AbstractIntroduction: The search for new compounds with antihypoxic and cardioprotective effects among 3-oxypyridine derivatives is promising. Research objectives:To study the anti-hypoxic and cardioprotective effects of 3-oxypyridine derivatives. Materials and methods:The search for compounds with an antihypoxic effect was carried out on blood leukocytes of rats in in vitro. To simulate hypoxia, Oil for Tissue Culture (SAGE) was used, 500 µl of which was applied into wells over a growth medium in order to block gas exchange. The cardioprotective effect of 3-oxypyridine derivatives was studied in the model of coronary-occlusive myocardial infarction (30 minutes of ischemia, 90 minutes of reperfusion). The level of troponin I (Tn I) was determined as a biochemical marker of myocardial damage. Results and discussion:In the in vitro experiments, when culting white blood cells, the lead compound in the group of 3-oxypyridine derivatives was identified under code LKhT 21-16, which increases the number of viable cells in the presence of hypoxia, surpassing the reference drugs. When confirming the chemical structure of the lead compound, LHT 21-16, a high sensitivity of the NMR spectroscopy method was revealed.In studying the cardioprotective activity in the model of coronary-occlusive myocardial infarction compound LHT 21-16 exerted a marked cardioprotective effect when reducing the size of the necrotic zone and the level of biochemical marker Tn I. Conclusions:3-oxypiridine derivatives have antihypoxic and cardioprotective effects, which shows in a high number of surviving cells in the presence of hypoxia in the in vitro model, a reduced size of the necrotic zone and a reduced level of Tn I in the coronary-occlusive myocardial infarction.

В экспериментах на изолированном по Лангендорфу сердце крысы изучали кардиопро-тективное действие эксенатида (10 мкг/кг/сутки) («Баета ® », Eli Lilly and Company, США) и вилдаглиптина (0,2 мг/ кг/сутки) («Галвус ® », Novartis, Швейцария), на сократительную функцию изолированного сердца, подвергшегося предворительной доксорубициновой (20 мг/кг, внутрибрюшинно за 48 часов) патологии. Кардиопротективное дей-ствие оценивали по результатам функциональной пробы с высокочастотной стимуляцией (480 уд./мин.) в условиях гиперкальциевой (5 ммоль) перфузии.Результаты. Полученные результаты свидетельствуют о том, что инктериномиметики эксенатид (10 мкг/кг/сут-ки) и вилдаглиптин (0,2 мг/кг/сутки) проявляют кардиопротективный эффект на доксорубициновой модели пато-логии, что выражается в снижении коэффициента диастолической дисфункции (S tТТI ), соответственно до 5,3±0,1 ус.ед. и 6,5±0,2 ус.ед. по сравнению с группой контроля 8,3±0,1 ус.ед.Заключение. Высказывается предположение о пути реализации кардиопротективного эффекта инкретиноми-метиков путем усиления экспрессии гем-оксигеназы-1(HO-1). При этом предотвращается катализируемое гемом образование высокоактивных гидроксильных радикалов из перекиси водорода. Индукция гем-оксигеназы-1 сопро-вождается увеличением активности ферритина, что оказывает антиапоптотический эффект за счет того, что фер-ритин связывает избыток свободного железа в клетках, подвергшихся оксидативному стрессу. Material and methods.In the experiments at the isolated the Langendorff heart of the rat, the cardioprotective effect of exenatide (10 μg/kg/day) (Baeta®, Eli Lilly and Company, USA) and vildagliptin (0.2 mg/kg/day) (Galvus®, Novartis, Switzerland), on the contractile function of an isolated heart that underwent anterior doxorubicin (20 mg/kg, intraperitoneally for 48 hours) pathology was evaluated. Cardioprotective effect was assessed by the results of a functional test with highfrequency stimulation (480 bpm) under conditions of hypercalcium (5 mmol) perfusion.
