Camellia sinensis (green tea) is largely consumed, mainly in Asia. It possesses several biological effects such as antioxidant and anti-inflammatory properties. The objectives were to investigate the neuroprotective actions of the standardized extract (CS), epicatechin (EC) and epigallocatechin gallate (EGCG), on a model of Parkinson's disease. Male Wistar rats were divided into SO (sham-operated controls), untreated 6-OHDA-lesioned and 6-OHDA-lesioned treated for 2 weeks with CS (25, 50, or 100 mg/kg), EC (10 mg/kg), or EGCG (10 mg/kg) groups. One hour after the last administration, animals were submitted to behavioral tests and euthanized and their striata and hippocampi were dissected for neurochemical (DA, DOPAC, and HVA) and antioxidant activity determinations, as well as immunohistochemistry evaluations (TH, COX-2, and iNOS). The results showed that CS and catechins reverted behavioral changes, indicating neuroprotection manifested as decreased rotational behavior, increased locomotor activity, antidepressive effects, and improvement of cognitive dysfunction, as compared to the untreated 6-OHDA-lesioned group. Besides, CS, EP, and EGCG reversed the striatal oxidative stress and immunohistochemistry alterations. These results show that the neuroprotective effects of CS and its catechins are probably and in great part due to its powerful antioxidant and anti-inflammatory properties, pointing out their potential for the prevention and treatment of PD.

RESUMO:Justicia pectoralis Jacq., Acanthaceae, é uma erva conhecida popularmente no Nordeste como chambá e, utilizada tradicionalmente no tratamento de doenças do trato respiratório, como a asma, tosse e bronquite. Essa espécie encontra-se na Relação Nacional de Plantas Medicinais de Interesse para o SUS. O objetivo do presente estudo foi elaborar protocolo para a preparação da droga vegetal a partir de J. pectoralis e realizar a sua caracterização visando seu emprego como matéria-prima farmacêutica. A parte aérea de J. pectoralis, após secagem em estufa com circulação e renovação de ar (35 °C) durante diferentes períodos de tempo (1-5 dias) mostrou a partir de 24 h de secagem um teor de umidade abaixo do valor máximo permitido para drogas vegetais. O pó da droga vegetal foi classificado como pó moderadamente grosso e, caracterizado quanto aos teores de cinzas totais, extrativos solúveis em água e etanol. Análise do extrato hidroalcoólico (etanol 20%) de J. pectoralis por Cromatografia Líquida de Alta Eficiência (CLAE-DAD) determinou um teor de cumarina e umbeliferona de 16,2 e 0,81 mg/g da droga vegetal, respectivamente. As condições de preparação da droga vegetal e os parâmetros de controle de qualidade determinados para J. pectoralis no presente estudo são de interesse no desenvolvimento de fitoterápicos que empreguem esse matéria-prima ativa.Unitermos: Justicia pectoralis, cumarina, umbeliferona, CLAE-DAD, controle de qualidade, matéria-prima vegetal.ABSTRACT: Justicia pectoralis Jacq., Acanthaceae, is a herb popularly known in Brazilian northeast as "chambá" and used in folk medicine for the treatment of respiratory tract conditions such as asthma, cough and bronchitis. This species is included in the National Register of Plants of Interest to the National Health System. The aim of the present study was to develop a protocol for the preparation of the plant drug from J. pectoralis and to characterise the plant drug for its use as a pharmaceutical raw material. The aerial parts of J. pectoralis, after drying chamber with forced air circulation (35 °C) for different periods of time (1-5 days), presented after one day a moisture content below the maximum allowed for plant drugs. The powder of the plant drug was classified as moderately coarse, and the total ashes content and the water-or ethanolsoluble extractives were determined. Analysis of hydroalcoholic (ethanol 20%) extract of J. pectoralis by high performance liquid chromatography-photo diode array (HPLC-PDA) determined the content of coumarin and umbelliferone (16.2 and 0.81 mg/g plant drug, respectively). Background Schizophrenia is a chronic mental disorder, characterized by positive, negative and cognitive symptoms. In general, several plants have shown activity in diseases related to the central nervous system (e.g., Erythrina velutina (EEEV), also known as “mulungu”). For this reason, we aimed to investigate the effects of standardized ethanol extract obtained from the stem bark of EEEV on the schizophrenia-like behaviors induced by ketamine (KET) administration. Methods Swiss mice were treated with KET (20 mg/kg, i.p.) or saline for 14 days. In addition, from 8th to 14th days, saline, EEEV (200 or 400 mg/kg, p.o.) or olanzapine (OLAN 2 mg/kg, p.o.) were associated to the protocol. On the 14th day of treatment, schizophrenia-like symptoms were evaluated by the prepulse inhibition of the startle reflex (PPI), locomotor activity evaluated by the open field test (OFT), spatial recognition memory evaluated by the Y-maze task and social interaction test (SIT). Results KET has caused deficits in PPI, and it has also has caused hyperlocomotion in OFT and deficits in SIT as compared to control. EEEV in both doses used, reversed behavioral changes induced by KET, likewise results obtained with the administration of OLAN. Conclusions Taken together, the results demonstrate that the standard extract of EEEV was able to revert schizophrenia-like symptoms, due to the administration in repeated doses of ketamine. Thus, our findings lead to a new perspective for the use of EEEV an interesting alternative for drug discovery in schizophrenia.
Objetivo: Avaliar o monitoramento sérico de vancomicina em pacientes neonatos em uma maternidade de referência no estado do Ceará. Método: Trata-se de um estudo transversal e quantitativo, com abordagem descritiva, sendo os dados coletados referentes ao período de janeiro a julho de 2021. Resultados: De um total de 432 recém-nascidos, 13,20 % (57) neonatos usaram vancomicina como antibioticoterapia. Da amostra 77,2% (44) eram prematuros com idade gestacional média de 30 semanas. Considerando que os níveis séricos de vancomicina estão entre 5-15 mcg/mL, evidenciou-se ser atingido em 59,7% (34) dos bebês já na primeira coleta, em 73,7% (42) na segunda e 69% (20) na última. Ajustou-se em 40,4% dos neonatos, os intervalos posológicos após a primeira vancocinemia, 26,4% após a segunda e 31% na última. Identificaram-se nas hemoculturas positivas o Staphylococcus epidermides como o microrganismo mais prevalente nas unidades neonatais com 66,7%. Dentre as intervenções farmacêuticas realizadas, destaca-se a solicitação de exame de vancocinemia, o aumento e a redução do intervalo entre as doses. Conclusão: O estudo demonstrou que a grande maioria dos neonatos se encontrava com níveis séricos de vancomicina na dosagem ideal. No entanto, a atuação do farmacêutico clínico na equipe multiprofissional passa a ser fundamental, pois o monitoramento e o ajuste posológico da antibioticoterapia são de extrema importância para a segurança do paciente.
RESUMO: Justicia pectoralis (Acanthaceae) é uma erva arbustiva conhecida como chambá, empregada popularmente no tratamento de afecções respiratórias, está presente nas Farmácias Vivas e na Renisus. Estudos farmacológicos demonstram atividades antiinflamatória, broncodilatadora e antiasmática do chambá, estando estas relacionadas à presença de cumarina (CM) e umbeliferona (UMB) na planta. Tais substâncias são, assim, consideradas marcadores químicos para espécie. Dessa forma, o presente trabalho tem por objetivo otimizar o processo de produção do extrato seco de J. pectoralis por spray drying a partir de seu extrato líquido padronizado. Inicialmente, foi avaliada a composição do excipiente farmacêutico a ser empregado como adjuvante no processo de secagem. Após sua escolha, as condições do aparelho para secagem do extrato líquido padronizado foram trabalhadas através de planejamento fatorial 2 3 para obtenção de um produto com características físico-químicas de interesse. O extrato seco selecionado mostrou um teor de CM e UMB de 19,79 ± 0,079 (0,40) e 1,69 ± 0,002 (0,14), respectivamente, umidade de 6,83%. Certamente, o extrato seco padronizado desenvolvido poderá ser empregado em formulações farmacêuticas com uso em potencial para o tratamento da asma leve a moderada.
