Capillary electrophoresis (CE) is being widely applied in pharmaceutical, biological, food, chemical, environmental, and clinical fields owing to its high efficiency, universality, sensitivity, trace consumption of sample, high sample throughput, and variety of operating modes. However, it is also facing great challenges in determination and analysis of ultratrace samples, semipreparation or preparation of samples, multidimensional analysis, and coupling of multiple technologies. Load capacity is a key factor that affects the solution of these issues. It is thought that the direct increase in capillary diameter is a shortcut. Many papers and patents reported the development of various electrophoresis methods in large capillaries (>150 µm), which are uniformly named wide‐bore electrophoresis (WBE) here. This article describes the progress in WBE, including the theory, methods, applications, and so on.

Density Functional Theory Study of the Interaction between Guanine and Catechin

The interacting patterns and mechanism of the catechin and guanine have been investigated with the density functional theory B3LYP method by 6-31G* basis set. Fourteen stable structures for the catechin-guanine complexes have been found which form two hydrogen bonds at least. The results indicate that the complexes are mainly stabilized by the hydrogen bonding interactions. At the same time, the number and strength of hydrogen bond play a co-determinant parts in the stability of the complexes which can form two or more hydrogen bonds. Theories of atoms in molecules (AIM) and natural bond orbital (NBO) have been adopted to investigate the hydrogen bonds involved in all systems. The interaction energies of all complexes have been corrected for basis set superposition error (BSSE), ranging from -38.86 to -14.56 kJ/mol. The results showed that the hydrogen bonding contributes to the interaction energies dominantly. The corresponding bonds stretching motions in all complexes are red-shifted relative to that of the monomer, which is in agreement with experimental results.

By precisely fixing an inner cooling capillary to the axial center of a quartz tube, a new and better wide-bore electrophoresis (WBE) system was built and has shown great potential in load capacity and separation efficiency. Further, a novel two-dimensional hybrid of HPLC and WBE system (HPLC–WBE) was developed with an injection valve as the interface. High-resolution and rapid analysis speed were observed in both the WBE system and the 2D HPLC–WBE system. Further work is currently underway.
