El objetivo de este estudio fue evaluar in vitro la actividad fijadora de nitrógeno y solubilizadora de fosfato de bacterias endófitas de pasto colosoana (Bothriochloa pertusa). Se colectó la raíz, los tallos y las hojas del pasto en cuatro zonas agroecológicas del municipio de Corozal, Sucre, Colombia, durante el segundo semestre del año 2012. Cada tejido fue sometido a proceso de desinfección superficial. Se aislaron bacterias endófitas en medio de cultivo agar R2A, a partir de cada tejido se determinó la densidad poblacional (UFC/g de tejido), por conteo en superficie; la separación de morfotipos se realizó mediante la forma, color, tamaño y apariencia. La actividad fijadora de nitrógeno y solubilizadora de fosfato se evaluó in vitro sobre medios de cultivos específicos. Todos los resultados obtenidos fueron analizados utilizando el programa estadístico R. Hubo diferencias significativas para densidades poblacional de bacterias con respecto a tejido, con mayores valores en raíz (1,61 x 1010/g raíz), seguida de tallo (7,44 x 109/g tallo) y los valores más bajos se reportaron para hojas (5,42 x 109/g hoja). Se encontró significancia entre la densidad poblacional con relación a los factores de zona, fincas y tipo de tejido analizado. Los resultados de la identificación con kit API20E confirmaron la presencia de las bacterias endófitas Aeromonas salmonicida y Pasteurella pneumotropica con capacidad simultánea de solubilizar fosfatos y fijar nitrógeno.
El presente estudio tuvo como objetivo, evaluar in vitro la actividad antifúngica de compuesto tipo antibiótico producido por bacterias endófitas aisladas de variedades de ñame contra el crecimiento micelial del hongo C. gloesporioides. Las muestras plantas de ñame fueron recolectadas aleatoriamente en forma de zig-zag en los sitios de muestreo seleccionados en el departamento de Sucre. Las muestras recolectados fueron desinfectados superficialmente, posteriormente se llevó a cabo el aislamiento, conteo y separación de morfotipos de bacterias endófitas mediante técnica de dilución seriada sobre superficie del medio agar. A cada morfotipo aislado fue utilizado para evaluar in vitro la actividad inhibitoria de células de bacterias endófitas contra el crecimiento del hongo C. gloesporioides. Los morfotipos de bacterias con mayor actividad inhibitoria fueron seleccionados para la obtención de metabolitos tipo antibiótico en dos fuentes de carbono (Glucosa y manitol) en tres concentraciones (0.5; 1,0 y 1,5 %). El morfotipo con mayor actividad inhibitoria fue 3AT1, los resultados del ensayo in vitro mostró mayor actividad cuando el compuesto tipo antibiótico fue obtenido en el medio con una concentración de 1.5% de glucosa. Los resultados de la identificación con kit API20E confirman con un 99.9 % de identidad con la especie de bacteria B. cepacia, constituyéndose este compuesto como un potencial biológico contra el crecimiento micelal del hongo C. gloesporioides, causante de la enfermedad conocida como antracnosis del cultivo del ñame en el departamento de Sucre.
<p>El objetivo de este estudio fue evaluar <em>in vitro </em>la resistencia de bacterias endófitas a diferentes concentraciones de plomo. El muestreo se realizó en el primer semestre de 2013, durante el cual se colectaron muestras de tejidos de variedades comerciales de arroz en etapa de macollamiento cultivadas en el municipio de Montería, Córdoba, Colombia. Cada tejido fue sometido a proceso de desinfección superficial. Se aislaron bacterias endófitas en medio de cultivo agar R2A; a partir de cada tejido se determinó la densidad poblacional (UFC/g de tejido) por el método de conteo sobre la superficie de medio R2A; la separación de morfotipos se realizó mediante la forma, color, tamaño y apariencia. Un total de 168 morfotipos fueron aislados de raíz, macolla y hoja de diferentes variedades comerciales de arroz. La prueba de resistencia a plomo se realizó <em>in vitro</em>, para esto se prepararon suspensiones de bacterias en fase <em>log</em>, se inocularon en medio mínimo con cinco concentraciones de plomo en forma de Pb(NO3)2. El ensayo se llevó a incubación con agitación por 150 rpm, a 32 °C por cinco días. Cada hora después de iniciado el ensayo se realizó lectura de la turbidimetría a 600 nm. Las bacterias mostraron la capacidad de crecer a concentraciones de 100% de Pb en forma Pb(NO3)2. La identificación con el kit API20E confirmó la presencia de <em>Burkholderia cepacia </em>y <em>Pseudomonas putida</em>, las cuales mostraron resistencia a diferentes concentraciones de plomo.</p>
