Limitations to large-scale propagation of purple pitahaya (Hylocereus costaricensis [F.A.C. Weber] Britton & Rose), a potential source of betalains for the food industry, can be overcome through utilization of in vitro culture technologies. In this work, successful in vitro propagation from areoles of adult purple pitahaya plants is reported. Factors affecting culture initiation, bud sprouting and growth, shoot multiplication, rooting, and acclimatization were studied. Best results for culture initiation were obtained from the central region of new joints by disinfection of large (5-7 cm in length) explants that were subsequently divided. Explants were sequentially treated with Extran® alkaline detergent for 10 min, followed by immersion in 70 % (v/v) ethanol for 15-30 s, a mixture of the fungicide Benomyl and the bactericide Agrymicin (2 gl −1 each) for 30 min, and disinfection in sodium hypochlorite (1.0 %w/v) for 15 min. Culture of sectioned individual areoles, without removing thorns, on Murashige and Skoog medium supplemented with 15 or 30 μM N 6 -benzylaminopurine (BAP) for 3 mo with monthly subcultures, followed by transfer to the same medium with reduced BAP (0-2 μM), induced bud sprouting in over 80 % of explants, adequate growth of the shoots, with production of lateral shoots, and spontaneous rooting within 160 d. These plants were successfully acclimatized in vermiculite and peat moss (1:1), or perlite and peat moss (2:1) in the greenhouse, with over 90 % survival rate. One hundred percent of the in vitroderived plants were successfully transferred to the field. Furthermore, these plants showed higher survival rates, larger height and increase in stem diameter than equivalent plants from the same genotype, derived from stem segments (the common clonal propagation system utilized for this species) that were simultaneously planted.
Existen dos clases de marcadores genéticos: los morfológicos y los moleculares (Tanksley 1983). En primer orden, la caracterización e identificación tradicional de variedades se ha basado en el empleo de caracteres morfológicos y/o agronómicos (Rallo et al. 2002). No obstante, el uso de marcadores morfológicos en las plantas tiene muchas limitantes, pues su expresión puede estar sujeta a factores ambientales o fenológicos; por ejemplo la presencia o ausencia de espinas en los cítricos. Con frecuencia estos marcadores solo es posible evaluarlos a nivel de toda la planta y cuando esta llega a su estado adulto. Para la gran mayoría de frutales tropicales esto significa una espera, no deseable, de varios años. Además, pueden ocurrir cambios epigenéticos que limitan el número de marcadores que pueden ser evaluados sin equivocación en la población segregante (Powell 1992; Phillips et al. 1995). Por otro lado, gracias a los avances en la biología molecular se han desarrollado métodos de identificación y caracterización basados en el uso de marcadores moleculares que superan, en la gran mayoría de los casos, las limitaciones de los métodos tradicionales.
Introduction: Rice is a staple food in Costa Rica representing almost 24 % of the total calorie intake in the diet. Consumers have become more quality conscious about the rice consumed, so producers need to focus on the quality of rice along with production. Objective: The aim of this study was to investigate the physicochemical profile and consumer preference of five Costa Rican rice varieties: Palmar 18, Basmatico, Chin Chin, INTA-Miravalles, and CR- 5272. Materials and methods: Proximate analysis, amylose, and mineral content were performed in the raw grain. Two different sessions were performed (n=120 each) to evaluate consumer preference measurements using the John Brown R-Index (RJB) index. The first sensory panel selected the best cooking preparation and the second, the preferred rice variety. Instrumental color measurements (L*, C*, hº, DE) were applied to the cooked rice varieties (76-96ºC for 38 min). Results: Regarding the physicochemical characterization, INTAMiravalles showed the highest protein content (8.2%), while Basmatico and Chin Chin, the lowest. Four varieties presented high amylose content (AC) and INTA-Miravalles reported an intermediate AC. The mineral content varied among varieties. INTA-Miravalles showed the highest N, followed by Palmar 18. The cooking preparation from the regulatory agency was preferred by consumers. The consumers’ preference of the varieties ranked from high to low: Basmatico>Chin Chin=CR5772>Palmar 18=INTA-Miravalles. Conclusions: This study suggests that consumer preference for cooked rice was influenced by physicochemical characteristics such as protein, AC, mineral content, and aromatic attributes. Consumer acceptance is an important factor to consider when new varieties are developed. The water-to-rice ratio during cooking played an important role in cooked rice sensory quality.
Introducción. El arroz en granza una vez cosechado, se almacena en silos con aireación para control de temperatura, esto se conoce como añejado del arroz. Durante este proceso, ocurren cambios que influyen en las características físico-químicas y fisiológicas del grano, los cuales afectan su calidad culinaria. Objetivo. Determinar el efecto del tiempo de añejado sobre las características físico-químicas y sensoriales de dos variedades de arroz. Materiales y métodos. El estudio se realizó en los laboratorios de la Facultad de Ciencias Agroalimentarias de la Universidad de Costa Rica desde setiembre 2018 a marzo 2019. Se estudiaron dos variedades de arroz, que fueron cultivadas en el cantón de Osa, Puntarenas. La primera cv. Lazarroz FL, del tipo índica, y la segunda cv. UCR-168-10, tipo aromática, almacenadas durante 4,5 meses. Durante este periodo, se determinó el contenido de almidón total, almidón resistente, amilosa, humedad, proteína y, con un panel sensorial entrenado, se evaluaron los atributos sensoriales. Resultados. Para las dos variedades estudiadas, la humedad, el almidón total y el resistente, no presentaron diferencias significativas durante los 4,5 meses de almacenamiento. En la amilosa se observaron variaciones sin tendencia clara. La proteína mostró una reducción durante el añejado hasta el mes 2,5. Según las pruebas sensoriales, atributos como sabor integral y pegajosidad, no presentan diferencias significativas a lo largo del estudio, mientras que los parámetros restantes sí, ya sea en el tiempo o entre variedades. La soltura aparente incrementó a lo largo del tiempo para las dos variedades, pero se dejó de percibir diferencias importantes a partir de los 2,5 meses de almacenamiento. Para las dos variedades el color se tornó más blanco. Conclusión. El almacenamiento de arroz por 2,5 meses permitió obtener los cambios en el contenido de proteína y en los atributos sensoriales deseados para las variedades de arroz evaluadas.
