α-Lipoic acid induces Endoplasmic Reticulum stress-mediated apoptosis in hepatoma cells

Abstract: Hepatocellular carcinoma (Hcc) is the most common liver cancer and a major cause of adult death. the current treatments for HCC suffer from drug resistance and poor prognosis; therefore, novel therapeutic agents are urgently needed. Phytochemicals have been proposed to treat a range of cancers. Among them, α-lipoic acid (α-LA), a naturally synthesized antioxidant found in various dietary animal and plant sources, prevents oxidant-mediated cell death in normal cells while inducing apoptosis in several cancer ce… Show more

“…ER stress led to activation of PERK (phospho- extracellular signal-related kinase) pathway and IRE1 (inositol-requiring enzyme 1) pathway leading to apoptosis. α-lipoic acid also inhibited ATF6 (activating transcription factor 6)-mediated pro-survival pathway [ 72 ].…”

“…48 Hence, interfering with or reversing of hypoxia and its effects or identifying a suitable way to increase ROS level in tumor cells can reduce the drug resistance of tumors and improve the therapeutic effect. 32,49,50 In turn, tumor cell aerobic glycolysis can influence HIF-1α by upregulating glutamine. The TCA cycle is the hinge of metabolism of glucose, fat and amino acid.…”
