Oxidative stress is one of the most important pathological processes in chronic heart failure caused by hypertension. These processes involve MYC-regulated mechanisms, including the induction of CYP2E1 as a potent prooxidant factor. In this work, we used qPCR, Western blot analysis, and biochemical markers of oxidative stress to investigate the ability of quercetin to inhibit oxidative stress by modulating MYC expression. We studied spontaneously hypertensive rats (SHRs) in which the onset of cardiac pathology was observed at least at 4 months of age and the development of pathology occurred during life up to 22 months of age. Wistar rats were used as normotensive controls. We observed overexpression of the transcription factor MYC (p=0.0024) in the myocardium of SHRs compared to normotensive controls, and an increased expression of MYC-target gene, CYP2E1, (p=0.0001) in the old SHR group compared to young SHRs. This probably contributed significantly to the development of oxidative stress in the cardiac tissue of old SHRs. We demonstrated that long-term treatment of old SHRs with quercetin resulted in dramatic inhibition of MYC (p=0.0000), and a significant decrease in CYP2E1 (p=0.0001) expression and CYP2E1 protein levels (p=0.0136). This probably contributed significantly to the decrease in lipid peroxidation (p=0.0000). Quercetin was also able to activate antioxidant activity, resulting in a significant improvement in the prooxidant-antioxidant balance in the heart. In turn, the elimination of oxidative stress could contribute to a decrease in blood pressure (p=0.0000) and relative heart weight (p=0.0071) in quercetin-treated old SHRs compared to the untreated old SHR group.
З використанням методу patch-clamp у конфігурації nucleus attached встановлено, що ядерна мемб-рана кардіоміоцитів щура містить різні типи іонних каналів з провідністю у діапазоні від 10 до 400 пСм. Зокрема, зареєстровано інозитол-1,4,5-трифосфатні ВСТУПЯдерна оболонка є напівпроникним бар'єром між цитоплазмою та нуклеоплазмою. Тран-спорт крізь ядерну мембрану забезпечуєть-ся наявністю у ній численних ядерних пор та іонних каналів з різними біофізичними властивостями. Незважаючи на велику різно-манітність іонних каналів у ядерній мембрані [1][2][3][4][5][6][7] та численні дослідження, присвячені внутрішньоклітинним сигнальним механіз-мам, зокрема Са 2+ -сигналізації, фізіологічна роль катіон-селективних іонних каналів у ядерній мембрані залишається остаточно нез'ясованою. Ключова роль іонів кальцію у скороченні кардіоміоцитів є загальновизна-ною. Значно менш дослідженим залишається питання механізмів участі Ca 2+ у регуляції транскрипції генів, адже відомо, що кальцієві сигнали поширюються не лише в цитозолі, але й у ядрі [8][9][10]. Іони кальцію можуть пасивно дифундувати із цитоплазми у ядро через ядерні пори [11]. Крім пор у ядерній мембрані наявні специфічні системи тран-спортування Са 2+ -інозитол-1,4,5-трифос-фатні [1,12] і ріанодинові рецептори [13,14]. Таким чином іонні канали ядерної мембрани є невід'ємною частиною системи Са 2+ -сигна-лізації, а відтак відіграють важливу роль у функціонуванні серцевого м'яза і можуть стати мішенню нових типів медичних препаратів для лікування захворювань серця. Також слід зазначити важливість досліджень іонних ка-налів ядерної мембрани на різних об'єктах, адже відомо, що їх експресія та, відповідно, характер поширення Са 2+ -сигналів можуть відрізнятися залежно від типу клітин. Раніше ми запропонували спосіб ізолювання ядер, придатних для дослідження методом patchclamp та описали високопровідні катіонні ка-нали (LCC-channels), інозитол-1,4,5-трифос-фатні рецептори та інші іонні канали ядерної мембрани нейронів центральної нервової системи [1].Метою цієї роботи було ідентифікувати іонні канали ядерної мембрани кардіоміо-цитів. Вчені давно намагаються підібрати оп-тимальні умови для виділення ядер із різних типів тканин. Перші протоколи ізолювання ядер із застосуванням центрифугування в градієнті цукрози розробив Svedberg і співавт. ще у 40-х роках ХХ ст. Пізніше Potter (1946), Schneider (1948 та інші дослідники вдосконалили цей метод, застосувавши гіпертонічні розчини цукрози. Проте виділи-ти цілісні ядра на той час ще не вдавалося (див. [9]). У ядерних мембранах нейронів та кардіоміоцитів у значній кількості експресуються катіонні канали великої провідності (LCC-канали). За допомогою електрофізіологічної реєстрації струмів крізь внутрішню ядерну мембрану нейронів Пуркіньє мозочка і кардіоміоцитів перевіряли дію на LCC-канали низки модуляторів (активаторів та інгібіторів) н-холінорецепторів. Серед досліджених нами речовин виражений вплив чинили рокуронію і піпекуронію броміди та нікотин. Фармакологічна чутливість LCC-каналів ядерних мембран нейронів Пуркіньє мозочка і кардіоміоцитів є майже ідентичною, що свідчить про наявність єдиної популяції цих каналів у досліджуваних об'єктах.У концентрації 0,2 ммоль/л нікотин вдвічі зменшував амплітуду струму через LCC-канали, в той час як рокуронію бромід та піпекуронію бромід у такій самій концентрації інгібували на 7 і 12% відповідно. Отже, серед перелічених речовин лише нікотин діє на LCC-канали з ефективністю, наближеною до впливу раніше дослідженого нами тубокурарину, відповідно саме вони залишаються найбільш перспективними для подальшого застосування як інструменту при вивченні фізіологічної ролі цих каналів. Large-conductance cation channels (LCC-channels) were found in both (inner and outer) nuclear membranes of cerebellar Purkinje neurons. They are the most common type of intracellular spontaneously active ion channels among other identified. Their structure and physiological functions are still unknown, but the previous findings confirmed their sensitivity to a number of agonists/antagonists of nicotinic acetylcholine receptors. The purpose of the investigation was to estimate the effect of other regulators of the N-cholinoreceptors functioning – neostigmine, hexamethonium, and methyllycaconitine (MLA) on the LCC-channels in the nuclear membrane of cerebellar Purkinje neurons of rats. The effect of the agents was estimated based on changes in the following biophysical parameters: current amplitude, Po, channel flickering effect. Ion currents through single channels were registered using the patch-clamp technique in a nucleus-attached mode in voltage-clamp configuration. Among the studied substances, only MLA and hexamethonium influenced the LCC-channels functioning. Hexamethonium at a concentration of 2 mM reduced the Po of the LCC-channels by 46%. Under the influence of MLA, a slight effect of channel flickering was observed ("Poisson surprise" was 2.14 in the control and 3.81 under the influence of 200 μM of the substance respectively). No significant change of the biophysical characteristics of the LCC-channels under the influence of neostigmine was detected. Despite the low efficiency as LCC-channels blockers, the lack or only slight effect is a strong argument in favor of the substance usage in medicine due to their wide therapeutic potential. The severity of their effects is necessary for a comprehensive analysis of the effect patterns of the abovementioned substances on the molecular dynamics of the studied channels. The results will also be important for the identification or synthesis of new and more effective inhibitors of the LCC-channels.
For seven years of researching the transport systems of nuclear membranes using the patch-clamp method, we observed a certain pattern: in winter, this method's efficiency significantly decreased. Since different seasons are characterized by different light and temperature indicators, we decided to pay attention to the latter's possible impact on the success of the research. Therefore, the purpose of this work was to test the influence of seasonal factors such as changes in daylight hours, temperature, atmospheric pressure, precipitation, and cloudiness on the quality of patchclamp recordings of ion currents through the LCC channels of the nuclear membrane of cardiomyocytes and cerebellar Purkinje neurons. We assumed that with decreasing daylength and decreasing temperature, the patch-clamp registrations' qualitative and quantitative indicators also decrease. We applied Pearson's correlation analysis with initial data on daylight hours, meteorological conditions, and calculated progress of registrations (%) for a specific day to test this assumption. Based on the results of this analysis, we found out that there is a direct pronounced linear dependence of the quality and number of registrations on the length of daylight hours (r = 0.6) and temperature (r = 0.6), as well as a weak inverse dependence on cloudiness (r = 0.3). Analysis of variance (ANOVA) also confirmed a significantly greater success of registrations performed in the summer compared to the winter of the same year. The obtained results can become the basis for optimizing the research activities of working groups studying intracellular transport systems' functioning by electrophysiological methods, in particular, patch-clamp.
