Wogonin Induces Apoptosis by Activation of ERK and p38 MAPKs Signaling Pathways and Generation of Reactive Oxygen Species in Human Breast Cancer Cells

Abstract: Wogonin is a one of the bioactive compounds of Scutellaria baicalensi Georgi which has been shown to have antiinflammatory, anticancer, antiviral and neuroprotective effects. However, the underlying mechanisms by which wogonin induces apoptosis in cancer cells still remain speculative. Here we investigated the potential activation of MAPKs and generation of reactive oxygen species (ROS) by wogonin on MCF-7 human breast cancer cells. These results showed that wogonin induced mitochondria and death-receptor-medi… Show more

“…Estimation of Intracellular and Mitochondrial ROS LevelsTo determine intracellular and mitochondrial ROS levels, dichlorofluorescin diacetate (Molecular Probes, Eugene, OR) and mitochondrial specific MitoSOX (Invitrogen) fluorogenic probes were used, respectively (27). Briefly, cells were incubated in media containing dichlorofluorescin diacetate (20 Subcellular Fractionation-The nuclear and cytoplasmic fractions were obtained from 90% confluently grown cells on 100-mm dishes as described previously with slight modifications (28).…”

“…Anti-cancer drugs targeting cancer metabolism are associated with mitochondria dysfunction to induce apoptosis, reactive oxygen species (ROS) generation and so on (Pathania et al, 2009;Yu and Kim, 2011). Thus, cancer metabolism-related genes can be valuable targets to establish anti-cancer therapeutic strategies.…”

“…The molecular mechanisms by which flavonoids initiate apoptosis have not yet been fully understood. There can be several mechanisms factors that may be involved, which may include inhibiting the activity of DNA topoisomerase I/II activity, [23,[34][35][36] and decrease in the ROS [37].…”
