Yeast‐mediated stereo‐selective reduction of estrone by continuous cell culture with dual stirred tanks for product yield improvement

Abstract: BACKGROUND: β-Estradiol is an important hormone for the treatment of breast cancer and osteoporosis. β-Estradiol can be produced via Saccharomyces cerevisiae mediated reduction of estrone. However, substrate inhibition and low production yield have been observed in the batch cell culture.

“…Therefore, in order to increase the productivity of cells, the authors proposed to perform the process continuously in a cascade of two stirred-tank reactors [54]. In the first reactor a continuous cell culture of the yeast was grown aerobically and continuously fed to the second reactor, in which reduction of 75 took place (Scheme 25).…”

“…Cheng and Tsai encountered the problem of substrate inhibition and low product yields during the reduction of estrone ( 75 ) to β-estradiol ( 76 ) in batch cell cultures of Saccharomyces cerevisiae expressing a native enzyme 17β-hydroxysteroid dehydrogenase. Therefore, in order to increase the productivity of cells, the authors proposed to perform the process continuously in a cascade of two stirred-tank reactors [ 54 ]. In the first reactor a continuous cell culture of the yeast was grown aerobically and continuously fed to the second reactor, in which reduction of 75 took place ( Scheme 25 ).…”

“…In a laboratory study, Cheng and Tsai (2010) observations of E1 to E2 transformation in dairy manure (Zheng et al (2012) led to the question of whether a similar transformation might occur in swine manure. In this Short Communication, we present limited but compelling evidence for a complete transformation cycle of E2 to E1 and then back to E2 in an aqueous suspension of swine manure colloids, with significant implications for the interpretation of E2's fate in the environment.…”

“…This paper focuses on the development of online SPME coupled liquid chromatography-electrospray ionization-ion trap mass spectrometric method for performing efficient chiral separation and analysis [7,[48][49][50][51][52] of trace secondary alcohol products R-and S-4-phenyl-2-butanol in a salt rich cell culture which was associated with the baker yeast S. cerevisiae catalyzed stereoselective biotransformation of the model compound 4-phenyl-2-butanone. In this method manual one-time fixed amount sample injection and online dual column switching technique were used to reach the goals of salt interference reduction, accurate, precise, and sensitive analysis.…”
