The conservation of threatened species poses many challenges but through cooperation and pooling of resources, individuals and organizations can work together to achieve better results. Here we describe our experience, working through a governmental and private alliance, studying one of the most threatened mammals in the Southern Cone. The huemul deer Hippocamelus bisulcus, one of two members of the Hippocamelus genus of South America, is endemic to Argentina and Chile and currently inhabits only a small fraction of its former range. Little is known about the huemul because it generally lives in remote areas with a harsh climate and rugged terrain. Using drive counts and fixed width transects over 5 consecutive years (2004–2008) we estimated density and abundance, and examined population changes and social structure, in three coastal huemul populations in the area of the Témpanos and Bernardo fjords of Bernardo O'Higgins National Park, Chile. Our results suggest that synergistic conservation actions, such as cattle removal and poaching control, can lead to the recovery of threatened huemul. The baseline information obtained from our surveys and the lessons learned through this governmental and private alliance will be useful for future monitoring of the huemul in the Patagonian fjords of Chile.

BackgroundChronic obstructive pulmonary disease (COPD) is a disorder associated to cigarette smoke and lung cancer (LC). Since epigenetic changes in oncogenes and tumor suppressor genes (TSGs) are clearly important in the development of LC. In this study, we hypothesize that tobacco smokers are susceptible for methylation in the promoter region of TSGs in airway epithelial cells when compared with non-smoker subjects. The purpose of this study was to investigate the usefulness of detection of genes promoter methylation in sputum specimens, as a complementary tool to identify LC biomarkers among smokers with early COPD.MethodsWe determined the amount of DNA in induced sputum from patients with COPD (n = 23), LC (n = 26), as well as in healthy subjects (CTR) (n = 33), using a commercial kit for DNA purification, followed by absorbance measurement at 260 nm. The frequency of CDKN2A, CDH1 and MGMT promoter methylation in the same groups was determined by methylation-specific polymerase chain reaction (MSP). The Fisher’s exact test was employed to compare frequency of results between different groups.ResultsDNA concentration was 7.4 and 5.8 times higher in LC and COPD compared to the (CTR) (p < 0.0001), respectively. Methylation status of CDKN2A and MGMT was significantly higher in COPD and LC patients compared with CTR group (p < 0.0001). The huemul ( Hippocamelus bisulcus ) is an endangered cervid endemic to southern Argentina and Chile. Here we report foot lesions in 24 huemul from Bernardo O’Higgins National Park, Chile, between 2005 and 2010. Affected deer displayed variably severe clinical signs, including lameness and soft tissue swelling of the limbs proximal to the hoof or in the interdigital space, ulceration of the swollen tissues, and some developed severe proliferative tissue changes that caused various types of abnormal wear, entrapment, and/or displacement of the hooves and/or dewclaws. Animals showed signs of intense pain and reduced mobility followed by loss of body condition and recumbency, which often preceded death. The disease affected both genders and all age categories. Morbidity and mortality reached 80% and 40%, respectively. Diagnostics were restricted to a limited number of cases from which samples were available. Histology revealed severe papillomatous epidermal hyperplasia and superficial dermatitis. Electron microscopy identified viral particles consistent with viruses in the Chordopoxvirinae subfamily. The presence of parapoxvirus DNA was confirmed by a pan-poxvirus PCR assay, showing high identity (98%) with bovine papular stomatitis virus and pseudocowpoxvirus. This is the first report of foot disease in huemul deer in Chile, putatively attributed to poxvirus. Given the high morbidity and mortality observed, this virus might pose a considerable conservation threat to huemul deer in Chilean Patagonia. Moreover, this report highlights a need for improved monitoring of huemul populations and synergistic, rapid response efforts to adequately address disease events that threaten the species.

This study examined landcover change during 1940-1988 in two contiguous landscapes of approximately 12,400 hectares in central Ohio, USA: an inertial till plain area and a more dynamic moraine area. Agriculture dominated both landscapes throughout, however, the cover of natural vegetation and urbansuburban development increased over time on the moraine while natural vegetation decreased and agriculture increased on the till plain. Markov process models for landcover change were constructed for three 14-1 7-year time intervals and for the entire forty-eight-year study period. Probabilities of sevreplacement for agricultural lands (0.91-0.97) were higher and those for forests (0.54-0.65) lower than those reported for other landscapes. Predictions of landcover percentages f o r j f t y years in the future were made using Markov process models derived from actual cover changes in 1940-1 957, 1971 -1 988, and 1940-1 988. All three models produced similar predictions for the inertial till plain area. In contrast, the three models' predictions difered considerably for the moraine area. Only the 1971 -1 988 transition matrix was sensitive to increases in urbanization and reforestation which began on the moraine in the 1970s. These results indicate that reliance on lengthy study intervals and net rates of change can obscure spatial and temporal patterns of landcover change that are caused by natural and socioeconomic factors operating on shorter time scales.

We evaluated the amounts and implications of changes in habitat connectivity on rural land‐scapes by modeling the colonization success and subsequent habitat colonization of a model edge organism within real landscapes. We first inventoried the changes in the fencerow and forest‐edge network of two contiguous Ohio (U.S.A.) landscapes, an agriculturally dominated till plain and a more diverse and dynamic moraine landscape, from 1940 to 1988. On the moraine the number of fencerows changed little from 1940 to 1971. The number increased by 86 during 1971–1988 as marginal farms were subdivided. The total length of fencerows on the moraine increased 2.5‐fold over 48 years. On the till plain the number and total length of fencerows remained relatively constant through the study period. The sum of fencerows and forest edges was used as a measure of total ecotonal edge. On the moraine total edge increased through the study period, whereas on the till plain it decreased. We selected two levels of landscape connectivity, low and high, to model animal habitat colonization success. As connectivity increased the earliest successful colonists preempted an increasingly large proportion of the total suitable habitat, and the probability of successful colonization by later‐arriving individuals decreased. The changes in connectivity that resulted from changes in both the fencerow network and the proportion of forested land have resulted in contiguous landscapes that present very different colonization potentials to organisms with long‐distance dispersal capability. Given the current uncertainty of the effects of corridors on species‐preservation efforts, we suggest that further modeling of this type prior to field testing will add useful insights, especially if conducted using specific species and landscape types.
