Objective: The objective of this review is to collect and document information on the Indian medicinal plants with anti-gout potential.Methods: Bibliographic investigation was carried out by consulting worldwide scientific databases, analyzing Ayurvedic text books and research journals. The search terms were "gout," "uric acid," "hyperuricemia," "xanthine oxidase (XO) inhibitor and uricosuric." Herbal keywords included "herbal medicine," "medicinal plant," "natural products," "phytomedicine" and "phytotherapy." Result and Conclusion:Medicinal plants have been used to treat various ailments since ancient times; hence, ethnobotanical investigations play an important role in pharmacological studies. In India, traditional medicines are being practiced for the treatment of gout and other rheumatic disorders from ancient time. This review provides a comprehensive summary of 130 Indian plants which have been mentioned in ancient literature or used traditionally for the treatment of gout. Out of these, 41 plants have been reported to possess XO inhibitory activity. Further, isolated phytoconstituents having promising XO inhibitor activity are also included in this review. Although a variety of medicinal plants with anti-gout potential have been found in the literature, there is limited information on evaluation of anti-gout activity of isolated phytoconstituents. The current review contains a detailed discussion of the potential of medicinal plants for treatment of gout.

Medicinal plants used in European folk medicine attached to Lamiales, Gentianales or Asterales orders are used to treat inflammatory disorders. Many targets have been identified but to date, implication of purinergic receptor P2X7 activation has not yet been investigated. We managed to evaluate the protective effect on P2X7 activation by plant extracts used as anti‐inflammatory in European folk medicine by the YO‐PRO‐1 uptake dye in vitro bioassay. Results revealed that among our selected plants, species from Scrophularia and Plantago genus were able to decrease significantly P2X7 activation (>50 % at 0.1 and 1 μg/mL). UPLC/MS, dereplication and metabolomic analysis of Scrophularia extracts, allowed us to identify the cinnamoyl‐iridoid harpagoside as putative inhibitor of P2X7 activation. These results open a new research field regarding the anti‐inflammatory mechanism of cinnamoyl‐iridoids bearing plants, which may involve the P2X7 receptor.

Inhibition of Xanthine Oxidase by Thiosemicarbazones, Hydrazones and Dithiocarbazates Derived from Hydroxy‐Substituted Benzaldehydes

Nonpurine xanthine oxidoreductase (XOR) inhibitors represent important alternatives to the purine analogue allopurinol, which is still the most widely used drug in the treatment of conditions associated with elevated uric acid levels in the blood. By condensing mono-, di- and trihydroxybenzaldehydes with aromatic thiosemicarbazides, aryl hydrazides and dithiocarbazates, three series of structurally related Schiff bases were synthesised, characterised and tested for XOR inhibitory activity. Hydroxy substitution in the para-position of the benzaldehyde component was found to confer high inhibitory activities. Acyl hydrazones were generally less potent than thiocarbonyl-containing Schiff bases. Within the thiosemicarbazone series, chloro and cyano substituents in the para-position of the thiosemicarbazide unit increased activities further, up to potencies approximately four-times higher than that of the benchmark allopurinol, as measured under the same assay conditions. In order to illustrate the potential of the Schiff bases to bind directly to the molybdenum centre in the active site of the enzyme, a representative example (H₂L) of each inhibitor series was co-ordinated to a cis-dioxomolybdenum(VI) unit, and the resulting complexes, [MoO₂(L)MeOH], were structurally characterised. Subsequent steady-state kinetic investigations, however, indicated mixed-type inhibition, similar to that observed for inhibitors known to bind within the substrate access channel of the enzyme, remote from the Mo centre. Enzyme co-crystallisation studies are thus required to determine the exact binding mode. Finally, the coordination of representative inhibitors to copper(II) gave rise to significantly decreased IC₅₀ values, revealing an additive effect that merits further investigation.
