The need for alternatives to animal use in pyrogen testing has been driven by the Three Rs concept. This has resulted in the inclusion of the monocyte activation test (MAT) in the European Pharmacopoeia, 2010. However, some technical and regulatory obstacles must be overcome to ensure the effective implementation of the MAT by the industry, especially for the testing of biological products. The yellow fever (YF) vaccine (17DD-YFV) was chosen for evaluation in this study, in view of: a) the 2016–2018 outbreak of YF in Brazil; b) the increase in demand for 17DD-YFV doses; c) the complex production process with live attenuated virus; d) the presence of possible test interference factors, such as residual process components (e.g. ovalbumin); and e) the need for the investigation of other pyrogens that are not detectable by the methods prescribed in the YF vaccine monograph. The product-specific testing was carried out by using cryopreserved and fresh whole blood, and IL-6 and IL-1β levels were used as the marker readouts. After assessing the applicability of the MAT on a 1:10 dilution of 17DD-YFV, endotoxin and non-endotoxin pyrogens were quantified in spiked batches, by using the lipopolysaccharide and lipoteichoic acid standards, respectively. The quantitative analysis demonstrated the correlation between the MAT and the Limulus amoebocyte lysate (LAL) assays, with respect to the limits of endotoxin recovery in spiked batches and the detection of no pyrogenic contamination in commercial batches of 17DD-YFV. The data demonstrated the applicability of the MAT for 17DD-YFV pyrogen testing, and as an alternative method that can contribute to biological quality control studies.
Neste estudo comparamos os resultados de ensaios de potência da alfaepoetina (EPOhr) realizados com camundongos de diferentes colônias e linhagens (Swiss Webster, NIH, C57BL/6 e BALB/c) com aqueles de ensaios conduzidos com o híbrido B6D2F1, o único camundongo recomendado pela Farmacopeia Europeia (FE). Fêmeas de diferentes colônias e linhagens, pesando 16-18 gramas, receberam uma única dose de EPOhr por via subcutânea (30, 90 ou 270 UI/animal, 0,2 mL/camundongo). As potências biológicas de apresentações de 4.000 UI/mL da EPOhr foram avaliadas utilizando um material de referência de trabalho de alfaepoetina (3.773 UI/mL) anteriormente testado junto ao padrão de referência internacional BRP (European Pharmacopeia Biological Reference Preparation). Os resultados indicaram que camundongos das colônias e linhagens examinadas atingiram critérios estatísticos (FE) para um ensaio válido de potência da eritropoietina e, portanto, podem ser considerados como alternativas ao uso do híbrido B6D2F1. Os ensaios com camundongos BALB/c, entretanto, foram os que produziram resultados mais semelhantes aos obtidos com os híbridos B6D2F1, em relação à contagem média de reticulócitos em resposta a 30, 90 e 270 UI/camundongo, e aos coefi cientes angulares (inclinação) e lineares (intersecção) da curva dose-resposta (curvas paralelas praticamente superpostas).
A necessidade de alternativas ao uso de animais no teste de pirogênio foi guiada pelo principio dos 3R’s, culminando no desenvolvimento e aceitação pela Farmacopeia Europeia do método alternativo in vitro, o Teste de Ativação de Monócitos (Monocyte Activation Test- MAT, 2.6.30/ 2010). O MAT utiliza como matriz, fontes de monócitos humanos (sangue total, PBMC, linhagem), respeitando o conceito dos 3R’s e excluindo riscos inerentes à extrapolação inter-espécies. É um método promissor e eficiente, que detecta um amplo espectro de pirogênios e supri limitações dos testes atuais preconizados pelas farmacopeias, o teste in vitro Teste de Endotoxina Bacteriana e o Teste de Pirogênio em Coelhos. Apesar das vantagens abordadas, alguns obstáculos técnico-científicos e regulatórios devem ser transpostos para a implantação efetiva do MAT na rotina industrial, em especial de produtos biológicos.
