Die Methode von Fresenius-Babo beruht auf von Dulfos und M i 11 on gemachten Beobachtungen, daß, um gewisse Metalle in organischen Substanzen zu bestimmen, keine vollständige Mineralisierung nötig sei. Es genügt meistens eine Destruierung des Eiweißes und eine starke Oxydation durch Chlor. Zu diesem Zwecke haben Fresenius-Babo die Behandlung der gut zerriebenen Substanz mit konzentrierter Salzsäure und Kaliumchlorat vorgeschlagen. KC10 3 + 6 HC1 = 6 Cl + KC1-f 3 H 2 0. An diese Methode *) knüpften sich im Laufe der Zeit eine Reihe von Beobachtungen an, die den Zweck hatten, sie"zu vereinfachen oder zu beschleunigen. So schlug G. Bruylants vor, den Zusatz von Kaliumchlorat in Tablettform vorzunehmen, da dadurch die Chlorentwieklung langsamer und gleichmäßiger vor sich geht und die Ausbeute eine viel bessere ist. 2) Um die Anhäufung von Gl-Salzen zu vermeiden, die durch den Kaliumchloratzusatz bewirkt werden, wurde von Jeserich 3) der Vorschlag gemacht, letzteres durch kleine Mengen Chlorsäure zu ersetzen.
