We determined whether a fixed-time AI (TAI) protocol could yield pregnancy rates similar to a protocol requiring detection of estrus, or estrous detection plus TAI, and whether adding a controlled internal device release (CIDR) to GnRH-based protocols would enhance fertility. Estrus was synchronized in 2,598 suckled beef cows at 14 locations, and AI was preceded by 1 of 5 treatments: 1) a CIDR for 7 d with 25 mg of PG F(2alpha) (PGF) at CIDR removal, followed by detection of estrus and AI during the 84 h after PGF; cows not detected in estrus by 84 h received 100 mug of GnRH and TAI at 84 h (control; n = 506); 2) GnRH administration, followed in 7 d with PGF, followed in 60 h by a second injection of GnRH and TAI (CO-Synch; n = 548); 3) CO-Synch plus a CIDR during the 7 d between the first injection of GnRH and PGF (CO-Synch + CIDR; n = 539); 4) GnRH administration, followed in 7 d with PGF, followed by detection of estrus and AI during the 84 h after PGF; cows not detected in estrus by 84 h received GnRH and TAI at 84 h (Select Synch & TAI; n = 507); and 5) Select Synch & TAI plus a CIDR during the 7 d between the first injection of GnRH and PGF (Select Synch + CIDR & TAI; n = 498). Blood samples were collected (d -17 and -7, relative to PGF) to determine estrous cycle status. For the control, Select Synch & TAI, and Select Synch + CIDR & TAI treatments, a minimum of twice daily observations for estrus began on d 0 and continued for at least 72 h. Inseminations were performed using the AM/PM rule. Pregnancy was diagnosed by transrectal ultrasonography. Percentage of cows cycling at the initiation of treatments was 66%. Pregnancy rates (proportion of cows pregnant to AI of all cows synchronized during the synchronization period) among locations across treatments ranged from 37% to 67%. Pregnancy rates were greater (P < 0.05) for the Select Synch + CIDR & TAI (58%), CO-Synch + CIDR (54%), Select Synch & TAI (53%), or control (53%) treatments than the CO-Synch (44%) treatment. Among the 3 protocols in which estrus was detected, conception rates (proportion of cows that became pregnant to AI of those exhibiting estrus during the synchronization period) were greater (P < 0.05) for Select Synch & TAI (70%; 217 of 309) and Select Synch + CIDR & TAI (67%; 230 of 345) cows than for control cows (61%; 197 of 325). We conclude that the CO-Synch + CIDR protocol yielded similar pregnancy rates to estrous detection protocols and is a reliable TAI protocol that eliminates detection of estrus when inseminating beef cows.

Muscle satellite cells were isolated from seven yearling steers implanted for 31 d with a combined implant that contained 120 mg of trenbolone acetate (TBA) and 24 mg of estradiol (E2) and from seven nonimplanted, control steers. Implanted steers had a 28% greater ADG and a 23% greater feed efficiency than did nonimplanted steers. Implanted steers had increased (P<.001) circulating IGF-I concentrations on d 6, 14, and 31 after implantation, and circulating IGF-I concentrations in control steers remained constant or decreased (P<.05) at these times. Maximum fusion percentage was greater (P< .005) in satellite cell cultures isolated from implanted steers (ISC cultures) than in satellite cell cultures isolated from control steers (NSC cultures) (72.8% vs 54.8%, respectively). Satellite cell cultures isolated from implanted steers (ISC cultures) also contained a greater (P<.001) number of myotube nuclei than did NSC cultures (7,998 nuclei/cm2 vs 5,150 nuclei/cm2, respectively). After 72 h in culture, the number of cells (corrected for plating density) was 43% greater (P<.05) in ISC cultures than in NSC cultures. [3H]Thymidine incorporation rates per 10(5) cells at 24 and 34 h after plating were greater (P<.05) in ISC cultures than in NSC cultures; however, incorporation rates did not differ at 72 h. These data indicate that TBA + E2 implantation may result in an in vivo activation of muscle satellite cell proliferation that can be detected in cell culture. This activation may play an important role in TBA + E2-enhanced muscle growth.

Three experiments with factorial arrangements of treatments were designed to test the efficacy of avian-derived polyclonal antibody preparations (PAP) against Streptococcus bovis (PAP-Sb) or Fusobacterium necrophorum (PAP-Fn) in reducing ruminal counts of target bacteria in beef steers supplemented or not with feed additives (300 mg of monensin/d and 90 mg of tylosin/d; MT). Feeding increasing doses of PAP-Sb in Exp. 1 or a single dose in Exp. 2 reduced S. bovis counts in a cubic fashion (P = 0.014). In Exp. 1 and 2, inclusion of MT in the diet had no effect (P > 0.05) on ruminal S. bovis counts. In Exp. 2, ruminal pH was increased (P < 0.05) by feeding PAP-Sb, MT, and PAP-Sb plus MT. Ruminal F. necrophorum counts were reduced by feeding PAP-Fn (P = 0.002) and MT (P < 0.001). Reduction in ruminal F. necrophorum counts was greater (P = 0.008) when feeding MT alone than when feeding PAP-Fn and MT together. In Exp. 3, ruminal S. bovis counts were not affected (P = 0.64) by PAP-Fn. Ruminal pH was not affected (P = 0.61) by feeding PAP-Fn, and the total anaerobic bacterial count was not affected (P > 0.05) by either PAP-Sb or PAP-Fn in Exp. 1 or Exp. 3. In conclusion, PAP of avian origin and against S. bovis or F. necrophorum were effective in reducing target ruminal bacterial populations. These PAP could be effective in preventing the deleterious effects associated with these bacteria, and possibly in enhancing animal performance.

Strip loins from two experiments were used to evaluate effects of feeding dry (DDG) or wet (WDG) distiller's grains on beef color, tenderness, and sensory traits of Holstein steers. In Exp. 1, conducted at the University of Illinois at Champaign-Urbana, dietary treatments consisted of a control whole corn-corn silage diet with soybean meal (SBM) or diets formulated with 12.5% DDG plus urea, 25% DDG, 25% WDG, 50% DDG, or 50% WDG (DM basis). In Exp. 2, conducted at Iowa State University, dietary treatments consisted of cracked corn-corn silage-hay diets with either SBM or urea (Urea) as the control diets, or diets formulated with 10, 20, or 40% DDG or WDG (DM basis). Within each study, strip loins from each of four steers (representing 45.7 and 66.6% of steers in Exp. 1 and 2, respectively) in four replicate pens per treatment were aged for 13 d at 4 degrees C for subsequent color, tenderness, and palatability evaluation. Color of steaks was measured objectively using a HunterLab Miniscan XE spectrophotometer and was subjectively evaluated by a trained panel. Tenderness was measured using the Warner-Bratzler shear force (WBSF) instrument on steaks cooked to 70 degrees C. For sensory evaluation, 95 consumers were recruited to evaluate tenderness, juiciness, and flavor of cooked steaks. In Exp. 1, steaks from steers fed 25% WDG had higher (P < 0.05) a* values after 138 h of simulated retail display than all other treatments, except for those from steers fed 12.5% DDG. In Exp. 2, a greater (P < 0.05) percentage of steaks from steers fed 40% DDG or 40% WDG were considered moderately undesirable during retail display (steaks that received a consumer acceptability score of 3 or less). There were no (P = 0.20 in Exp. 1, and P = 0.33 in Exp. 2) differences among treatments in Exp. 1 and Exp. 2 for WBSF (1.47 +/- 0.66 kg and 1.58 +/- 0.72 kg, respectively) or taste panel tenderness (5.7 +/- 0.30 and 6.2 +/- 0.22, respectively), beef flavor (6.0 +/- 0.23 and 6.2 +/- 0.22, respectively), and juiciness (5.6 +/- 0.31 and 5.8 +/- 0.23). Feeding distiller's grains at up to 50% of the dietary DM did not affect tenderness or sensory traits, and seems to be a viable feed alternative without negatively impacting sensory attributes.

Strip loins from 236 carcasses from crossbred yearling steers were collected on each of 2 slaughter dates (slaughter 1 or 2) to determine the effects of feeding corn or sorghum distillers grains (DG) on beef color, fatty acid profiles, lipid oxidation, tenderness, and sensory attributes. Dietary treatments consisted of a steam-flaked corn (SFC) diet without (control) or with 15% (DM basis) corn dry or wet DG (CDDG and CWDG) or sorghum dry or wet DG (SDDG and SWDG) and alfalfa hay (R). Additional treatments included SDDG or SWDG with no alfalfa hay (NR). In slaughter 2, steaks from steers fed SFC had lesser L*, but greater a* (P < 0.05) values than those from steers fed DG. When comparing sorghum and corn DG steaks, the same color differences were detected. Steaks from steers fed sorghum DG had lower L*, but greater a* (P < 0.05) values than those from steers fed corn DG. Also, L* values in steaks from steers fed SWDG with R were greater (P < 0.05) than those from steers fed SWDG with NR. In slaughter 1, feeding DG increased (P < 0.05) steak n-6 fatty acid concentrations compared with SFC. In both slaughter groups, feeding dry DG increased (P < 0.05) steak linoleic acid concentrations compared with wet DG. In slaughter 2, feeding corn DG diets increased (P < 0.05) linoleic acid concentrations of steaks compared with sorghum DG diets. In addition, increased (P < 0.05) concentrations of alpha-linolenic acid in steaks resulted from feeding SDDG or SWDG with R compared with those sorghum treatments with NR. In each slaughter group, feeding DG increased (P < 0.05) the n-6:n-3 ratio of steaks compared with SFC, and feeding corn DG increased (P < 0.05) this ratio compared with sorghum DG. Furthermore, steaks from steers fed corn DG had greater (P < 0.05) concentrations of trans-vaccenic acid than those from steers fed sorghum DG. In slaughter 1, the CLA isomer 18:2, trans-10, cis-12 was greater (P < 0.05) in steaks from DG diets. On d 1 of retail display, steaks from steers fed SDDG with R in slaughter 2 had greater (P < 0.05) thiobarbituric acid reactive substances values than those from steers fed SDDG with NR. Feeding DG at 15% of the dietary DM did not affect sensory attributes or Warner-Bratzler shear force values of steaks. Feeding DG from either corn or sorghum as either a wet or dry by-product had no effect on beef sensory attributes.
