The expressions of trypsin and chymotrypsin in the pyloric caeca of Atlantic salmon (Salmo salar L.) were studied in three experiments. Two internal (trypsin phenotypes, life stages) and three common external factors (starvation, feeding, temperatures) influencing growth rates were varied. Growth was stimulated by increased temperature and higher feeding rate, and it was depressed during starvation. The interaction between trypsin phenotype and start-feeding temperature affected specific activity of trypsin, but not of chymotrypsin. Trypsin specific activity and the activity ratio of trypsin to chymotrypsin (T/C ratio) increased when growth was promoted. Chymotrypsin specific activity, on the other hand, increased when there was a reduction in growth rate whereas fish with higher growth had higher chymotrypsin specific activity resulting in lower T/C ratio value. During a rapid growth phase, trypsin specific activity did not correlate with chymotrypsin specific activity. On the other hand, a relationship between specific activities of trypsin and chymotrypsin could be observed when growth declined, such as during food deprivation. Trypsin is the sensitive key protease under conditions favouring growth and genetically and environmentally affected, while chymotrypsin plays a major role when growth is limited or depressed. Trypsin specific activity and the T/C ratio value are shown to be important factors in the digestion process affecting growth rate, and could be applicable as indicators for growth studies of fish in captive cultures and in the wild, especially when food consumption rate cannot be measured.

An earlier study demonstrated that under-yearling (0+) Atlantic salmon (Salmo salar L.) smolt had a lower vertebral mineral content and mechanical strength and higher prevalence of vertebral deformities than 1+ smolt during the early seawater (SW) phase. The present study aimed to examine if commercial extruded high-energy diets need to be supplemented additional minerals for proper bone mineralization and prevention of bone deformities in fast growing 0 + smolts. We studied vertebral morphology with radiology, and bone mineral content and mechanical strength in 60 g 0+ smolt fed diets with a normal (NM) or elevated (HM) bone mineral (P and Ca) contents from SW transfer (week 0) until 10 times weight increase at week 17. Thereafter, both groups were fed a commercial diet until a mean slaughter weight of 4100 g after 57 week. There were no differences in body weight and length between the dietary groups during the study, while the condition factor differed significantly at the final sampling (NM 1.40; HM 1.29). The most common bone deformity observed was compressions in the tail region of the vertebral column. Lower incidences of vertebral deformities (percent individuals with one or more deformed vertebrae) was observed in the HM group in week 17 (HM 20%; NM 47%) and week 57 (HM 37%; NM 73%), also reflected by higher vertebral length/dorso-ventral diameter ratio in weeks 17 (HM 0.99; MN 0.92) and 57 (HM 0.97; NM 0.88). The HM group had significantly higher vertebral mineral content (HM 550 g kg )1 ; NM 480 g kg )1 ) and mechanical strength (HM 9050 g mm )1 ; NM 4600 g mm )1 ) than the NM group after 8 week feeding. Plasma levels of Ca, P and D-vitamin metabolites recorded in week 8 reflected changes in P homeostasis, but could not explain the preventive effect of the HM diet on development of bone deformities. The results suggest that elevated dietary mineral content during the early SW phase may reduce the prevalence of vertebral deformities in fast growing 0 + salmon smolts. KEY WORDS

In August 1998, 3000 Atlantic salmon Salmo salar L. parr were divided into 7 groups with 2 replicates. Every 6 wk until March of the following year 1 group was vaccinated. One group was held as an unvaccinated control. The fish were transferred to seawater in May 1999, and slaughtered in February 2000. Temperature, fish size and photoperiod at vaccination, and the time between vaccination and sea transfer thus varied among the groups. In all vaccinated groups, growth was reduced for 1 to 2 mo following vaccination. Intra-abdominal lesions developed faster, and stabilised at a higher level in the groups vaccinated early at the highest temperature and the smallest fish size. Growth in seawater was influenced by the time of vaccination. At the end of the experiment, the group vaccinated last (MAR) was the heaviest of the vaccinated groups (4.0 kg), and the group vaccinated first, i.e. in August (AUG) was smallest (3.2 kg). Growth rate in seawater differed only in the summer when specific growth rate was above 1.45 in all groups. There was a correlation between adhesion, condition factor and number of weeks from vaccination to sea transfer. The AUG group had the highest condition factor, with a top level of 1.64 in autumn, and this group also displayed the highest incidence of deformed vertebra. The experiment shows that side effects of vaccination can be significantly reduced when planning the vaccination strategy, by taking environmental factors and fish biology into consideration.

This study investigates the effects of water temperature (T) on vaccine-induced abdominal lesions (i.p. injection with oil-adjuvant vaccine) and vertebral deformities in Atlantic salmon. Quadruple groups of vaccinated (V) or unvaccinated (U) underyearling smolts were reared in tanks under four different temperature regimes for 6 weeks in fresh water (FW) followed by 6 weeks in sea water (SW). The four different T regimes were 10 °C FW-10 °C SW (10-10), 10 °C FW-16 °C SW (10-16), 16 °C FW-10 °C SW (16-10) and 16 °C FW-16 °C SW (16-16). After the temperature regimes were finished, the fish were group-tagged and transferred to a common sea cage for on-growth until harvest size. At termination, weight was significantly affected by both T and V, while lesion score and deformities were affected by T only. The weight difference between the largest and smallest U group was 20.3% (16-10 U: 2.4 kg, 10-16 U: 1.89 kg), while the largest difference between U and V fish within a T regime was 28.7% (16-16 U: 2.1 kg, 16-16 V: 1.5 kg). Fish from the 16-16, 16-10 and 10-16 regimes had a significant higher lesion score than those from the 10-10 regime. Fish from the 10-16 and 16-16 regimes displayed a significantly higher prevalence of vertebral deformities (palpation : 13-27%, radiology: 88-94%) than fish from the 10-10 and 16-10 regimes (palpation: 2-3%, radiology: 27-65%). Vertebra number 26 (located beneath the dorsal fin) was the most frequently affected vertebra in smolts, while vertebra number 43 (located above the anal fin) was most frequently affected in adults.

The aim of the present study was to elucidate whether vertebral deformities influence on the growth performance of salmon, and if so, at what level of severity the growth performance ⁄ welfare is affected. To do this, Atlantic salmon postsmolts were individually tagged and reared in a common seacage from January 2005 until January 2006. At the end of the experiment, dissected vertebral columns were radiographed, evaluated for vertebral deformities, and the location and type of deformity was recorded. For statistical analysis the fish were grouped according to the number of deformed (d) vertebrae (v); 0 dv (n = 173), 1-5 dv (n = 43), 6-10 dv (n = 11), 11-20 dv (n = 18) and 21-44 dv (n = 31). In fish with less than six dv, the deformities were found within the tail fin region (v 50-58). Fish with 11-20 dv also had a high occurrence of dv within the tail region (V31-49), and fish with more than 20 dv also had a high occurrence of dv within the caudal trunk region (V9-30). Growth was significantly influenced by the number of dv. At the end of the experiment, fish with more than 10 dv were shorter and had a higher condition factor than normal fish, and fish with more than 20 dv were shorter, had lower weight and a higher condition factor than normal fish. In the first period in seawater (January-June) there was no detectable differences in growth (length or weight) between the groups, but the condition factor of the individuals with a high number of dv increased. In the second period (June-January), fish with more than 10 dv, had a significantly lower growth rate. This study indicate that a low occurrence of vertebral deformities have little effect on the growth performance ⁄ welfare of salmon.
