KEYWORDSNoble metal nanoparticles were synthesized directly by pulsed laser ablation (Nd:YAG, λ=1064 nm) of gold and silver plates immersed in pure water. Concentrations of the nanoparticles were determined by atomic absorption spectroscopy measurement. Atomic force microscope and transmission electron microscope analysis were used to characterize the size and size distributions of the metals nanoparticles. The objective of this work is studying the effects of presence gold and silver nanoparticles on the levels of triiodothyronine hormone (T3) in saliva of patients with chronic renal failure their thyroid had disorder (hypothyroidism). Also the study characterized the binding between the antiT3antibody with its antigen i.e., T3 in saliva patients in the presence affixed size concentration of nanoparticles to improve and modify a competitive ELISA method. It is found that both gold and silver nanoparticles demonstrated activation effect on the binding between anti-T3 antibody and antigen (T3) in the saliva of patients and these effects increased with increasing the concentrations and size for both nanoparticles (gold was more activation effect than silver). Optimization of experimental conditions for the binding in the saliva were: 40 μL and 20 μL of saliva (T3) in presence both silver and gold nanoparticle, respectively, pH = 7.4 of 0.2 M phosphate buffer was selected as the detection solution, the temperature of incubation was 22 °C while the incubation time were 30 min, 90 min for silver and gold nanoparticles, respectively.
INTRODUCTIONSynthesis and evaluation of efficacious antimicrobial, due to limited number of effective drugs 1 and antioxidant agents to protect human body from free radicals 2 represent the main goal for the several research groups worldwide. Recently, considerable attention has been given to isoxazoline derivatives due to their varied pharmacological activities like antimicrobial 3 , anti-inflammatory 4 , anti-tubercular 5 , antidepressant 6 , antioxidant 7 , antitumor and DNA Methyltransferase 1 Inhibitors 8 . Therefore, our attempts to find novel antimicrobial and antioxidant agents focus on the isoxazoline derivative. A novel series of isoxazolines (3-10) were synthesized and characterized by spectral data. Isoxazoline derivatives (3-ABSTRACT A novel series of isoxazoline derivatives (3-10) was synthesized and characterized by using spectral analysis. The synthesized derivatives were in vitro screened against several bacterial species as well as Candida albicans and exhibit moderate to potent activity. All the synthesized products were screened qualitatively for their antioxidant property by using TLC technique and the percent DPPH radical scavenging activity of the potent derivatives (4-6,8) were evaluated. Docking study of isoxazoline derivatives 7 and 8 against L-Glutamine: D-fructose-6-phosphate amidotransferase (glucosamine-6-phosphate synthase), the target enzyme in antimicrobial chemotherapy, was evaluated to explore the interactions of the synthesized hits inside the amino acid residues of the enzyme active site. The docking outcomes strongly supported the in vitro assay of new derivatives against several microbial species.
In current research, the synthesis of gold nanoparticles was achieved via reducing of gold ions in aqueous solution with Garcinia mangostana (G. mangostana) peel extract. The optimum concentration of gold (Au) solution, concentration ratio of Au solution and extract, temperature, time and pH, the synthesized AuNPs (G. mangostana-gold nanoparticles) were studied by using UV-Vis, FT-IR, AAS, AFM, SEM and Zitasizer. The absorbance peak is noticed between 535-550 nm via UV-Vis spectroscopic method. The SEM, AFM analysis were proofed the particle as spherical in structure and their size between 15-100nm. Therefore, mechanism of AuNPs synthesis had been suggested. Also, the antibacterial activity was examined using different bacteria as well as free radical scavenging activity was tested using 1, 1-Diphenyl-2-picrylhydrazyl (DPPH). The AuNPs produced through biosynthesized method indicated a much elevated antioxidant activity as compared to peel extract of G. mangostana. Toxicity of the NPs and extract were tested via giving orally dose 50 mg/b.w. to mice. Diagnosis of the data (pathological changes) indicated that the AuNPs was non-toxic. The G. mangostana peel extract and AuNPs synthesized by this extract were converted to a cream and used as a wound healing cream. As a results, the AuNPs exhibited important role in wound healing progression compared to control, which may be attributed to their anti-inflammatory, antibacterial and antioxidant activities. Therefore, this research confirms its important use of AuNPs and can be utilized as promising agents for in the development of new drugs.
A novel derivative of 3,5-disubstituted-2-isoxazolines (3-11) have been synthesized and characterized by spectral analysis. All the synthesized derivatives were screened qualitatively for their antioxidant property using TLC technique and the percent DPPH radical scavenging activity of the potent derivatives (5, 11) were evaluated. The 2-pyrazolines (3-11) were in vitro screened against different bacterial strains as well as candida albicans and found exhibiting moderate to potent activity. Docking study of 2-isoxazoline derivative 11 against glucosamine-6-phosphate synthase, the target enzyme for the antimicrobial agents, was explored to study the interactions of the discovered hit (11) within the binding pocket residues of the enzyme.
Hypothyroidism is a common disorder of the thyroid gland in which the gland cannot compete with the body requirement of the thyroid hormones, triiodothyronine (T3) and thyroxine (T4). Hypothyroidism is classified into overt (OH) and subclinical (SbH), in which the first comprises the cases that have elevated levels of thyroid- stimulating hormone (TSH) and reduced levels of T4 while the second comprises the cases with elevated TSH but normal T4 levels. The goal of the present study is to investigate the level of malondialdehyde (MDA) and some minerals (calcium, magnesium, and phosphorus) in patients with SbH and OH. For this purpose, 80 patients from Al-Yarmook Teaching Hospital were enrolled in the study half of them were diagnosed with OH and the other half were diagnosed with SbH. The study has been controlled with 40 healthy people of comparable age and all of the three groups were contained 50% males and females. The anthropometric examination of subjects has revealed that the body mass index (BMI) and waist to hip ratio (WHpR) were significantly (p<0.05) altered among control, OH, and SbH subjects.
