Tape-stripping of the skin is a useful method for removing the stratum corneum and obtaining more information about the function of this skill layer as the main barrier for skin penetration. The amount of stratum corneum removed is of relevance in establishing the concentration profile of chemicals within the stratum corneum after topical application. Weighing is the preferred method for measuring the amount stripped, but because it is often subject to artifacts, alternative methods are sought. We present a simple, colorimetric method for determining the amount stratum corneum removed by sequential adhesive-tape-stripping of human skin in vivo. The method is based on quantification of the sodium hydroxide soluble protein fraction using a commercially available protein assay similar to the Lowry assay. The method is shown to be an accurate and reproducible alternative to weighing, also demonstrating uniform removal of stratum corneum layers following the very initial strips.

Optically active D-arylglycines, which are of interest for preparation of semisynthetic penicillins and cephalosporins, were isolated from the racemic mixtures of their derivatives using immobilized proteolytic enzyme subtilisin (EC No. 3.4.4. 16). The performance of these reactions in two-phase systems, consisting of water and an immiscible organic solvent, improved the yield, purity, and economics of the process by increasing the substrate solubility and reducing the rate of nonenzymatic hydrolysis. The proportion of the organic phase can be as much as 75% of the overall volume without seriously impairing the enzymatic activity. The optically pure D-and L-arylglycines were liberated from their D- and L-derivatives by acid hydrolysis. The substituent influence of the various arylglycine derivatives on the rate of the enzymatic cleavage reaction was investigated.

Zusammenfassung: Durch Gelfiltration an Sephadcex G-50 konnte aus Handels-Insulin einehöherrmolekulare Fraktion b gewonnen werden, die durch TIrypsineinwirkung teilweise in Des-Ala-Insulin ütbergeht, wie elektrophoretisch nachgewiesen werdcen konnte. An lonenaustauschersäulen konnten aius dieser Fraktion zwei Präparate b 2 und b 3 isoliert werden, die sich durch Trypsin quantitativ in D)es-Ala-Insulin umwandeln ließen. Aber nur eine diieser Fraktionen (b 3 ) erfüllte die für einen einbettigen Insulinvorläufer notwendige Voraussetzung, nach Sulfitolyse ebenfalls nur ein einziges Siummary: Bovine pro insulin: isolation, properties amd activation by trypsin. The gel-filtration of commercial insulin on Sephadex G-50 revealed a high molecular weight fraction b. By electrophoresis, it was shown that fraction b was partly converted into des-Ala-insulin by the action of trypsin. Two preparations, b 2 and b 3 , were separated from fraction b by ion exchange chromatography. Both b 2 and b 3 were converted quantitatively into des-Ala-insulin by the action of trypsin.
