BackgroundRecombinant hemagglutinin (rHA) and neurominidase (rNA) developed in our investigation are amino acid sequence consensus variants of H1N1 2009 subtype influenza virus strain, also including immunogenic epitopes typical for other influenza virus subtypes (H3N1 and H5N1). Substitutions were made: typical for Russian virus isolates (in HA – S220T, NA – D248N) and in active centers of molecules – R118L, R293L, R368L; C92S, C417S to increase recombinant proteins stability in E. coli. The aim of the present work was to study immunogenicity of the obtained rHA and rNA.Material/MethodsFragments aa 83–469 of NA and aa 61–287 of HA were chosen because they include the main B-cell epitopes and are the minimal structures for correct folding of target proteins. The designed nucleotide sequences were synthesized and purified and the expression of rNA and rNA were analyzed. For immunization and virus challenge we used influenza viruses A/California/04/2009 (H1N1), A/PR/8/34 (H1N1), A/Perth/16/2009 (H3N2), A/Chicken/Kurgan/05/2005 R.G. (H5N1), and B/Florida/04/2006. Specific IgG levels were determined by ELISA in 96-well ELISA plates. Significant differences of survival in mouse groups were analyzed by Mantel-Cox (log-rank) and Gehan-Breslow-Wilcoxon tests.ResultsThe obtained results demonstrate the high immunogenicity and ability of indicated proteins mixture to provide similar cross-protection against influenza viruses of the H1N1 subtype.ConclusionsThe data obtained suggest efficient pluripotent vaccine creation based on HA and NA conservative regions.

Резюме. Вирус гриппа вызывает высококонтагиозное заболевание людей, птиц и млекопитающих. Подсчита-но, что ежегодно от эпидемий гриппа погибает от 250 000 до 500 000 человек и эта цифра может увеличиваться до 1 млн в период возможных пандемий вируса гриппа. Самым эффективным путем профилактики болезни или ее тяжелых последствий является вакцинация. На данный момент существует несколько подходов для создания противогрипозных вакцин, но они имеют ряд недостатков, которые снижают общую эффектив-ность вакцинных препаратов. Соответственно, создание универсальной вакцины, способной обеспечить на-дежную защиту от существующих и возможных реассортантных штаммов вируса гриппа, на данный момент является приоритетной задачей. Использование вакцин на основе рекомбинантных белков позволяет избе-жать рисков, связанных с введением вируса в организм, пусть и инактивированного. Данная работа посвя-щена получению высокоочищенного гибридного рекомбинантного белка, включающего фрагменты белков гемагглютинина вирусов гриппа А и В, а также компоненты флагеллина в качестве адъюванта. Указанные фрагменты белков представляют собой консервативные части гемагглютининов H1, H3, H5 и В, к которым в процессе естественной инфекции образуются специфичные антитела, перекрестно реагирующие с гомоло-гичными эпитопами среди различных штаммов вирусов гриппа А и В. Выбранные участки белков наиболее иммуногенны. Их присутствие обеспечивает как В-клеточный, так и Т-клеточный иммунный ответ. Исполь-зование эпитопов нескольких белков позволяет увеличить эффективность вакцины, а использование гибких мостиков между эпитопами позволяет сохранить правильную пространственную укладку белка и, соответ-ственно, обеспечивает полноценное функционирование каждого эпитопа. Гибридный белок, содержащий В-и Т-клеточные эпитопы разных субтипов вирусов гриппа А и В, а также флагеллин, позволят обеспечить сильный иммунный ответ при меньших затратах на производство. Нами был смоделирован гибридный ре-комбинантный белок Flu-Chim, содержащий иммуногенные эпитопы вирусов гриппа А/H1N1, A/H3N2, A/ H5N1 и В, слитые с фрагментами флагеллина. Был создан высокопродуктивный штамм-продуцент, на основе клеток E. coli BL21(DE3). Была изучена экспрессия гибридного гена flu-chim при различных условиях индук-ции. Также был получен высокоочищенный гибридный рекомбинантный белок Flu-Chim с использованием металлоаффинной хроматографии. Чистота белка после финальной стадии очистки составила 98%. В даль-нейшем планируется изучение иммунологических свойств полученного рекомбинантного белка.
Already about one hundred years BCG remains the only widely used tuberculosis (TB) vaccine. It is known that intensity of the BCG-induced Th1 immune response decreases over time and comes to naught within 10-15 years. It significantly distinguishes BCG from the vaccines providing a lifelong protection such as vaccines against poliomyelitis or measles, and can be bound to natural restriction of duration of a persistence of the BCG-induced CD4 + T-cells. Data on insufficient ability of BCG to stimulate lifelong immunological memory is accumulating. Earlier in our lab on the model of rather resistant to TB C57BL/6 mice infection with the virulent laboratory strain of Mycobacterium tuberculosis (Mtb) H37Rv protective activity (comparable to that of BCG Russia) of 3 subunit vaccine variants was demonstrated as assessed by lung and spleen CFU counts and life span of animals after infection. The aim of this study was to study the characteristics and duration of the immune response induced by the most effective variant of these vaccines. Groups of C57BL/6 mice were vaccinated intramuscularly twice with two-week intervals with 10 μg protein conjugated to 200 μl of an aluminum hydroxide emulsion. Immune response (production of specific antibodies, vaccine proteinstimulated production of interferon gamma and proliferation in vitro) was monitored during 10 months after vaccination. We have shown that the test vaccine induces in mice the formation of long-term immunological memory to a bacterial antigen. Moreover, in the presence of glutoxim the immunological memory spectrum shifts to a "protective" type, i.e. the predominance of the cellular component of the immune response over the antibody response is stimulated. The next step will be the investigation of vaccine effectiveness for revaccination after primary BCG immunization.
