α-Onocerin: An Acetylcholinesterase Inhibitor fromLycopodium clavatum

Abstract: In order to find new acetylcholinesterase inhibitors from natural sources which have therapeutic potential in the treatment of Alzheimer's disease, the chloroform : methanol (1 : 1) extracts of five Lycopodium species ( L. clavatum L., L. selago L., L. annotinum L., L. alpinum L., and L. complanatum ssp. chamaecyparissus A. (Br.) Döll.), growing in Turkey (Lycopodiaceae), were screened in vitro by the Ellman spectrophotometric method at 1 mg/ml concentration. Among these extracts, only the extract of L. clavat… Show more

“…While Lycopodium species have been extensively searched for their alkaloid contents [22][23][24], we have encountered few studies on its nonpolar components; namely sterols, hydrocarbons, and fatty acids [31,32]. We also reported the isolation of some non-polar compounds; namely palmitic acid, cerotic acid, tetradecyl acetate, and 24-alpha-methylcholest-5-enol, and a serratane-type triterpene, alpha-onocerin [33,34]. It has been reported that amphotericin B complexed with cholesterol is at least four times more active than amphotericin B alone in L. donovani-infected hamsters [35].…”

“…Os resultados obtidos com a huperzina A estimularam Orhan et al 24 a estudarem outras 5 espécies de Lycopodium, à procura de outro metabólitos inibidores de AChE. Após uma avaliação preliminar dos extratos pelo ensaio de Ellmann 20 , o extrato das partes aéreas de L. clavatum foi selecionado e o fracionamento bio-guiado resultou no isolamento da α-onocerina (12a).…”

“…Among them, artocarpuate A, artocarpuate B and a-onocerin have also been cited. 3,19 Some biological activities have been reported for lanostane-type triterpenes, but none have been found to be related to an activity toward the enzyme AChE. 20,21 Further semi-synthetic derivatives are necessary to disclose some structure-activity relationship.…”
