In Saccharomyces cerevisiae, one of two cytosolic lysine-tRNAs is partially imported into mitochondria. We demonstrate that three components of the ubiquitin/26S proteasome system (UPS), Rpn13p, Rpn8p and Doa1p interact with the imported tRNA and with the essential factor of its mitochondrial targeting, pre-Msk1p. Genetic and biochemical assays demonstrate that UPS plays a dual regulatory role, since the overall inhibition of cellular proteasome activity reduces tRNA import, while specific depletion of Rpn13p or Doa1p increases it. This result suggests a functional link between UPS and tRNA mitochondrial import in yeast and indicates on the existence of negative and positive import regulators.
Synthesis of 6(E)-and 6(Z)-(3-Ethoxycarbonylpropyl)oximes of 16α,17α-Cyclohexanopregn-4-ene-3,6,20-trione and Study of Their Interaction with Proteins of the Rat Uterine Cytosol and Blood Serum.-The suitability of steroids (VII) and (VIII) as ligands in affinity chromatography for the isolation of proteins is investigated by studying their ability to bind to proteins from the rat uterine cytosol and blood serum using competitive analysis. The more efficient binding is shown by (E)-isomer (VII), due to the fact that in (VII), the conjugated ketone in ring A is more spatially accessible for the reaction with the protein.-(LEVINA, I. Aim. The paper presents the experience of treatment of echinococcal cysts in children, which reflects most of the existing methods of treatment.Materials and methods. From 2010 to 2021, 81 patients with hepatic echinococcosis aged from 3 to 17 years (average 14.3 ± 1.3) were treated at the bases of the Department of Pediatric Surgeons of the Pirogov Russian State Medical University. All patients were examined according to the standard procedure, including: general clinical examination, examination of liver function, ultrasonography, chest X-ray, CT scan, blood test for IgG to echinococcus antigens. All children were divided into 2 groups according to the type of the surgery: in the group 1, 27 (33.3%) children underwent laparoscopic surgery; in the group 2, 54 (66.7%) children – laparotomy.Results. The treatment duration accounted 12.5 ± 3.1 days in group 1 and 9.2 ± 1.3 days in group 2 (p < 0.0067). In the group 1 the drainage duration was higher (6.8 ± 0.6) than in the group 2 (4.1 ± 0.3) (p < 0.03). Complications occurred in 7 patients, relapse of cysts – in 2 patients.Conclusion. Children with echinococcal cysts in Central Russia occur with a constant frequency and require to apply modern approaches in diagnosis and treatment. The classification and treatment protocol of echinococcosis, adopted by WHO, are easy-to-use and proves its effectiveness. There is no significant difference in germicidal agents’ effect on echinococcus and the effect on the development of relapse. The method of selection in the treatment of solitary and superficial cysts is laparoscopic interference with the mandatory application of a PAIR and a high-power aspirator. An important stage of the interference is the treatment of the fibrous capsule of the cyst, if it is impossible to completely remove it, with an argon-plasma coagulator.
Aim. To study current trends in the incidence and mortality of prostate cancer in Kemerovo Region.Materials and Methods. We conducted a descriptive retrospective study by investigating cancer registry of Regional Clinical Oncology Dispensary collected in 2010-2018.Results. During the study period, 4965 cases of prostate cancer have been documented. The average standardised incidence rate was 50,080/0000 (95% CI = 46.01; 54.43). We detected a trend towards an increase in standardized incidence from 32.410/0000 in 2010 (95% CI = 28.76; 36.05) to 73.000/0000 in 2018 (95% CI = 67.86 – 78.15) with the average annual increment of 10.22% (р < 0.0001). A significant proportion of cases (48.17% in 2018) occurred in elderly (≥ 70 years). Standardized mortality rates increased from 9.240/0000 in 2010 (95% CI = 7.29; 11.19) to 28,830/0000 in 2018 (95% CI = 25.51; 32.15), average annual increment 13.99% (р = 0.002). Component analysis showed that an incidence increase by 155.81% is primarily associated with existing risk factors (128.19%) rather than with population aging (16.71%) or their combined effect (10.91%).Conclusion. Currently, there is a clear trend to increasing incidence and mortality from prostate cancer in Kemerovo Region, mainly due to existing risk factors.
The interactions between E- and Z-isomers of 3-O-methoxyimino-pregn-4-ene-20-one and its 17α-hydroxy derivative and transcortin from human blood were investigated. The substitution of the progesterone 3-oxo group for a 3-O-methoxyimino group was shown to diminish the affinity of the steroid for transcortin by approximately one order of magnitude irrespective of the substituent's orientation. The data suggests that progesterone derivatives substituted thereby must have higher bioavailability compared to progesterone and must not significantly affect the biodynamics of glucocorticoid in vivo.
