“…Srinivasan et al showed that WA, by inducing the proapoptotic protein in the prostate apoptosis response-4 (Par-4), caused the arrest of prostate cancer cell growth ( 13 ). Several studies have indicated that WA can stop the G2/M cell cycle and decrease the expression of NF-kβ and TNF-α, and thus it has antiproliferative activity and induces apoptosis in cancerous cells ( 34 , 35 ). It was found that WA also increased the activation of p21 in prostate cancer cells ( 36 ).…”

“…In this study, we demonstrated the in vitro antileukemic effect of the root extract of W. somnifera in a T-lymphoblastoid cell line. The high-performance liquid chromatography (HPLC) analysis performed on WE revealed the presence of WFA, WDA and to a lesser extent withanolide B. Withanolides, in particular WFA and its acetyl derivative, are highly bioactive and show anticancer activity [ 19 , 20 , 21 ]. Of note, its dihydroderivative is not active, thus suggesting that an unsaturated lactone moiety in ring A of WFA is important for its biological activity.…”

“…The study of natural products' biosynthetic pathways and patterns is important because it offers new approaches in total synthesis. Withanolides are a signature secondary metabolite (Sangwan et al 2004) and have significant and multiple bioactivities including immunomodulation, anti-cancer, neuroprotection, anti-hyperglycemic and the reversal of Alzheimer's disease (AD) (Choudhary et al 2013;Kushwaha et al 2012;Akizuki et al 2013;Sehgal et al 2012). Although structure and pharmacological activity have been studied and summarized during the last two decades, limited attempts have been made to illuminate the biosynthetic routes and identification of the key regulatory genes involved in withanolide biosynthesis.…”

“…The complex structures and diverse bioactivities of withanolides have attracted increasing attention from numerous researchers, and several reviews about their classification, complicated and changeable structures, and extensive biological activities have been published over the past thirty years (Kirson and Glotter 1981;Glotter 1991;Anjaneyyulu et al 1998;Chen et al 2011;Misico et al 2011;Choudhary et al 2013). The withanolides exhibit characteristic spectroscopic behaviors that contribute to their structural elucidation.…”
