One of the causal of the low rubber production is the presence of plant diseases. Important diseases in rubber plants generally cause symptoms of leaf fall, due to Colletotrichum spp., Corynespora sp., and Oidium sp. Currently, there is an incidence of new rubber leaf fall disease with symptoms that are different from the diseases previously found in rubber plantations. This disease is widespread in Sumatra, Kalimantan, and Southeast Asia. There are many suspicions about the pathogens that cause the disease, but there is no precise diagnosis. Therefore, an accurate diagnosis is needed as a basis for determining an effective and efficient disease control strategy. This research was carried out to determine the causal agents of leaf fall disease with symptoms of round leaf spots that can cause leaf fall. Diagnosis was carried out by isolating the pathogen from several rubber clones, inoculating the pathogen to healthy rubber plants, identifying morphologically and molecularly, and re-isolating from inoculated plants. The results of Koch's postulates and morphological and molecular identification determined that the causal agents of leaf fall disease at rubber plants with round spots was the fungus Pestalotiopsis microspora.
Penyakit jamur akar putih (JAP) merupakan salah satu penyakit penting di perkebunan karet Indonesia karena dapat menyebabkan kematian tanaman dan kerugian ekonomi yang cukup tinggi. Salah satu usaha pengendalian penyakit JAP adalah pengobatan tanaman sakit dengan menggunakan biofungisida. Tujuan dari pengujian ini untuk mengetahui efektivitas biofungisida berbahan aktif beberapa mikroorganisme antagonis terhadap penyakit JAP pada skala laboratorium, rumah kaca, dan lapangan. Penelitian dilaksanakan di Balai Penelitian Sembawa mulai Juli 2012 sampai April 2013. Bahan utama yang digunakan pada penelitian ini adalah formulasi biofungisida yang mengandung cendawan antagonis Trichoderma viridae, T. harzianum, Paecilomyces lilacinus, dan bakteri antagonis Bacillus subtilis. Percobaan terdiri dari tiga kegiatan yaitu pengujian antagonisme formulasi biofungisida terhadap R. microporus di laboratorium dengan menggunakan metode uji ganda, studi efektivitas formulasi biofungisida terhadap penyakit JAP pada bibit karet dalam polibeg klon PB 260 di rumah kaca dengan menggunakan rancangan acak lengkap enam perlakuan dan tiga ulangan yang terdiri dari kombinasi biofungisida + pupuk hayati berbahan aktif mikoriza serta perlakuan fungisida kimia sebagai pembanding, dan studi efektivitas formulasi biofungisida terhadap penyakit JAP pada tanaman karet belum menghasilkan klon PB 260 di lapangan dengan menggunakan rancangan acak kelompok sembilan perlakuan dan tiga ulangan yang terdiri dari beberapa perlakuan biofungisida dan fungisida kimia pembanding. Hasil penelitian di laboratorium menunjukkan biofungisida mampu menekan perkembangan R. microporus dengan rata-rata penghambatan sebesar 57,62%. Pengujian di rumah kaca dengan perlakuan kombinasi biofungisida 100 g dan pupuk hayati 200 g cukup efektif menurunkan intensitas serangan JAP sebesar 5,56% dan mampu meningkatkan pertumbuhan tanaman yang terlihat dari pertumbuhan akar, tinggi tanaman, dan biomassa kering yang lebih baik dibandingkan dengan perlakuan lain. Pengujian biofungisida pada TBM menunjukkan penurunan intensitas serangan penyakit sebesar 18,33% s.d. 23,33% yang tidak berbeda nyata dengan fungisida kimia pembanding dan perlakuan biofungisida 20 g/pohon memiliki penurunan intensitas serangan penyakit paling tinggi dibandingkan perlakuan biofungisida lainnya. Dari ketiga pengujian menunjukan biofungisida tersebut efektif digunakan untuk mengendalikan penyakit JAP. Penyakit gugur daun Corynespora yang disebabkan oleh cendawan Corynespora cassiicola (C. cassiicola) merupakan salah satu penyakit daun penting yang dapat menyebabkan penurunan produksi lateks pada perkebunan karet. Salah satu tahapan penting untuk melepaskan klon karet baru adalah mengidentifikasi karakter sekunder seperti resistensi terhadap penyakit. Pengujian resistensi klon karet IRR seri 300 dilakukan di laboratorium dan rumah kaca dengan menggunakan Rancangan Acak Lengkap dua faktor yaitu faktor jenis (genotipe) klon (26 jenis klon) dan isolat C. cassiicola (3 isolat). Selain itu, pengamatan serangan penyakit secara langsung juga dilakukan pada tanaman belum menghasilkan di lapangan. Hasil pengujian menunjukkan semua isolat C. cassiicola berpengaruh nyata terhadap resistensi 26 klon IRR seri 300 baik di laboratorium maupun di rumah kaca. Hasil pengamatan pada tiga kegiatan menunjukkan bahwa 13 klon karet yaitu IRR 301, IRR 302, IRR 303, IRR 304, IRR 305, IRR 307, IRR 308, IRR 309, IRR 312, IRR 315, IRR 316, IRR 318, dan IRR 323 memiliki tingkat resistensi tinggi terhadap penyakit gugur daun Corynespora.
Penyakit gugur daun Colletotrichum merupakan salah satu penyakit penting yang dapat menyebabkan penurunan produksi lateks pada perkebunan karet. Pengendalian penyakit gugur daun Colletotrichum yang paling efektif adalah dengan penggunaan klon resisten. Pengujian resistensi klon karet IRR seri 300 dilakukan di laboratorium dan rumah kaca dengan menggunakan rancangan acak lengkap dua faktor yaitu jenis klon (26 jenis klon) dan isolat C. gloeosporioides (tiga isolat: CG-PR 303, CG-RRIM 600, dan CG-GT 1). Selain itu, pengamatan serangan penyakit secara langsung juga dilakukan pada tanaman belum menghasilkan di lapangan. Hasil pengujian menunjukkan bahwa semua isolat C. gloeosporioides memiliki pengaruh nyata terhadap tingkat ketahanan 22 klon IRR seri 300 baik di laboratorium maupun di rumah kaca. Isolat CG-PR-303 memiliki tingkat virulensi paling tinggi dibandingkan isolat lainnya. Berdasarkan hasil pengamatan pada tiga kegiatan dapat disimpulkan bahwa 13 klon karet yaitu IRR 300, IRR 301, IRR 302, IRR 307, IRR 308, IRR 309, IRR 310, IRR 311, IRR 313, IRR 315, IRR 316, IRR 318, dan IRR 321 memiliki tingkat resistensi yang tinggi terhadap penyakit gugur daun Colletotrichum.
Penyakit jamur akar putih (JAP) yang disebabkan oleh cendawan Rigidoporus microporus (Sw.) Overeem masih menjadi penyakit penting dan merugikan pada perkebunan karet Indonesia hingga saat ini. Upaya pengendalian JAP secara biologi terus dilakukan untuk melengkapi cara lain yang telah ada (kimia dan kultur teknis) agar diperoleh hasil yang lebih efisien dan efektif. Studi pendahuluan pemanfaatan bahan tanaman antagonis telah memberikan capaian yang prospektif. Diperolehnya ekstrak tanaman antagonis kunyit (C. domestica) yang efektif untuk pengendalian JAP. Penelitian ini dilakukan dengan tujuan untuk mengetahui efektifitas, persistensi, dan fitotoksisitas bahan nabati tersebut yang diekstrasi menggunakan berbagai bahan pelarut dan dibuat dalam berbagai bentuk formula. Hasil penelitian yang dilakukan di laboratorium dan rumah kaca Balai Penelitian Sembawa menunjukkan bahwa pada kondisi laboratorium ekstrak kunyit efektif menekan perkembangan cendawan R. microporus sebesar 43,25% s.d. 65,13% terhadap kontrol. Pada kondisi rumah kaca diperoleh hasil bahwa formula ekstrak kunyit 20 EC + pelarut n-hexane dapat menurunkan intensitas serangan penyakit JAP sebesar 20,80%. Semua jenis formula yang diuji tidak toksik terhadap tanaman karet dan memiliki persistensi yang baik sampai empat hari setelah aplikasi formula ke media tanam (tanah) dalam polibeg.
