Aim: To investigate the short-term safety and pharmacokinetic behavior of a new intraocular lens containing a dexamethasone drug delivery system (IOL-DDS) in rabbit eyes. Methods: A modified polymethylmethacrylate IOL containing a biodegradable dexamethasone DDS was implanted into the posterior chamber of the right eyes of 9 New Zealand white rabbits. Serial slitlamp and indirect ophthalmoscopic examinations (including grading of intraocular inflammation) were performed. After 3, 6 and 9 days, the rabbits were euthanized and the globes were removed for histological examination and for determination of dexamethasone levels in the aqueous humor and in the vitreous. Analysis of dexamethasone concentrations was performed by ELISA. Results: Therapeutic concentrations of dexamethasone were detectable in the aqueous and vitreous of the study eyes throughout the 9-day period in all tested animals. The mean aqueous dexamethasone concentration (ng/ml, ±SD) was 1,015.42 (±43.05), 970.11 (±32.47) and 757.58 (±30.19) and the mean vitreous concentration (ng/ml, ±SD) was 399.82 (±38.05), 287.38 (±34.47) and 268.15 (±32.00) at 3, 6 and 9 days after the surgical procedure, respectively. No corneal or retinal histological changes were observed during the study period. Conclusion: The IOL-DDS is effective in delivering therapeutic concentrations of dexamethasone to the aqueous and vitreous, without acute damage to the cornea and retina. Further controlled studies in the same animal model are under way to determine the potential value of this lens in the prevention and treatment of inflammation following cataract surgery.

Microscopicamente, o colágeno presente estava envolvido por tecido conjuntivo frouxo, foi mais intenso na porção intraarticular e, ao longo do tempo, apresentou-se em feixes paralelos, com fibras onduladas e organizando-se. As fibras elásticas estavam presentes em pequena quantidade, esparsas e ao redor do colágeno, predominando nas extremidades do enxerto. SUMMARYThe fascia lata was used for the intracapsular reconstruction of experimentally ruptured cranial cruciate ligament in dogs. In gross appearence, 90 days after surgery, the autografts were thick in intracapsular portion, presented bigger diameter than the normal ligament, and showed tendon aspect. Microscopicaly, collagen fibers were involved by slack conjunctive fibers, they were more intense in the intracapsular portion, and, later in time, presented an arragement of longitudinally oriented collagen fiber bundles. The elastic fibers were present in small amount, sparse and around the collagen fibers and prevailed in graft extremities.
Foram utilizados 36 coelhos da raça Nova Zelândia Branca, machos, com peso médio de 3,5kg e idade entre cinco e seis meses para avaliação da coleta, do preparo e da técnica de aplicação do auto-enxerto percutâneo de medula óssea (MO). Em 20 coelhos anestesiados, uma agulha para punção de MO foi inserida na crista ilíaca, sendo a medula aspirada com auxílio de seringa de 20m<img src="http:/img/fbpe/cr/v31n6/a16img01.gif"> heparinizada. As 20 amostras obtidas foram preparadas de duas maneiras, utilizando-se 10 amostras em cada procedimento: 1) medula integral: após a coleta de 1m<img src="http:/img/fbpe/cr/v31n6/a16img01.gif">, a amostra foi homogeneizada e, em casos de presença de coágulo, ela foi descartada; 2) medula centrifugada: após a coleta de 2m<img src="http:/img/fbpe/cr/v31n6/a16img01.gif"> a medula foi centrifugada por 10 minutos, sendo o sobrenadante removido e 1m<img src="http:/img/fbpe/cr/v31n6/a16img01.gif"> do sedimento homogeneizado. Em ambos os procedimentos, as amostras foram submetidas à contagem de células nucleadas. Nos 16 coelhos restantes, para permitir a avaliação da técnica de aplicação da MO, foi criada, cirurgicamente, uma falha óssea em ambos os rádios, pela remoção de um segmento osteoperiosteal com 1,0cm de comprimento, localizado a três centímetros da articulação rádio-carpo-ulnar. Após cinco dias da realização das falhas ósseas, os 16 coelhos foram separados em dois grupos iguais (I e II), sendo novamente anestesiados e preparados para a enxertia. No grupo I, cada animal recebeu 1,0m<img src="http:/img/fbpe/cr/v31n6/a16img01.gif"> de medula integral, imediatamente após a coleta, em um dos rádios (tratamento) e 1,0m<img src="http:/img/fbpe/cr/v31n6/a16img01.gif"> de solução salina fisiológica no rádio contralateral (controle). No grupo II, cada animal recebeu 1,0m<img src="http:/img/fbpe/cr/v31n6/a16img01.gif"> de medula concentrada em um dos rádios (tratamento) e 1,0m<img src="http:/img/fbpe/cr/v31n6/a16img01.gif"> de solução salina fisiológica no rádio contralateral (controle). Em função da característica física da medula, a coleta das amostras constituiu-se em procedimento simples, envolvendo trauma mínimo. Foi necessária prévia heparinização da seringa de coleta, o que não impediu a osteogenicidade das células. Com relação ao preparo das amostras, observou-se ampla variação individual na contagem celular nos dois procedimentos. A concentração da medula por centrifugação não interferiu negativamente no seu potencial osteogênico. A aplicação por via percutânea mostrou-se um método simples, com mínimo trauma aos tecidos, não introduzindo tecido desvitalizado e, desta forma, reduzindo o risco de infecção e de interferência na reparação óssea. A avaliação radiográficados membros operados, realizadas no dia da enxertia e uma semana após, demonstrou a eficiência da aplicação da MO e sua atuação na formação de osso.
The objectives of this study were to demonstrate the lesions that affect athlete bulls and to correlate the disorders with weight, age, affected limb and region of the limb. The present study was accomplished using radiographic images of athletic rodeo bulls collected from the medical and surgical records of the large animal service at the veterinary hospital. Radiographic images were evaluated for 136 bulls that were taken care of at the Veterinary Hospital, ranging in age from 4 to 13 years, with an average weight of 800kg and proven prior physical activity through participation in rodeos. The chi-square or Fisher’s exact test was used to assess the association between the studied variables. It was observed that 71.6% of the bulls studied and suffering from lameness had radiographic lesions, predominantly in experienced animals. Enthesopathy in starter and experienced bulls, septic arthritis in starter bulls, and fractures and degenerative joint disease in experienced bulls were the most frequent radiographic lesions diagnosed. The region of limb where the majority of radiographic changes occurred was the digits. Risk factors and occurrence of diseases of the locomotor system in athletic bulls are similar to those in sport equines.
