However, there is little information available regarding the optimum time for harvest. The aim of this 2-year study was to compare the yield, fermentation characteristics and feeding value of kale harvested at different stages of growth, and ensiled with and without a bacterial inoculant. During Year 1 the crop was harvested after 15, 18 and 20 weeks of growth. The yield at each harvest was similar, but as the crop matured the crude protein (CP) concentration and buffering capacity decreased significantly, and there was a marked increase in the water-soluble carbohydrate (WSC) concentration. During Year 2 the crop was harvested at 14 and 17 weeks of growth. Again, the stage of maturity did not affect yield and, on this occasion, the chemical composition of the crop was unaffected by harvest date. However, the WSC and CP concentrations of the crop were lower in Year 2 than in Year 1, possibly as a result of differences in fertilizer regime. Each of the silages produced in Year 2 was offered to six Suffolk cross wether lambs, aged 10 months, to measure voluntary intake, in vivo digestibility and nitrogen retention. Neither harvesting date nor the use of an inoculant affected voluntary intake or nitrogen retention by the lambs. However, in vivo digestibility was higher in the kale silage harvested after 14 weeks of growth and when an inoculant was applied. The results obtained suggest that harvesting kale after 14 weeks of growth can produce highly digestible silage with a high CP concentration. Although delaying harvest until 18 weeks of growth will probably result in a decrease in the CP concentration of the crop, it should also lead to an increase in the WSC concentration of the crop, ensuring a more reliable fermentation.Peer reviewe

Here, a factorial experiment was used to investigate the interactive effects of a UV-B episode and concurrent progressive drought on the growth, chemistry, and reproductive success of A. thaliana. Both drought and UV-B negatively affected rosette growth, although UV-B had the greater effect. Acclimation to UV-B involved adjustment of leaf morphology, while drought induced accumulation of soluble sugars and phenolics. All plants recovered from treatments, but the cost of recovery was a developmental delay resulting in alteration in phenological timings. Combined treatments interacted causing additive negative effects on growth following exposure. This may be linked with inhibition of soluble sugar accumulation by UV-B, restricting the capacity for osmotic adjustment in response to drought. Following cessation of treatments, relative growth rate (RGR) and net assimilation rate (NAR) were significantly stimulated in plants treated with combined drought and UV-B. This interaction alleviated subsequent impacts of elevated UV-B on silique yield and reproductive timings. This study demonstrates the potential for interaction between these two common environmental factors. Furthermore, it shows the changeable nature of these interactions over the course of exposure and recovery through to reproduction, highlighting the need for sustained assessment of such interactions over a plant's lifecycle.

High-value coproducts can greatly improve the feasibility of utilizing plant feedstocks for biorefining and biofuel production. Plant polyphenolics have potential application in the pharmaceutical and cosmetic industries. Orchard grass varieties have been noted for accumulation of polyphenolic compounds, and the current study determined the soluble phenol profile and content in the orchard grass variety 'Abertop'. Hydroxycinnamates and flavonoids were monitored during the transition from vegetative to flowering stage at maximum crop yield. Caffeic acid derivatives, related to bioactives in the Asian medicinal herb Salvia miltiorrhiza , and novel hydroxycinnamate-flavone conjugates were also identified in extracts. Harvest yields of hydroxycinnamates and flavonoids ranged from 2.6 to 4.0 kg/ha and from 2.1 to 5.1 kg/ha, respectively. Abundant compounds showed high levels of antioxidant activity comparable with that of trolox. Minimal changes in soluble phenol content and composition were observed after ensiling with the exception of increases in caffeic acid, a caffeic acid derivative, and a caffeic acid breakdown product, dihydroxystyrene.

Xylitol is a low calorie sweetener that can be produced through a bioconversion approach from lignocellulosic biomass that requires pre-treatment prior to the bioconversion of xylose to xylitol. Steam explosion (SE) is an industrially scalable pre-treatment (PT) process with the potential to liberate xylose monomers, however SE-PT has not been optimized for xylose release from multiple feedstock. The effect of pressure, substrate weight, phosphoric acid loading concentration and residence time on four feedstock [wheat straw (WS), corn stover (CS), Miscanthus (M), and willow (W)] for xylose release and minimal fermentation inhibitor production [furfural and 5-hydroxymethylfurfural (HMF)] was investigated using the Taguchi methodology for design of experiment (DoE) with variation at four levels (4 4). An L 16 orthogonal array design was utilized and all factors indicated influence on xylose release and inhibitor formation and the resulting xylose rich hydrolysate assessed for bioconversion to xylitol. The L 16 DoE gave hydrolysates containing 75-95% of xylose content in the original biomass, whilst retaining cellulose and lignin components in the fiber. The level of inhibitors were within boundary limits to enable microbial fermentation of the hydrolysates to xylitol. Fine tuning of the overall evaluation criteria (OEC) model imbibing 1.5 kg feedstock in 1.2% w/v orthophosphoric acid, 12 bar(g) and 6 min residence time resulted in 90% xylose recovery and production of >1,000 L of wheat straw hydrolysate for bioconversion to xylitol. The advantages and limitations of the Taguchi OEC model and further improvements to this process are discussed in a biorefining context.

Laminarins are storage polysaccharides found only in brown seaweeds, specifically Laminarialaes and Fucales. Laminarin has been shown to have anti-apoptotic and anti-tumoural activities and is considered as a nutraceutical component that can positively influence human health. The structure is species dependent, generally composed of linear ß(1–3) glucans with intrachain β(1–6) branching and varies according to harvest season and environmental factors. Current methods for analysis of molar mass and DP length are technically demanding and are not widely available. Here, we present a simple inexpensive method which enables rapid analysis of laminarins from macroalgal biomass using high-performance anion exchange chromatography with pulsed amperometric detection (HPAEC-PAD) without the need for hydrolysis or further processing. This is based on the linear relationship observed between log10 DP and retention time following separation of laminarins on a CarboPac PA-100 column (Dionex) using standard 1,3-β-d-gluco-oligosaccharides ranging in DP from 2 to 8. This method was applied to analyse laminarin oligomers in extracts from different species harvested from within the intertidal zone on Welsh rocky shores containing laminarin polymers with different ranges of DP. The degree of polymerisation and extrapolated molar mass agreed well with values estimated by LC-ESI/MSn analysis and those reported in the literature.

Oil rich seeds of Allanblackia floribunda, a tree from tropical Africa, have traditionally been used in food preparation. Furthermore, the therapeutic properties of various parts of this tree have long been exploited in traditional medicine. As both food and pharmaceutical industries show growing interest in tropical tree crops, this study aimed to investigate whether A. floribunda seeds could also be used as a source of potentially bioactive compounds. The polyphenol profile revealed six predominant compounds which were identified by HPLC-PDA-ESI/MS as the biflavonoids morelloflavone, Gb-2a and volkensiflavone and their respective glucosides. A range of less abundant flavones, flavonols and flavan-3-ols was also detected. All six major compounds showed antioxidant activity, with the activity of morelloflavone, its glucoside and Gb-2a-glucoside comparable with that of ascorbic acid. The main compounds accounted for approximately 10% of dry weight, making the seeds used for oil production a rich source of biflavonoids as a by-product.
