Background: Amaryllidaceae plants are known to be a great source of alkaloids, which are considered an extensive group of compounds encompassing a wide range of biological activities. The remarkable cytotoxic activities observed in most of the Amaryllidaceae alkaloids derivatives have prompt the chemical and biological investigations in unexplored species from Brazil. Objective: To evaluate the cytotoxic and genotoxic properties of alkaloids of Griffinia gardneriana and Habranthus itaobinus bulbs and study the role of caspase-3 as a molecular apoptosis mediator. Methods: Methanolic crude extracts of Griffinia gardneriana and Habranthus itaobinus bulbs were submitted to acid-base extraction to obtain alkaloid-enriched fractions. The obtained fractions were fractionated using chromatographic techniques leading to isolation and identification of some alkaloids accomplished via HPLC and 1H-NMR, respectively. Molecular docking studies assessed the amount of free binding energy between the isolated alkaloids with the caspase-3 protein and also calculated the theoretical value of Ki. Studies have also been developed to evaluate in vitro cytotoxicity and genotoxicity in such alkaloids and apoptosis activation via the caspase pathway using both tumor and normal cell lines. Results: Seven alkaloids were isolated and identified. Among these, 11-hydroxyvittatine and 2-α-7- dimethoxyhomolycorine were not cytotoxic, whereas tazettine, trisphaeridine, and sanguinine only showed activity against the fibroblast lineage. Lycorine and pretazettine were 10 to 30 folds more cytotoxic than the other alkaloids, including cancerous lines, and were genotoxic and capable of promoting apoptosis via the caspase-3 pathway. This result supports data obtained in docking studies wherein these two compounds exhibited the highest free energy values. Conclusion: The cytotoxicity assay revealed that, among the seven alkaloids isolated, only lycorine and pretazettine were active against different cell lines, exhibiting concentration- and time-dependent cytotoxic actions alongside genotoxic action and the ability to induce apoptosis by caspase-3, a result consistent with those obtained in docking studies.

Species delimitation remains a central problem in systematic, taxonomic and evolutionary studies. However, the precise delimitation of species depends on the criteria used to identify lineages and the specific species concept that is applied. Recently, multidisciplinary studies using different data sources have significantly improved the delimitation of species within complex taxonomic groups, leading to an integrative taxonomy. To investigate the species delimitation within the Atlantic clade of Epidendrum (subg. Amphyglottium), four different species criteria were examined (phenetic differentiation, mono–phyly, diagnosability, absence of genetic intermediates). Morphometrics, plastid DNA sequences and nuclear microsatellite markers were used to explore the agreement between patterns recovered and species criteria tested. The conflicts among species criteria are discussed in light of pollination ecology, patterns of gene flow, reproductive isolation mechanisms and selective pressures currently acting in deceptive orchid species. Four currently recognized species from the Atlantic clade could be delimitated, including one newly described species, Epidendrum flammeus. Three out of five species satisfied the monophyly criterion, and few diagnostic flower characters were found among species. In contrast, nuclear microsatellite data correctly assigned individuals to their respective species, even in the presence of weak reproductive isolation and extensive hybridization events reported in the literature. One important implication of this finding is that phylogenetic studies in Epidendrum spp. should make use of single– or low–copy nuclear loci instead of plastid markers, which may be true for other plant groups. The results also indicate that the generalized pollination syndrome found among species of the Atlantic clade, the different levels of gene flow observed between nuclear and plastid markers, and hybridization events are commonly observed as the main evolutionary forces within this orchid group. Finally, we discuss evolutionary processes and taxonomic limits among these species, and we highlight the need to increase the inter–disciplinary approach to investigate species limits in complex plant groups.

Pradosia restingae Terra‐Araujo, a new species of Sapotaceae (Chrysophylloideae) from the Brazilian Atlantic forest is described and illustrated. It is only known from the southern coast of the State Rio Grande do Norte and is likely restricted to the coastal sand dune ecosystem, locally known as the restinga forest, from which the epithet is derived. The species is naturally common, but due the low number of known subpopulations we assign P. restingae a preliminary conservation status of ‘Endangered’ (EN).

Abstract—A new species of Dichorisandra from the Brazilian Atlantic Forest is herein described, illustrated, and compared to morphologically similar species. Dichorisandra striatula is characterized by leaf blades with small and discontinuous white stripes on the adaxial surface, hirsutulous indumentum on both surfaces, obovate to broadly obovate petals, and ellipsoid fruits. Information on phenology, habitat, conservation status, and a geographical distribution map are also provided.

The Dichorisandra acaulis group has its circumscription broadened to include two additional species, viz. D. oxypetala and the herein newly described D. forzzae. Dichorisandra oxypetala has previously not been included in this morphological group due to its deviant vegetative morphology. However, its reproductive morphology is congruent with the D. acaulis group and, thus, it is here included as a member of the later. Dichorisandra forzzae is here described to accommodate the specimens with anthers dehiscent through one apical pore, erroneously identified as D. acaulis in the state of Espírito Santo, Brazil. We provide illustrations, taxonomic comments and an updated identification key for the species of the D. acaulis morphological group. Furthermore, a table of comparison among the morphologically similar species of the Dichorisandra acaulis group is presented.
