRCA1 and BRCA2 genes in 54 families with breast/ovarian cancer. Families were selected from three Institutions following the standard criteria for hereditary breast/ovarian cancer. PCR amplification of all exons was performed, followed by SSCP, heteroduplex, PTT and sequencing analysis. We identified eight truncation mutations, three in the BRCA1 gene and five in the BRCA2 gene. Three of these mutations have not been reported previously by other groups: 308insA in one family, 3936 C>T in two families, for BRCA1, and 4970insTG in one family for BRCA2. In addition two families having Ashkenazi Jewish ancestors present the well known mutations 185delAG and 6174delT. Interestingly, 5 out of 11 families have mutations recurrent in Spanish families. Among the 54 families selected, seven have breast and ovary cancer cases, and only two presented a mutation in BRCA1 or BRCA2 genes. Other cancers as prostate and stomach are frequent among relatives carrying the mutation. Five cases of very early onset (<31 years old) breast cancer were detected. The frequencies of BRCA1 (0.074) and BRCA2 (0.13) mutations in our families is low but similar to the incidence found in other populations, like in Spain. Since is widely known that risk factors that modulate the development of breast cancer such as lifestyle risk factors, geographic location, country of origin and socioeconomic status, besides a familial history of breast cancer our findings suggest that the history of colonization and immigrations is very relevant when studying hereditary factors associated to breast cancer.
Resumo: Este estudo insere-se no campo da Responsabilidade Social Empresarial -RSE. Procurouse analisar o desempenho socioambiental da Petrobras, através do coeficiente de correlação entre o faturamento líquido e os investimentos em ações sociais e ambientais. Os dados foram coletados através dos balanços sociais da empresa no modelo proposto pelo Instituto Brasileiro de Análises Sociais e Econômicas -IBASE, entre o período de 1998 e 2009. Metodologicamente, trata-se de uma pesquisa documental e descritiva, com uma abordagem quantitativa. Os resultados indicaram que existe uma correlação muito forte entre a receita líquida e os investimentos socioambientais, o que significa que a empresa possui um alto desempenho socioambiental, estando este fortemente relacionado com o desempenho financeiro. Porém, a associação mais forte encontrada em relação à receita líquida, foi quanto aos indicadores sociais internos e externos, com o coeficiente de correlação r igual a 0,982 e 0,977, respectivamente, enquanto o coeficiente de correlação encontrado na relação com indicador ambiental resultou em r igual a 0,722. Considerando-se por partes, o desempenho social da empresa é maior que o seu desempenho ambiental. Palavras-chave:Petrobras. Balanço Social. Desempenho Socioambiental. The aim was to examine the social and environmental performance of Petrobras, though the correlation coefficient between net income and social and environmental investments. The data were collected through the company's social balance sheets in the model proposed by the Instituto Brasileiro de Análises Sociais e Econômicas-IBASE, between 1998 and 2009. The results indicated that there is a strong correlation between net income and their social and environmental investments, which means that the company has a high social and environmental performance, the latter being strongly related to financial performance. However, the strongest association found in relation to net revenues was related to internal and external social indicators, with the correlation coefficient r equal to 0.982 and 0.977, respectively, while the correlation coefficient in relation to environmental indicator resulted in r equal to 0.722.
O objetivo deste estudo foi analisar a percepção do usuário do Comitê de Ética em Pesquisa (CEP) acerca da submissão do Protocolo de Pesquisa. Esse é um estudo exploratório-descritivo com abordagem qualitativa realizado no CEP. Os sujeitos foram os estudantes de graduação e pós-graduação usuários do CEP. Utilizamos entrevista semi-estruturada e análise do discurso do sujeito coletivo. Verificou-se que a maioria dos sujeitos desconhece o processo de construção da folha de rosto e não realiza leitura da Resolução CNS 196 de 1996. Embora submetam os trabalhos por obrigatoriedade da Universidade, reconhecem que o CEP proporciona respaldo científico para a pesquisa.
