Sequential carbophilic addition of organolithium reagents and N-acyliminium ion cyclization of N-phenethylimides 1 affords the substituted isoquinolones 3 in high yields, with the possibility of varying the substituent at the C-1 position of the isoquinoline ring by changing the organolithium reagent. Ready access to the isoquinoline nucleus via Parham-type cyclization of imides 2 is also described. We have shown that iodinated imides 2 tolerate the metal-halogen exchange in the presence of the imide group, and the intramolecular cyclization of the so-obtained aromatic organometallic derivatives leads to the corresponding enamides 4. Both approaches have allowed the efficient preparation of various types of the isoquinoline class of alkaloids, just by changing the substitution pattern on the readily available starting imides. Thus, we have developed convenient alternative routes for the synthesis of benzo[a]quinolizidones and their 2-oxa analogs, isoindoloisoquinolones, dibenzo[a,h]quinolizidones, and thiazolo- and oxazolo[4,3-a]isoquinolones.

The antituberculosis activity of fully‐aromatized quaternary benzo[c]phenanthridine bases prepared in our laboratory was tested under the Tuberculosis Antimicrobial Acquisition and Coordinating Facility (TAACF) program. Professor Armin de Meijere zum 65. Geburtstag gewidmet Die Entwicklung effizienter und umweltverträglicher Synthesemethoden ist eine wichtige Aufgabe der modernen Chemie. Hierbei hat sich das Dominokonzept als sehr erfolgreich erwiesen.[1] Bei Dominoreaktionen werden Bindungen und neue Funktionalitäten aufgebaut, die in nachfolgenden Schritten unter identischen Bedingungen wiederum unter Bindungs-und Funktionalitätsbildung weiterreagieren. Je größer die Zahl der gebildeten Bindungen und je höher die Komplexität des Produkts ist, desto besser ist die Qualität einer Dominoreaktion. Zwar ist inzwischen eine Fülle von Zwei-Stufen-Dominoreaktionen bekannt, Drei-Stufen-Transformationen sind jedoch eher die Ausnahme. Hier beschreiben wir eine neuartige Dominoreaktion zum Aufbau von drei Bindungen in aufeinander folgenden Schritten, die einen effizienten Zugang zum Gerüst der Erythrina-und B-Homoerythrina-Alkaloide ermöglicht.Die Erythrina-Alkaloide [2] wie Erysodin (1) sind eine weit verbreitete, strukturell interessante Naturstoffklasse mit ausgeprägter biologischer Aktivität.[3] Zahlreiche Verbindungen dieser Familie zeigen Curare-ähnliche Wirkungen sowie hypotensive, sedative und ZNS-depressive Eigenschaften. [4] Die Retrosynthese des Gerüsts der Erythrina-und B-Homoerythrina-Alkaloide 2 a und 2 b im Rahmen des Dominokonzepts führt über die N-Acyliminiumionen 3 a bzw. 3 b sowie die metallierten Amide 4 a bzw. 4 b zum Amin 5 a und den Cyclohexenderivaten 6 a bzw. 6 b (Schema 1). Hierbei sollten die intermediären N-Acyliminiumionen 3 durch intramolekulare Addition eines Aluminiumkomplexes des primären Carbonsäureamids [5] an die Enolacetat-Komponente von 4 mit anschließender Eliminierung von Essigsäure zugänglich sein. Der Aluminiumkomplex würde in situ durch Reaktion des primären Amins 5 mit der Esterfunktion der Enolacetate 6 gebildet. Die erforderlichen Enolacetate 6 a und 6 b ließen sich einfach durch Umsetzen der bekannten Ketone [6] 7 a und 7 b mit Isopropenylacetat in Gegenwart katalytischer Mengen an p-Toluolsulfonsäure in 86 bzw. 93 % Ausbeute erhalten.Zur Synthese von 2 a und 2 c wird das Amin 5 a bzw. 5 b mit Trimethylaluminium in Benzol versetzt, eine Stunde bei Schema 1. Retrosynthese und Synthese des Erythrina-und B-Homoerythrina-Gerüsts 2 sowie der Enamine 8 und 9.
