Fur das aus Colchicum a u t u m n a l e L. und einigen anderen verwandten Liliaceen isolierte Demecolcin (fruher Substanz F genannt) wurde als wahrscheinlichste Struktur die Formel XVII eines N-Methyl-desacetyl-colchicins abgeleitetl). Dieselbe Formel wurde auch von Kiselew und Mitarb. p ) 2, fur Colchamin vorgeschlagen, das wahrscheinlich mit Demecolcin identisch ist. Kiselew & Menschikown) haben diese Formel kiirzlich fiir Colchamin weitgehend sicherstellen konnen. Unabhangig davon hat UenoP) ein ahnliches Resultat beim Demecolcin erreicht. I n beiden Arbeiten wird gezeigt, dass Colchamin bzw. Demecolcin ein Abkommling des N-Methyl-desacetyl-colchicins ist; es fehlt lediglich ein strenger Beweis, dass Demecolcin auch in1 Ring C genau gleich gebaut ist wie Colchicin und nicht dcr Iso-Reihe angehort. Hier wird nun die Formel XVII fur Demecolcin durch eindeutige Teilsynthese bewiesen3), ferner werden einige neue Derivate des Colchicins und Demecolcins beschrieben. Als Stiitze fur die Formel XVII wurden zunachst folgende Umsetzungen ausgefuhrt : Allocolchiceina)b) wurde durch Erhitzen mit konz. HC1 in krist. Desacetyl-allocolchicein (I) ubergefuhrt, das einen amorphen Methylcster I1 (Desacetyl-allocolchicin) lieferte. Dieser gab bei erschopfender Methylierung mit Methyljodid das krist. N-Trimethyldesacetyl-allocolehicinium-jodid (111), das beim Erhitzen leicht Trimethylamin abspaltete. Dasselbe Salz liess sich durch analoge Methylierung aus Allodemecolcein (IV) bereiten. Um einen eindeutigen Strukturbeweis xu fuhren, versuchten wir zunachst Colchicin (XI) am Stickstoff zu methylieren. Bei erfolgreicher Methylierung hatte dabei das bekannte N-Acetyl-demecolcin (XV) entstehen mussen, falls Formel X V I I fiir Demecolcin richtig ist. Da Colchicin gegen heisses Alkali empfindlich ist, wurde eine moglichst schonende Methode versucht. I n Vorversuchen zeigte es sich, dass Acetanilid rnit Dimethylsulfat und K-tort.-Amylat bei 18O glatt in N-Methyl-acetanilid ubergeht. Colchicin blieb unter gleichen Bedingungen weitgehend unverandert. Auch langeres Kochen mit Rg,O und l) 35. Mitt., F. &antat$, R. Winkle? & T. 19Methyljodid fiihrte nicht zum Ziel. Wir hofften, dass N-Formyl-desacetylcolchicin (,,Substanz B") etwas reaktionsfafiiger sein wurde. Die analogen Versuche fuhrtcn aber auch hier nicht zum Ziel; das kiirzlich beschriebene N-Formyl-demecolcinl) liess sich dabei nicht isolieren.Da freie NH,-Gruppen leichter methylierbar sind als acetylierte, beschlossen wir, die Methylierung des Desacetyl-colchicins ( X I I ) oder seiner Isoverbindung XI11 zu versuchen. Um die Resultate solcher Versuche auf eine sichere Basis zu stellen, haben wir zunLchst die friihere) beschriebene Methylierung des Demecolceins (XXI) rnit Diazomethan wiederholt. Ausser Demecolcin (XVII) gelang es nach eini...
