Yorkie: the final destination of Hippo signaling

Abstract: The Hippo signaling pathway is a key regulator of growth during animal development, while loss of normal Hippo pathway activity is associated with a wide range of cancers. Hippo signaling represses growth by inhibiting the activity of a transcriptional co-activator protein, known as Yorkie in Drosophila and Yap in vertebrates. In the five years since the first report linking Yorkie to Hippo signaling, intense interest in this pathway has led to rapid increases in our understanding of the action and regulation … Show more

“…Since several excellent reviews have been published on these signaling pathways (for recent reviews, see Kango-Singh and Singh, 2009;L. Zhang et al, 2009;Grusche et al, 2010;Halder and Johnson, 2010;Oh and Irvine, 2010;Pan, 2010;Yi and Kissil, 2010;Zhao et al, 2010), we have not described these pathways in detail in this review. Rather, we aim to review recent advances in determining leg segment size and PVs in the leg segment, focusing on the Ds-Ft and Wts-Hpo signaling pathways.…”

“…The protocadherins Ds and Ft are linked to the unconventional myosin Dachs (D), and the FERM domain proteins Expanded (Ex) and Merlin (Mer) are linked WW domain protein Kibra (Baumgartner et al, 2010;Genevet et al, 2010;Yu et al, 2010), regulate Hippo/Salvador/ Warts (Hpo/Sav/Wts) tumor suppressor signaling, which in turn stimulates cell proliferation through the Yorkie (Yki) and Scalloped (Sd) transcriptional complex in Drosophila and mammals (for a review, see Reddy and Irvine, 2008;Oh and Irvine, 2010;Pan, 2010;Zhao et al, 2010;Halder and Johnson, 2011). The Ds/Ft signaling pathway also regulates planar cell polarity (PCP) and proximodistal tissue patterning (Reddy and Irvine, 2008).…”

“…Lft was recently shown to modulate Ds/Ft signaling in wing and leg morphogenesis by its interactions with the intracellular domains of both Ds and Ft (Mao et al, 2009), for review, see (Oh and Irvine, 2010;Pan, 2010;Zhao et al, 2010). Vertebrate homologues of Lft are known as Lix1 in chicken and mouse (Swindell et al, 2001;Moeller et al, 2002).…”

“…63 Yki has been classified as an oncogene, as it promotes proliferation and growth under normal conditions, and thus inhibits apoptosis. 15 Consequently, inactivation of the core kinase cascade causes tissue overgrowth because of Yki derepression. If the caffeine-dependent ablation of eye tissue we observed in hpo-or wts-RNAi animals were indeed a consequence of Yki activity, then one would expect Yki overexpression (ey4yki-cDNA) to produce a comparable eye phenotype.…”

“…9 When Hippo signaling is inactive, Yki localizes to the nucleus where it forms complexes with transcription factors such as Scalloped, [10][11][12] Homothorax 13 or Mad 14 to activate the expression of target genes that promote cell growth, survival and proliferation. 15 Yki therefore acts as a growth promoter, whereas Hpo, Wts, Mats and Sav are considered tumor suppressors.…”
