Xiaoaiping Induces Developmental Toxicity in Zebrafish Embryos Through Activation of ER Stress, Apoptosis and the Wnt Pathway

Abstract: The aim of the study was to determine the developmental toxicity of the traditional Chinese medicine Xiaoaiping (XAP) and to investigate its underlying mechanism of action. Zebrafish embryos were incubated with 0.4, 0.8, 1.2, and 1.6 mg/mL XAP. Endpoints such as mortality, hatching rate, malformation, body length, morphology score, swimming behavior, histological changes, reactive oxygen species (ROS) production, total superoxide dismutase (T-SOD) activity, and the mRNA expression of genes related to oxidative… Show more

“…The rate of hatching, which normally occurs with zebrafish embryo from 48 to 96 hpf, is a critical indicator of the developmental state of these embryos [64,65]. At a concentration of 25 μg/L, AEO-7 eliminated hatching (Fig.…”

“…However, they focused on two endpoints in their experiment: abnormal spastic behaviors (72 hpf) and alternations in the larval photomotor responses assay (120 hpf). Li et al [ 99 ] exposed zebrafish to Xiaoping at high doses, and many larvae opted to swim at a medium to a low rate, which led to decreased locomotion ability. Changes in larval behavior have been confirmed to be associated with damage to the central nervous system.…”

“…Hatching is a critical stage in the embryogenesis of Danio rerio for it aids in the evaluation of developmental delays and toxicity caused by different substances [93]. A normally developing Danio rerio typically hatches between 48 to 72 hpf [94].…”
