Double experimental designs, Plackett–Burman design coupled with Box–Behnken design, were used to optimize the simultaneous extraction process of arabinogalactan and taxifolin from Larix gmelinii by ultrasonic microwave synergistic extraction (UMSE) method. The optimal extraction conditions were determined with the extraction yields of 223.40 ± 7.82 mg/g for larch arabinogalactan and 125.02 ± 5.30 mg/g for taxifolin. Compared to conventional methods, UMSE method showed the merits of higher extraction efficiency, saving time, and energy. The molar ratio of galactose to arabinose is found to be 6:1 through the monosaccharide composition analysis of larch arabinogalactan. The obtained taxifolin structure was identified by mass spectrometry, Fourier transform infrared spectroscopy, and nuclear magnetic resonance. Interestingly, larch arabinogalactan and taxifolin showed completely opposite influences on α‐amylase, α‐glycosidase, and pancreatic lipase; it was inferred that larch arabinogalactan could act as an inhibitor while taxifolin as an accelerator. Practical applications In this study, response surface methodology of Plackett–Burman design and Box–Behnken design performed well to obtain the optimal conditions and satisfactory extraction yields for the extraction of arabinogalactan and taxifolin from Larix gmelinii by ultrasonic microwave synergistic extraction method, which could be a good paradigm for the efficient isolation of bioactive substances from plant matrix in the field of separation science. Meanwhile, the results from the evaluation of the effects of two target compounds on activities of α‐amylase, α‐glycosidase, and pancreatic lipase in vitro revealed that larch arabinogalactan and taxifolin enjoyed the completely opposite influences on the three digestive enzymes, namely larch arabinogalactan could act as an inhibitor while taxifolin an accelerator. These results will facilitate the application of larch arabinogalactan and taxifolin in the food and pharmaceutical industries.

Proanthocyanidins were separated for the first time from Cinnamomum longepaniculatum leaves. An experiment-based extraction strategy was used to research the efficiency of an ultrasound-assisted method for proanthocyanidins extraction. The Plackett-Burman design results revealed that the ultrasonication time, ultrasonic power and liquid/solid ratio were the most significant parameters among the six variables in the extraction process. Upon further optimization of the Box-Behnken design, the optimal conditions were obtained as follows: extraction temperature, 100°C; ethanol concentration, 70%; pH 5; ultrasonication power, 660 W; ultrasonication time, 44 min; liquid/solid ratio, 20 mL/g. Under the obtained conditions, the extraction yield of the proanthocyanidins using the ultrasonic-assisted method was 7.88 ± 0.21 mg/g, which is higher than that obtained using traditional methods. The phloroglucinolysis products of the proanthocyanidins, including the terminal units and derivatives from the extension units, were tentatively identified using a liquid chromatography with tandem mass spectrometry analysis. Cinnamomum longepaniculatum proanthocyanidins have promising antioxidant and anti-nutritional properties. In summary, an ultrasound-assisted method in combination with a response surface experimental design is an efficient methodology for the sufficient isolation of proanthocyanidins from Cinnamomum longepaniculatum leaves, and this method could be used for the separation of other bioactive compounds.

The chemical profiles of four ginseng roots samples from three species of ginseng (Panax quinquefolius, Panax ginseng and Panax notoginseng) and two commercial ginseng products containing P. quinquefolius and red P. ginseng were compared using ultra-performance liquid chromatography coupled with quadrupole time of flight mass spectrometry (UPLC/QTOF-MS). Principal component analysis allowed a holistic approach in showing distinct chemical differences between the three ginseng species and correct classification of the two commercial products to their respective species. Further investigation of the chemical profile variations yielded ten main markers that were distinct for the three species. This study shows the potential of chemical profiling for the classification of complex natural product samples, such as ginseng, and application to commercial products sold in the market. This methodology can assist the industry in authenticating the various species of ginseng and providing a quick assessment of the quality of commercial ginseng products. Chen (Tienchi or Sanchi, notoginseng) are the most commonly used and studied ginseng herbs.1 Due to the continual increases in sales, there has been an increase in concern of adulteration of herbal products sold in the market. 4 Practices including misidentification of herbal species and the addition of unlabeled pharmaceuticals 5 can cause harm to the consumer and potential mistrust in the herbal and traditional medicine industry. Specifically for ginseng, intentional substitution of the wrong species in ginseng products in the market is a known issue. 6 To address this growing concern, many analytical methods have been developed to authenticate and classify the various species of ginseng.Morphological examinations 7 to genetic testing 8 has been commonly used to authenticate different species of ginseng. However, certain products in the market can be packaged ground into powder or prepared differently (extracts) which can decrease the effectiveness of these methods.
