Kappaphycus alvarezii (Doty), a red seaweed, is the most widely cultivated commodity in Indonesia. However, there is a decline in the cultivation production in the last three years. One way to overcome this problem is by improving the quality of seedlings through a development of farming. Efforts made from these are to develop farming techniques using different planting distances (PD) treatments. This study aimed to determine the effect of different PD on growth and carrageenan yield of K. alvarezii using seedlings produced from mass selection combined with tissue-cultured method. This research was conducted in Marobo coastal waters, Bone District, Muna Regency, SE Sulawesi, Indonesia. This study used a Completely Randomized Design (CRD) with three treatments based on different PD and five replications. The treatments were A: 10 cm PD, B: 15 cm PD, and C: 20 cm PD. The results show that the daily growth rate (DGR) of all treatments tended not to differ significantly. For 10 cm PD, the DGR tended to be higher and significantly different from the other two treatments, 15 cm and 20 cm. DGR averagely were 6.00±1.19%/day, 5.36±1.17%/day, and 5.27±1.62%/day for 10, 15, and 20 cm PD, respectively. The ratio of fresh weight (FW) and dry weight (DW) showed no significant differences among treatments. Ratio of FW:DW treatments of 10 cm, 15 cm, and 20 cm were 8.34:1, 9.58:1, and 8.98:1, respectively. For carrageenan yield, the treatment of 20 cm PD showed highest yield and it was significantly different from other treatments. Seaweed diseases and epiphytes found during this study were ice-ice and epiphytes (Sargassum polychystum).

The purpose of this study was to determine the effect of different seedling sources on growth and carrageenan yield of K. alvarezii. There were three various seedlings sources used in this study: local strain, tissue-cultured seedlings, and seedlings resulting from mass selection combined with tissue–cultured method called “prof’. This research was over three months (August-October 2018) in Marobo coastal waters, Bone District, Muna Regency SE Sulawesi using the longline method. The results showed that the daily growth rate (DGR) during this study tended to fluctuate among treatments “prof’ and tissue-cultured seedlings had higher the DGRs than local strain seedlings. Ratio fresh weight (FW) and dry weight (DW) showed no significant differences among different seedling sources. The mean FW: DW ratio showed the local strain had the highest ratio (10.10:1) followed by tissue-cultured seedlings (9.88:1) and prof (9.06:1). Furthermore, the local strain showed a significant difference in carrageenan yield from the “prof’ and tissue-culture seedlings. Local strain seedlings showed the highest carrageenan yield (40.58±2.51%) followed by tissue-cultured seedling (35.35±2.79%) and prof seedlings (33.88±2.09%). Ice-ice disease and epiphyte (Sargassum polychystum) were found during this study. This study showed that prof and tissue-cultured seedlings greater potential for improved seaweed farming.

This research was conducted for four months from January to May, 2019 in Sasara coastal Waters, Kulisusu Bay, Buton Utara, SE Sulawesi, Indonesia. The aim of this research was to determine the best oblique incision length (OIL) to use in slide-slipped grafting using tissue cultured and local seedlings in terms of the growth and carrageenan yield of the seaweed Kappaphycus alvarezii. The treatments used in this study were A: 25-mm OIL, B: 30-mm OIL and C: 35-mm OIL. After 18 days, grafts using all OILs were successfully attached together. The new seedlings were then cultivated for 45 days. The results showed that the daily growth rate (DGR) did not differ significantly between treatments. The 25-mm OIL treatment had the highest DGR (3.98±0.97 %.day−1) followed by 35-mm OIL(3.90 ± 1.32%.day−1 and 30-mm OIL (3.69 ± 0.91 %.day−1). The ratio of fresh weight (FW) to dry weight (DW) also showed no significant differences among treatments. FW:DW ratios of treatments 25 mm OIL, 30 mm OIL, and 35 mm OIL were 8.13:1, 7.42:1, and 5.78:1, respectively. For carrageenan content, 30-mm OIL seedlings of showed higher content but it was not significantly different from the other treatments. Water quality parameters were: salinity 24-32 ppt; temperature 29-31 ° C; nitrate 0.336-0.432 mg.l−1; phosphate 0.195-0.542 ppm; and turbidity 3,124-9,331 NTU.

AbstrakTujuan dari penelitian ini adalah untuk mengetahui pengaruh waktu perendaman terhadap kualitas fisik, kimia dan sensorik udang Litopenaeus vannamei. Sampel dibagi menjadi tiga kelompok berdasarkan supplier udang yang berbeda, kemudian dilakukan empat perlakuan waktu perendaman, sehingga penelitian ini menggunakan rancangan acak kelompok (RAK).Peningkatan berat badan, kadar air, TVB dan, nilai TMA, serta kualitas sensorik diukur selama proses perendaman. Kandungan proksimat diukur sebagai data pendukung. Hasil penelitian menunjukkan bahwa level TVB dan TMA cenderung meningkat sepanjang waktu perendaman yaitu 5,56; 7.10; 9,60; dan 14,96 (mgN/100) untuk TVB sementara 2,8; 3.1; 3.7; dan 6,8 mgN / 100 untuk TMA. Persentase bobot tubuh cenderung meningkat terutama selama dua jam pertama, yaitu 9,7; 3.3; 0,7% untuk setelah 2, 4, dan 6 jam. Kadar air berturut-turut 79,09% (T0); 85,29% (T1); 74,64% (T2) dan 81,31% (T3). Kadar abu 25,96% (T0) dan 21,87% (T3), protein 58,87% (T0) dan 54,23% (T3), lemak 17,13% (T0) dan 21,90% (T3). Kata kunci: Bobot tubuh, Litopenaeus vannamei, ,TVB, TMA, waktu perendaman AbstractThis aim of this research was to determine the effect of soaking time on physical,  chemical and sensory quality of Litopenaeus vannamei. The samples collected were devided into three groups based on different farmer  as block then were treated by four defferent soaking time treatment, therefore the experiment was arrange under Randomized Block Design. Body weight increation, water content, TVB and, TMA value, also sensory quality were monitored during time soaking processing. Proksimate content was measured as supporting data. The result showed that both TVB and TMA level tend to increased along the extend of the time soaking which were 5.56; 7.10; 9.60; and 14.96 (mgN/100) for TVB while 2.8; 3.1; 3.7; and 6.8 mgN/100 for TMA. The body weight percentatively tend to increased especially during the firs two hours, which were 9.7; 3.3; 0.7% for after 2, 4, and 6 hour. Moisture content consecutively were 79.09% (T0); 85.29% (T1); 74.64% (T2) and 81.31% (T3).  Ash content were 25.96% (T0) and 21.87% (T3), protein were 58.87% (T0) and 54.23% (T3), Fat were 17.13% (T0) and 21.90% (T3). Key words: Body weight, Litopenaeus vannamei, TVB, TMA, soaking time
